Chinese Journal of Tissue Engineering Research 2005;9(27):208-209
BACKGROUND: The MRJ studies of acute myocardial infarction (AMI)were more frequently focused on the changes of myocardial perfusion. OBJECTIVE: To investigate the characteristics of infarction and myocardial reperfusion in MRI, and make comparison of them with the results of their staining pathologic sections. DESIGN: Completely randomized grouping design and randomized controlled study. SETTING: Magnetic Resonance Department of Air-force General Hospital of Chinese PLA and Radiological Department of General Hospital of Chinese PLA. MATERIALS: The experiment was completed in the Medical Animal Experimental Center, General Hospital of Chinese PLA in December 2003.Fourteen miniature pigs were divided into 2 groups: infarction group and reperfusion group with 7 pigs in each group. One-month before and after the operation, MRI was performed in order to strengthen the scanning examination. One-month after operation, axial plane pathologic slices, which were corresponding to the MRI, were given TTC and SE staining. According to the TTC staining results, samples of the infarcted myocardium (corresponding to the anterior wall of the left ventricular) and normal myocardium (corresponding to the posterior wall of the left ventricular) were taken and given HE staining to observe the extent of infracted myocardium. MAIN OUTCOME MEASURES: The changes in cardiac shape and function and the changes of myocardial T1 and T2 relaxation time. RESULTS: Fourteen Chinese miniature pigs were used with 7 in each group. ①Relaxation time of T1 and T2 of infarcted myocardium in infarction group was obviously longer than that of thenormal myocardium [1 159.54±78.67, (60.15±6.31) ms, 1 056.15±70.95, (47.46±7.94) ms, t=2.63,5.38, P< 0.05, 0.01] and that of the infarcted myocardium in reperfusion group was also obviously longer than that of normal myocardium [1 171.14±139.98,(56.64±6.16) ms, 1 074.64±97.61, (44.57±4.25) ms, t=2.64, 6.24, P < 0.05,0.01].②MRI in single infracted myocardium and reperfused infracted myocardium was both changed obviously, but without significant differences in relaxation time and stress.Dilatation of left ventricle in single infracted group was more obviously than that in the reperfusion infracted group.③Theresult of MRI was coincidence with that examinated with TTC staining CONCLUSION:①MRI is a useful imaging modality in the evaluation of AMI and the left ventricle remodeling after myocardial infarction.②It is significant for reperfusion to treat remodeling of left ventricle after acute myocardium infarction.③There is good correlation between MRI and pathological examination.

Objective To explore the characteristics and risk factors of type 2 diabetes mellitus (T2DM) onset in pedigrees. Methods A total of 865 subjects were screened and diagnosed by oral glucose tolerance test (OGTT) based on American Diabetes Association (ADA) criteria. Type 1 diabetes mellitus (T1DM) , maturity onset diabetes of the young (MODY) and chondriosome diabetes were excluded by clinical features and laboratory test of insulin and autoantibodies including glutamic acid decarboxylase antibody, insular cellular antibody and insulin autoantibody. A total of 182 pedigrees of T2DM were obtained. Results No gender difference was found in the prevalence of T2DM (42. 59% in male and 48. 18% in female respectively, P >0. 05) , nor was the newly diagnosed rate(9. 89% in male and 11. 82%in female, P > 0. 05). The onset age was (63. 3 ± 12. 4) years old in the first generation [(64. 4 ± 12. 5)years in male and (62. 3 ± 10. 3) years in female] , (47. 1 ± 8. 7) years old in the second generation [(48. 2 ±9. 3)years in male and (46. 1 ± 8. 1) years in female] , (29. 6 ± 10. 2) years old in the third generation [(28. 9 ±9. 5)years in male and (30. 0 ± 10. 4)years in female]. Compared with normal glucose tolerance (NGT) subjects , newly diagnosed T2DM and impaired glucose regulation (IGR) subjects had higher prevalence of hypertension, hyperlipidemia and smoking but less physical activities. Statistical differences were shown in body weight five years before diagnosis, one years before diagnosis and at diagnosis in newly diagnosed T2DM[(68. 4 ±12. 4)kg, (69. 5 ± 11. 0)kg and (69. 1 ±9. 6)kg] and IGR[(66. 1 ±10.7)kg, (65.9 ± 10.7) kg and(65.7 ± 10.4) kg] , when compared with NGT [(61.0 ± 10.2) kg,(59. 5 ±11.0) kg and (60. 1 ± 10. 4) kg, all P < 0. 05] . The same results were obtained with waist circumference and waist-hip ratio [(4. 1 ± 12. 5) cm and 0. 92 ± 0. 36 in newly diagnosed T2DM while (89. 1 ± 10. 7) cm and 0. 90 ± 0. 64 in IGR] , when compared with NGT[(82. 5 ± 10. 1) cm and 0. 82 ±0. 25] , all P <0. 05. Conclusions No gender difference was found in the onset characteristics of T2DM.High prevalence of obesity, hypertension, hyperlipidemia and smoking with less physical activities were associated with T2DM.

This study explored a novel systemic community-based model for detecting and managing people living with HIV/AIDS (PLWHA). Both quantitative and qualitative research methods were used in this study. A quantitative questionnaire investigation was conducted in a sample of 1192 subjects which were randomly selected from two areas with high HIV prevalence, Xiangfan City and Shiyan City of Hubei Province, China. Twenty-two medical and health service staffs were interviewed by semi-structured questionnaire focusing on awareness, status, problems, and suggestions about community-based Voluntary Counseling and Testing and Provider Initiated Testing and Counseling (VCT/PITC). And they were organized to discuss about the aforementioned issues in Xiangfan City and Shiyan City, respectively. Our results showed that the accessibility and availability of the general VCT/PITC were bad. About 28.3% had known and only 4.9% had made use of VCT/PITC. Developing community-based VCT/PITC had some special advantages that can overcome some existing problems to remedy the aforementioned defects. We are led to conclude that, to maximize the availability and uptake rate of the VCT/PITC, we plan to detect PLWHA by developing the community-based VCT/PITC through 4 paths. Then we establish the community HIV health care center constituted of 8 sectors to provide an overall management. Thus, we can effectively detect and manage the PLWHA with a new systemic community-based model.

Clinical Medicine of China 2009;25(7):718-720
Objective To examine the serum homocysteine, folic acid and Vitamin B12 levels in macrocytic anemia patients and observe their dynamic changes following therapy. Methods Homocysteine, folic acid and Vita-min B12 were analyzed by electro-chemiluminescence immunoassay. Complete blood cell count was analyzed by opti-cal method and resistance method. Results The homocysteine is significantly higher in nutritional megaloblastic a-nemia[ (71.26±27.84)μmoL/L ] than in drug-induced megaloblastic anemia[(11.44±5.06)μmol/L],in myel-odysplastic syndrome[ (9.51±4.13)μmol/L] and in the normal control group[(8.74±5.42)μmoL/L] (P<0.01). After treated with low-dosage folic acid and Vitamin B12,the patients with megaloblastic anemia presented slow declining but eventually normal homocysteine levels, compared with those received high-dosages. Conclusion Homocysteine can be used for differential diagnosis of macrocytic anemia. The duration of remaining of abnormal ho-mecysteine levels is related to the dosage of folic acid and Vitamin B12.

Objective:To determine the effect of IL-12 on the cellular immune response of PBMC from patients with chronic hepatitis B virus infection, and provide basic scientific information for clinic therapy of this disease. Methods:PBMCS were prepared from peripheral blood of individuals with chronic HBV infection and cultured in the presence or absence of HBsAg and HBcAg with or without IL-12.The level of IFN-?in culture supernatants, the frequency of IFN-?-producing cells, and the subpopulation of IFN-?-producing cells were detected by either ELISA,ELISPOT or FACS.Results:Less than 30% patients and very low level of IFN-? were observed when PBMCs were stimulated with HBsAg or HBcAg alone. Addition of IL-12 to the cultures resulted in significant increase in IFN-?production and IFN-?-producing cells. In addition, IL-12 induced expression of IFN-? not only by CD8~+T cells, but also by non-T cell populations.Conclusion:IL-12 can promote the cellular immune response to the chronic hepatitis B virus by the enhancement of IFN-?production.
