Apolipoprotein E (ApoE) was purified from human serum and used as the antigen to immunize BALB/C mice. The splenic cells of the mice fused with NS-1 mouse myeloma cells. Three hybridoma cell lines(E3C3, E6C3 and E4C2)secreting anti-human ApoE McAb were established. The immunological property of the McAb was studied. The ascites fluids titer were 8?10~(-5)2?10~(-6). The McAbs did not cross react with the other apolipoprotins. ApoE was purified by immunoaffinity chromatography prepared by using the ApoE McAb. McAbs recognized two distinct respective epitopcs on ApoE. The subclass of immunoglabulin of three McAbs were IgG_1. A double McAb sandwich EL1SA was developed to evaluate ApoE levels in normal and hyperlipidemia human serum.

Objective To observe the effect of injection needle scalpel combined with Tongbi tincture heat deposited on the erythrocyte sedimentation rate, CRP and IL-1 in knee osteoarthritis.Methods 96 cases of knee osteoarthrosis according to the treatment divided into the control group(n=48) and experimental group(n=48), the control group was treated with injection needle scalpel, the experimental group on the basis of injection needle scalpel combination of tongbi tincture heat deposited, all lasted for 14 days.The clinical efficacy, CRP, IL-1, blood sink, knee joint functional impairment score, and safety was observed in the two groups.Results The total effective rate of the experimental group was 93.75%,higher than the control group 79.17%(P<0.05).After treatment, the CRP, IL-1, blood sedimentation of experimental group was lower than the control group[(7.09±0.97)mg/L,(6.54±0.82) ng/L,(16.47±2.03) mm/h vs.(10.35±1.28) mg/L,(7.21±0.93) ng/L,(18.25±2.21) mm/h](P<0.05).The scores of knee joint function impairment in the experimental group was higher than that in the control group(P<0.05).There was no significant difference in safety between the two groups.Conclusion The therapeutic effect of injection needle scalpel combined with Tongbi tincture heat deposited in treatment knee osteoarthrosis is better than injection needle scalpel.It can promote the absorption of inflammation, relieve knee function.

Objective To investigate and analyze the data of continuing medical education in order to understand the real need of the learners.Methods Reviewing and making statistic analysis of the data of Renji hospital's continuing medical education courses from 2007 to 2012,including the amount,title,discipline,geographical distribution,training content and the questionnaire survey.Results It shows that the learners from coastal and developed areas have a higher demand for cuttingedge technology and the new progress in medicine(49.1％),while the learners from inland and western regions prefer to learn the basic medical theory and technology (41.9％).Senior doctors tend to study cutting-edge technology and new progress medical courses (51.7％) while the junior persons prefer the basic medical knowledge (46.6％).These mean that the needs of content of continuing medical education is associated with the regions and the level of the doctors (P＜0.05).Conclusion The conclusions prompt that medical development between different regions of China may be unbalanced.Chinese government should pay more attention in terms of allocation of health resources and personnel training.It also provides advice for future courses so that it may improve the efficiency of the medical education resources.

Objactive:To study the color changes of SY-1 silicone elastomer before and after polymerization.Methods:L*a*b*color parameters of 10 samples of SY-1 silicone elastomer were measured with Minolta chroma-ticity instrument(CS-321) before and after polymerization.The color difference(△E) between precure and post-cure was calculated as△E =[(△L)2+(△a)2+(△b)2]1/2.Results:After polymerization L*and a*valueswere increased(P

0.05).Conclusions:Laser welding joint gap at 0.00~1.00 mm does not affect the mechanical property of laser-welded titanium.

AIM: To evaluate the effects of all-trans retinoic acid (atRA) on the proliferation in cultured mouse cerebral microvascular endothelial cells (bEnd.3) . METHODS: Cultured cells were divided into five groups randomly, one as control group, the other four groups were 10-9, 10-8, 10-7 and 10-6 mol/L group. Effects of atRA on proliferation in bEnd.3 cells were detected by flow cytometry and immunocytochemitry of PCNA and MTT at 24 h, 48 h and 72 h. The effects of atRA (10-6 mol/L group) on the expressions of angiogenic genes in bEnd.3 cells were studied using microarray. RESULTS: The results of MTT and flow cytometry showed that all- trans retinoic acid at concentration of 10-6 mol/L significantly inhibited the proliferation of bEnd.3 cells. Immunocytochemical staining showed the expression of PCNA was markedly decreased in bEnd.3 cells at 24 h after treatment with atRA. Microarray results demonstrated that there were 11 down - regulated angiogenic genes and 2 up - regulated angiogenic genes in 10-6mol/L atRA group. CONCLUSION: All - trans retinoic acid at concentration of 10-6mol/L may significantly inhibit the proliferation of bEnd.3 cells treated for 24 h ire vitro via down-regulation of angiogenic genes and PCNA expression.

Objective To investigate the intraoperative nursing cares in 25-gauge(25G) trans-conjunctiva sutureless vitrectomy(TSV25G). Methods The nursing cares,such as preparation,closely intraoperative cooperation,postoperative measurements and health education,were put into effect in 45 patients undergone TSV25G.Results 45 patients had undergone TSV25G successfully with the proper intraoperative cares.No severe complications was found.Conclusion Preoperative preparation,closely intraoperative cooperation,postoperative measurements and health instructions are important factors to make sure the TSV25G successful.

Objective To observe the effect of unilateral ureter obstruction(UUO) on the expression of complement 5a receptor(C5aR) in rat renal tissue,and to investigate the roles played by C5aR in the development of renal intersititial fibrosis.Methods Thirty six male Sprague Dawley rats were randomly assigned into two groups(18 each): sham-operated group and UUO group.The left ureter of the rats in UUO group was obstructed,while in sham-operated group the left ureter was not.Six rats in each group were executed respectively on the 5th,10th and 15th day after surgery.Histological changes in renal tubular interstitium were observed with H.E.and Masson staining,and the expression of C5aR in renal tissues was detected by iminunohistochemical ELiVisionTM plus at each time point.Results Renal tubular dilatation and partial interstitial cells degeneration were found in UUO group on the 5th day after surgery.Necrosis of the interstitial cells was found in renal tubule,and the interstitial fibrosis was found on the 10th day.On the 15th day after surgery diffuse thickening and shrinkage of renal tubular basement membrane and obvious fibrosis were found(P0.05).Conclusion The C5aR expression in renal tissue increases significantly at the early stage of UUO,while the process is slowed down in the later stage.C5aR may take part in the process of renal intersititial fibrosis at the early stage.

Objective To investigate the diagnostic value of laparoendoscopic exploration and peri-toneum biopsy in patients with ascites of unknown origin and to study the feasibility of replacing hard laparo-endoscope with ultrathin gastroendoscope. Methods A total of 35 patients with ascites of unknown origin underwent laparoendoscopic exploration and biopsies were taken at suspected sites on peritoneum. Results Definite diagnosis was made in 33 patients after laparoendoscopy (94. 3% ), among which there were 10 tuberculosis peritonitis (28. 6% ), 7 metastatic peritoneal carcinoma (20%), 5 liver cancer, 3 liv-er cirrhosis, 3 primary peritoneal carcinoma, 2 ovary cancer, 1 hepatovascular sarcoma, 1 spleen lymphoma and 1 acidophilic enteritis. No procedure related complications were observed. Conclusion Laparoendosco-py is safe and effective in target peritoneal biopsy, and is of great importance in diagnosis of ascites of un-known origin.
