1.Study on the floating characteristics of the calcium alginate-chi-tosan intragastric floating beads

Objective To prepare calcium alginate-chitosan intragastric floating beads and investigate the influences of various factors on the floating characteristics of beads.Methods Sodium alginate solution added with frother was slowly dripped into chitosan-calcium chloride solution added with acetic acid,calcium alginate-ehitosan intragastric floating beads were obtained after drying.The effects of frother,addition amount of acetic acid,the concentration of alginate solution,chitosan and calcium chloride solution on the floating characteristics of beads were investigated.Results Frother and addition amount of acetic acid were the main influence factors on the floating characteristics,and the concentrations of alginate solution, chitosan and calcium chloride solution also had some influences.Conclusion The calcium alginate-chitosan intragastrie floating beads with favorable floatability could be achieved through adjusting the formula,it laid a foundation for the furether development of the preparations in intragastric floating preparations.
2.Efficacy and Safety of Oxcarbazepine combined with Levetiracetam on Children with epilepsy

Objective To investigate the efficacy and safety of oxcarbazepine combined with levetirac-etam on children with epilepsy. Methods According to different treatments, 62 cases of children with epilepsy were divided into the OXC group (treated with oxcarbazepine, 40 cases) and the OXC combined with LEV group (treated with oxcarbazepine and levetiracetam, 20 cases). Then the therapeutic efficacy, EEG changes and ad-verse reactions of patients in the two groups were studied. Results The total efficacy was 87.5% in the OXC group, and was 95.5% of the OXC combined with LEV group, with no significant difference. EEG improvement rate was 52.5% in the OXC group and 81.8% in the OXC combined with LEV group , with significant difference (P < 0.05). The rate of adverse reactions in the OXC combined with LEV group was lower than that in the OXC group (P < 0.05). Conclusion Both levetiracetam and oxcarbazepine has good curative effect on children with epilepsy , but combination with levetiracetam can significantly decrease the epileptic discharges on EEG , with higher security.

4.Alcohol exposure during pregnancy causes non-compaction cardiomyopathy in offspring mice

Objective To investigate the relationship of alcohol exposure during pregnancy and non-compaction cardiomyopathy (NCC) in offspring mice.Methods Pregnant mice of ED3.5-ED18.5 were given 56% alcohol by gavage at a dose of 5 mL/kg.The ED19.5 mice were sacrificed,and the heart of the fetal mice was harvested.Transmission electron microscopy (filaments,mitochondria and sarcoplasmic reticulum) and HE staining were used to verify the changes of structure and ultrastructure of the obtained myocardial tissues.Echocardiography was used to evaluate the cardiac function and ventricular myometrium of the offspring mice after growing up.Results Alcohol exposure during pregnancy caused the disorganized and dissolved myofilaments in the fetal mice.Some offspring mice (31.25%,5/16) had NCC.The ratio of non-compacted myocardium to compact myocardium at the end of systole (N/C) was 2.49 ± 0.6 in the offspring mice of the alcohol exposure group,significantly higher than that in the control offspring mice (0.62 ± 0.23,t =10.397,P =0.000).The volume of heart was decreased in the offspring mice of the alcohol exposure group while the left ventricule was enlarged.Echocardiography showed cardiac dysfunction and thickened ventricular septal/left ventricular posterior walls in the grown-up mice of the exposure group.Conclusion Large dose of alcohol exposure during pregnancy cause trabeculations and non-compaction in ventricular myocardium,and it might be one of causers for NCC in the offspring.
5.Functional MRI study of insula in the task-state and resting-state

Objective To study the activated status of insula during the performance of word generation task, and to explore the function of different part of insula with functional MRI (fMRI). Methods Twenty-eight subjects were recruited in this study (male 15, female 13), all of them underwent block-designed fMRI with word generation tasks and resting-state scan. SPM 5 and REST 1.3 were used to process the data. Bilateral anterior insula and posterior insula were selected as seeds to calculate the connectivity coefficiency with other voxels, and differences between the anterior and the posterior insula were compared. Results Bilateral anterior insula was significantly activated, while bilateral posterior insula was significantly deactivated. The functional connectivity areas with left anterior insula included right anterior insula, right posterior insula, supplementary motor area (SMA), left superior temporal gyrus, left middle frontal gyrus, left superior frontal gyrus, left inferior parietal lobe, middle cingulate gyrus, right striatum and right inferior frontal gyrus. The functional connectivity areas with left posterior insula included right posterior insula, left anterior insula, right superior temporal gyrus, middle cingulate gyrus, right precentral gyrus and right striatum. The functional connectivity areas with right anterior insula included SMA, left inferior frontal gyrus, right inferior parietal lobe, left inferior parietal lobe, left superior temporal gyrus, right precentral gyrus, right striatum, middle cingulate gyrus, left middle frontal gyrus, left striatum, right middle frontal gyrus, right inferior frontal gyrus and left transverse temporal gyrus. The functional connectivity areas with right posterior insula included right precentral gyrus, left superior temporal gyrus, left anterior insula, left posterior insula, right supramarginal gyrus and middle cingulate gyrus. Conclusion Anterior insula and posterior insula are functionally connected with different areas, and concerned with the language function in different manners. Left lateral anterior insula may play an important role in the language function.

Targeting such setbacks as difficulty to registerand difficulty to see doctors at outpatient clinic services,the hospital adopted the lean management since August 2010.For example,diversified reengineering of the outpatient service process in place,which has improved outpatient traffic at the registration section by means of innovated appointment mode and outpatients total reservation;workload balance by means of through-counters for registration and fee payment as well as the timeinterval outpatient visits to doctors; reduced human costs and outpatient back-forth movement by means of integrated service and resources integration.Lean management plays a positive role in hospital management,as proven by much shortened outpatient waiting time,much higher outpatient satisfaction,for a win-win of both patients and the hospital.
7.Nucleotide sequencing of E/NS1 gene segment of dengue type 1 viruses isolated in Guangdong Province.

OBJECTIVETo analyse the genetic relationship of local strains of dengue type 1 viruses isolated in different years and regions from Guangdong Province, and to explore the genetic links with strains of adjacent countries by comparing with the sequences of relevant strains in Genbank.
METHODSThe viral RNAs were extracted and used for one-step reverse transcriptase polymerase chain reaction (RT-PCR) to amplify the partial nucleotide fragments in E/NS1 gene junction which were then cloned into the plasmid pBluescript II SK for sequencing, the results were analysed by DNASTAR software.
RESULTSThe phylogenetic tree of the sequenced 14 strains of dengue type 1 viruses branches into two genotypic groups. The nucleotide sequences showed a maximal homologies of 99.2% with Indonesia strains, 100% with Philippines strains and 98.8% with Thailand strains.
CONCLUSIONSThe dengue type 1 viruses of Guangdong Province are closely related to the Philippines, Indonesia and Thailand strains, which may indicate the possibility of importation from those countries.

8.Expression of Myocardial Specificity Markers MEF-2C and Cx43 in Rat Bone Marrow-derived Mesenchymal Stem Cells Induced by Electrical Stimulation In Vitro.

Bone marrow-derived mesenchymal stem cells (BMSCs) for repairing damaged heart tissue are a new kind of important treatment options because of their potential to differentiate into cardiomyocytes. We in this experiment investigated the effect of different electrical stimulation time on the expression of myocardial specificity gene and protein in rat bone marrow mesenchymal stem cells (rBMSCs) in vitro. The rBMSCs of second or third generation were randomly divided into three groups, i.e, electrical stimulation (ES) group, 5-Azacytidine (5-Aza) group and the control group. The rBMSCs in the ES groups with complete medium were exposed to 2 V, 2 Hz, 5 ms electrical stimulation for 0. 5 h, 2 h, 4 h, and 6 h respectively every day for 10 days. Those in the 5-Aza group were induced by 5-Aza (10 μmol/L) for 24 h, and then cultured with complete medium for 10 days. Those in the control group were only cultured with complete medium, without any treatment, for 10 days. The rBMSCs' morphological feature in each group was observed with inverted phase microscope. The mRNA expression of myocyte-specific enhancer factor 2C (MEF-2C) and connexin 43 (Cx43) were examined with Real-Time quantitative PCR and the protein expression of MEF-2C, Cx43 were detected with Western Blot method. The results showed that the mRNA expression level of the MEF-2C, Cx43 and the protein expression level of MEF-2C, Cx43 were significantly higher in the ES group and 5-Aza group than those in the relative control group (P < 0.05). It suggests that electrical stimulation could play a part of role in the induction of the rBMSCs to differentiate into the cariomyocyte-like cells in vitro and the effectiveness of the electrical stimulation with 2 h/d had the best in our experiment. But the mechanism how electrical stimulation promotes the differentiation of rBMSC into cardiomyocyte is still unclear.

9.Solanine induces the apoptosis of human prostate cancer cells via ROS/p38 signaling pathway

Objective To investigate the molecular mechanism of solanine-induced apoptosis of prostate cancer cells Du145 and LNCaP.Methods The effects of solanine on the viability of Du145 and LNCaP cells were evaluated by MTT assay.The generation of intracellular reactive oxygen species (ROS) and solanine-induced apoptosis were measured by flow cytometry.The protein levels of p38 and p-p38 expressions were examined by Western blot.Results Solanine significantly inhibited the viability of Du145 and LNCaP cells in a dose-dependent manner (P<0.01).The inhibition of solanine on cell viability was suppressed by the ROS scavenger NAC.ROS generation,apoptosis and phosphorylation of p38 were induced by treatment with solanine at 40 μmol/L for 24 h.The expression of p38 and solanine-induced apoptosis were suppressed by NAC and SB203580.Conclusion Solanine induces the apoptosis of human prostate cancer cell via the RO.S-p38 signaling pathway.
10.Prognostic value by combination of angiographic and clinical characteristics in stenting patients with unprotected left main coronary artery lesion

Objective To compare the predicting values for Prognosis among Global Risk Classification (GRS),Synergy Between Percutaneous Coronary Intervention With TAXUS and Cardiac Surgery (SYNTAX) score,the European System for Cardiac Operative Risk Evaluation (EuroSCORE) in patients who received stenting because of unprotected left main coronary artery (ULMCA) lesion.Methods Totally 105 successive elderly patients with ULMCA lesion who received stenting were divided into 2 groups:with and without main adverse cardiac events (MACE).The clinical and angiographic characteristics were analyzed and then compared among GRC,SYNTAX score and EuroSCORE.Results As compared with none MACE group,MACE group had higher EuroSCORE score (2.0±2.3 vs.6.5±2.9,t=8.18,P=0.002),and more trivessel disease and left main bifurcation lesion (x2 =8.96,6.96,P =0.011,P =0.008).High risk GRC showed more MACE than medium or low risk GRC [55.9% (19/34) vs.20.5%(9/44),7.4% (2/27),x2 =19.77,P=0.001].AUC(95%CI )of GRC,SYNTAX score and EuroSCORE were [0.821 (0.730-0.912),0.586(0.462-0.709) and 0.631 (0506-0.757)],respectively.Compared with SYNTAX score and EuroSCORE,GRC was superior in the MACE predicting value (Z=3.29,2.63,P<0.01 or P<0.05).
