The activity of the negative regulator of cell cycle, P27, affects the normal process of cell cycle. As a type of cell cycle diseases, tumor is closely correlated with the activation of P27. It is shown that the level of P27 decreases in most kinds of tumors.By its influence on cell proliferation and apoptosis, P27 plays an important role in the occurrence and development of tumor. The activity of P27 is regulated by several pathways either at or after transcription, so various ideals which directly change the protein level of P27 or indirectly interfere the activities of elements in regulative pathways of P27 will provide many novel and efficient ways for therapies in tumors.

Objective To establish a HPLC determination method for the effective constituents extracted from Fructus Schisandrae Chinensis by low-temperature water extraction.Methods The assay was conducted on Kromasil C18 column(250 mm? 4.6 mm,5 ? m)using a gradient elution with acetonitrile-water as mobile phase.The flow rate was set at 1.0 mL/min,column temperature was kept at 30 ℃,and the detection wavelength was at 240 nm.Results The linear range for schizandrol A was 5.0～ 200.0 ? g/mL(r=0.999 9),and the average recovery was 101.27 % with RSD being 1.97 %(n=6).Conclusion The method is simple,reliable and reproducible for the determination of schizandrol A.It can be used for quality control of preparations including Fructus Schisandrae Chinensis.

Objective To investigate the characteristics of Wallerian degeneration (WD) of secondary pyramidal tract after supratentorial cerebrovascular diseases, and to study the relationship between secondary WD and clinical performance. Methods We retrospectively reviewed and investigated the results of brain CT films of 272 cases suffered from supratentorial cerebral vascular diseases during 2001 till 2004. The number of supratentorial disorders was counted respectively in WD group and non-WD group, the distribution feature of these disorders was analyzed, and the changes in the cerebral peduncles of the midbrain and Activities of Daily Living Scale (ADLs) were determined. The information obtained was analyzed statistically respectively. Results Seventy-four cases of secondary WD were found, the rate of WD outbreak was 27.2%. Objective:To explore the effects of CICARE communication model in emergency nursing .Meth-ods:The CICARE communication model was applied from the beginning of administration to rescue and transfer processes .The nurses were required to apply the CICARE communication skill to the whole nursing process through training , scenario simulation , role playing , practice and assessment .Results:The communication ability of nurses and satisfaction of patients improved significantly .Doctor -patient disputes decreased .Conclusions: CICARE communication model , as an effective way of communication plays a important role in promoting the comprehensive quality of nurses and the nursing quality .

BACKGROUND: Our previous studies have found that the Kangguzengsheng Capsules can promote fracture healing.OBJECTIVE: To explore the influence of Kangguzengsheng Capsules-containing serum on the proliferation andosteogenic differentiation of bone marrow mesenchymal stem cells.METHODS: Forty Sprague-Dawley rats were randomly divided into four groups. The capsule powder was resolved into1 mL natural saline and intragastrically administered into rats according to 1.16 g/100 g, 3.48 g/100 g, 10.44 g/100 g in low,medium and high dose groups. Rats in control group were given equal volume of natural saline. After consecutiveadministration for 12 days, blood samples from the abdominal aorta were collected and serum samples were isolated andpreserved until use. Bone marrow mesenchymal stem cells were isolated and purified by the whole marrow adhesion method,and identified by flow cytometry. Harvested cells were divided into four groups and cultured in the osteogenic culture mediumcontaining different kinds of serum samples as described above. MTT method was adopted to test the cell proliferation at 24,48 and 72 hours of culture. Alkaline phosphatase activity in cells was detected at 7 and 14 days of culture. Alkalinephosphatase staining was performed at 7 days of culture, and alizarin red staining performed at 14 days of culture.RESULTS AND CONCLUSION: Compared with the blank control group, bone marrow mesenchymal stem cells showedno significant changes in the proliferative ability and alkaline phosphatase activity in the low dose group, but these twoindices were significantly increased in the high and medium dose groups at 48 and 72 hours of culture or at 7 and 14days of culture, respectively (P < 0.05). Cells positive for alkaline phosphatase and alizarin red staining were observed inthe low, medium and high dose groups, and the cell staining was most remarkable in the high dose group. To conclude,Kangguzengsheng Capsules-containing serum can promote the proliferation and osteogenic differentiation of bonemarrow mesenchymal stem cells.

Objective To analyze the correlation of nonalcoholic fatty liver disease (NAFLD) with waist,hip circumference and body mass index in order to explore the prevention countermeasures.Methods The datum of routine physical examination and questionnaire survey among 2 503 employees of 12 enterprises in November 2013 were collected.The indexes of height,weight,waist circumference,hip circumference,blood pressure,blood lipid and blood glucose of the subjects were measured.The grouping was according to whether the subjects suffering from NAFLID.The correlation and epidemiological characteristics between each group and the risk factors of body weight,waist,hip circumference and body mass index were analyzed.Results 2 503 subjects were collected including 490 NAFLID patients (19.57％).The body weight and body mass index of NAFLID patients were significantly lower than those of the control group.The result of BMI classification showed that the subjects of the control group were overweight while the subjects with NAFLID were obesity.The waist circumference and hip circumference of NAFLID patients were significantly larger than that of the control group.Conclusions Larger waist and hip circumference and overweight are risk factors of NAFLID.Effective intervention measures,scientific control of body weight,rational diet,the strengthening of physical exercises should be taken in order to prevent and control the development of fatty liver.

Objective To investigate the CT and MRI features of ovarian fibroma.Methods CT and MRI findings of 42 patients with ovarian fibroma confirmed by operation and pathology were analyzed retrospectively.Twenty-one patients were examined by CT,26 cases was examined by MRI,and 5 cases were examined by CT and MRI.Results In 42 cases of ovarian fibroma,there were simple type in 29 cases,degenerative type in 11 cases,and special type in 2 cases.All the cases had single lesion,morphology were round or elliptic or lobulated,clear boundary in 40 cases,partial fuzzy boundary in 2 cases,3 cases with calcification,hemorrhage in 1 case.The maximum tumor diameter were 1.4-26.7 cm,median value was 5.5 cm.Simple type of ovarian fibroma were equal density on CT,low signal on T1WI and T2WI.Degeneration type was patchy,fissure,low density areas on CT or high signal on T2WI,and the tumor parenchyma was almost no enhancement or only slight enhancement.A large number of hemorrhages had been found in 1 special type patient,and significantly enhanced in the other special type patient.Special type were misdiagnosed as malignant tumor.Conclusion CT and MRI performance of ovarian fibroma has some characteristics,but diagnosis still need to rely on pathology.

Objective To explore the ultrasonographic manifestations of the fetal with asplenia syndrome. Methods At 21751 cases of fetal systemic ultrasound examination during 20-40 weeks, 4 cases with asplenia syndrome were diagnosed. and the ultrasonographic manifestations of the 4 cases were analyzed retrospectively. Results The ultrasonographic features of fetal asplenia syndrome;①with complex and serious cardiovascular malformation; ②visceral transposition; ③ absence of spleen. Conclusions When the complex cardiovascular malformations were found by prenatal ultrasound,the position of fetal spleen and internal organs in order to early diagnosis of fetal asplenia syndrome.

Objective To study the characteristics of patients with acute arsine poisoning and its possible treatments. Methods The only use of drugs,or drugs with plasma exchange(PE)were used to treat 36 patients with acute arsine poisoning.The blood haemolysis,enzymes of creatinc kinase(CK),lactate dehydrogenase(LDH),alkaline phosphatase (ALP),alanine aminotransferase(ALT),aspartate aminotransferase(AST),?-hydroxybutyric dehydrogenase(HHBD), total bihrubin(TBIL),indirect bilirubin(IBIL),direct bilirubin(DBIL),blood urea nitrogen(BUN),serum creatinine (Cr)were observed.Results There was an exposure time-effect relation in clinical characteristics,and a linear correlation between the concentrations of arsenic in blood and urine(r=0.718,P=0.019),but no significant correlations were found between the concentrations of arsenic in blood or urine with CK,LDH,ALP,ALT,AST,HHBD,TBIL,IBIL, DBIL,BUN,Cr(P＞0.05).In patients with severe acute arsine poisoning,PE quickly controlled hemolysis within 24 hours,and prevented secondary damage in kidney and other organs,oliguria stage got much shorter,and CK,LDH,ALP, AST,HHBD,TBIL,IBIL,BUN significantly reduced at 24 to 72 hours after PE treatment(P＜0.05).Conclusions The only use of drug was enough for the treatment of mild acute arsine poisoning.To the patients with severe acute arsine poisoning,PE may be an effective method to control its blood hemolysis and prevent complications,which should be taken as early as possible.

Objective:To construct a short hairpin RNA(shRNA) eukaryotic expression vector of the survivin gene,and investigate its inhibitory effect on the survivin expression in human cervical cancer cells.Methods:We designed and synthesized 2 pairs of survivin-specific small interfering RNA primers(s1 and s2),cloned them into the eukaryotic expression vector pSilencer 2.1-U6 neo by DNA recombined techniques after annealing connection reaction,and transfected them respectively into human cervical cancer cell line HeLa using LipofectAMINE2000 after identificationby restrict endonuclease digestion and DNA sequencing.Then we selected the positive clones by G418 and detected the expression of survivin mRNA by semi-quantitative RT-PCR and that of the survivin protein by Western blot.Results:The survivin shRNA eukaryotic expression vectors pSilencer2.1-s1 and pSilencer2.1-s2 were successfully constructed.Positive clones were obtained by screening with G418 for 24 days,the survivin expression in HeLa cells decreased in different degrees after transfected with pSilencer2.1-s1 and pSilencer2.1-s2,and the latter showed a better interference efficiency.Conclusion:The shRNA eukaryotic expression vector of the survivin gene we constructed,with its improved interfering efficiency,has paved the way for further research on its role in regulating the biological behavior of cervical cancer cells.
