1.Vascular endothelial growth factor promotes the proliferation and osteogenic differentiation of mouse bone marrow mesenchymal stem cells

BACKGROUND:Vascular endothelial growth factor (VEGF) is a crucial factor for regulating bone marrow mesenchymal stem cells in microenvironment, which can promote endothelial differentiation of bone marrow mesenchymal stem cells. But there is no report about VEGF effect on regulating the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells. OBJECTIVE:To investigate the regulatory role of VEGF in the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells. METHODS:The bone marrow mesenchymal stem cells were isolated and cultured from mouse long bones. cellCounting Kit-8 was used to test the proliferation of bone marrow mesenchymal stem cells cultured with different concentrations of recombinant VEGF proteins. The appropriate concentration of VEGF recombinant protein was selected to test their effects on the osteogenic differentiation of bone marrow mesenchymal stem cells. The mRNA and protein levels of Osterix, Runx2, alkaline phosphatase, osteocalcin and heme oxygenase-1 in bone marrow mesenchymal stem cells under induction of VEGF were detected by molecular biology methods. RESULTS AND CONCLUSION:(1) VEGF promoted the proliferation of bone marrow mesenchymal stem cells dose-dependently, and 100μg/L was the optimum concentration. (2) VEGF promoted the expression of Osterix, Runx2, alkaline phosphatase and osteocalcin in bone marrow mesenchymal stem cells under osteogenic induction. Similar results were obtained by alizarin red staining and quantification of numbers of calcium nodules. (3) VEGF induced the expression of heme oxygenase-1 in bone marrow mesenchymal stem cells at mRNA and protein levels. These findings indicate that VEGF can induce the expression of heme oxygenase-1 i in bone marrow mesenchymal stem cells, and promote the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.

3.Risk factors of incision infection and spinal canal hematoma after lumbar posterior surgery

Objective To analyze the risk factors of incision infection and spinal canal hematoma after lumbar spinal posterior surgery.Methods Data of 33 patients with incision infection and 25 patients with spinal canal hematoma after posterior surgery for lumbar spinal disease from January 2010 to December 2014 were retrospectively analyzed.For the patients with incision infection,20 of them were males and 13 were females,with an average age of 58.85±8.76 years (range 38-79 years).Of these patients,5 were diagnosed with lumbar disc herniation,9 with lumbar spondylolisthesis,15 with lumbar spinal stenosis and 4 with lumbar vertebral fracture.For patients with spinal canal hematoma,17 of them were males and 8 were females,with a mean age of 60.96±11.37 (range,38-77).The diagnoses of them were lumbar disc herniation in 18 patients,spondylolisthesis in 3 patients and spinal stenosis in 4 patients.From the same period database,patients who underwent lumbar posterior lumbar surgery with no postoperative complications were selected by 3:1 ratio as the control group according to age,gender and diagnosis.Results For patients with incision infection,it was found that diabetes mellitus,preoperative RBC,hemoglobin,total protein,albumin,serum calcium,operation time,number of segment fusion,intraoperative blood loss,postoperative WBC,RBC,hemoglobin and platelet were significantly different from those with non-infection group.Moreover,multivariate logistic analysis showed that diabetes mellitus (OR=3.716,P=0.032),preoperative serum calcium (OR< 0.001,P=0.001),intraoperative blood loss (OR=1.002,P=0.014),postoperative hemoglobin (OR=0.923,P=0.018) and postoperative platelet (OR=1.007,P=0.017) were independent risk factors for postoperative incision infection.For patients with spinal canal hematoma,it was found that patients' preoperative total protein,albumin,serum calcium,platelet,operation time,intraoperative blood loss and postoperative total protein were significantly different from non-hematoma group.Multivariate logistic analysis showed that preoperative serum calcium (OR< 0.001,P=0.001),preoperative total protein (OR=1.298,P=0.043),intraoperative blood loss (OR=1.003,P=0.021) and postoperative total protein (OR=1.080,P=0.028) were independent risk factors for postoperative spinal canal hematoma.Conclusion The preoperative diabetes mellitus,serum calcium,intraoperative blood loss,postoperative hemoglobin and platelet were important risk factors for lumbar incision infection.And preoperative serum calcium,total protein,intraoperative blood loss and postoperative total protein were the risk factors for spinal canal hematoma.
4.Comparison of serum specific IgE and skin prick test in allergic rhinitis patients sensitive to dust mite.

OBJECTIVE:
To investigate the correlation of serum specific IgE and skin prick test in allergic rhinitis patients sensitive to dust mite and the difference between the results.
METHOD:
Data of 349 allergic rhinitis patients who had positive result in either Dermatophagoides pteronyssinus(Der p) or Dermatophagoides farinae (Der f) by serum specific IgE and skin prick test were statistically analyzed.
RESULT:
Grades of Skin prick test and specific IgE levels were notably relevant in these two dust mites(Der p r= 0. 568, Der f r= 0. 506, P<0. 01). There was significant difference of positive rates between serum specific IgE and skin prick test in Der p(Chi2 = 11. 605, P<0. 01)but not in Der f(Chi2 =0, P>0.05). There was no correlation between positive degree of two methods and score of clinical symptoms in allergic rhinitis.
CONCLUSION
Serum specific IgE and skin prick test were notably relevant in allergic rhinitis patients sensitive to dust mites. The positive rates of the two methods were different due to different allergen. Therefore, they could not substitute for each other. The level of serum specific IgE and positive degree of skin prick test could not reflect the degree of symptom in allergic rhinitis.

OBJECTIVE:
To study the effects of subcutaneous allergen immunotherapy by Alutard SQ allergy vaccination in management of perennial allergic rhinitis.
METHOD:
Thirty-eight cases of the perennial allergic rhinitis received the subcutaneous allergen immunotherapy by Alutard SQ allergy vaccination for 3 years or more than 3 years. The clinical symptoms and signs were compared before and after treatment.
RESULT:
Good result were obtained in 21 of 38 cases, 7 cases effective, and 10 cases ineffective. Total effective rate was 73.68%.
CONCLUSION
The subcutaneous allergen immunotherapy by Alutard SQ allergy vaccination was very effective for the perennial allergen rhinitis.

6.Protective Effect of Liraglutide on Hypoxia and High Glucose-induced Oxidative Stress Injury in Cardiomyocyte

Objective:To explore the effects and possible mechanism of liraglutide on hypoxia and high glucose-induced oxidative stress injury in cardiomyocytes. Methods:The neonatal rat cardiomyocytes were separated and cultured in vitro. The hypoxia and high glucose-induced injury model was established in neonatal rat cardiomyocytes. The cells were divided into six groups:the normal control group, liraglutide control group, hypoxia and high glucose model group, liraglutide treatment group, GLP-1R antagonist group and hyperosmotic control group. The metabolic ability of the cells was detected by MTT assay, the activities of LDH and CK-MB were detected by colorimetric method,SOD activity and MDA content were determined by xanthine oxidase method and thiobarbituric acid method,ROS level was measured by chemiluminescence method. The mRNA and protein expression of adaptor protein p66Shc was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. Results:Compared with those in the normal control group, the cells in hypoxia and high glucose model group had poorly metabolic ability,the content of LDH, CK-MB, MDA and ROS increased (P < 0.01), the activity of SOD decreased (P <0.01), and the expression of adaptor protein p66Shc greatly increased(P <0.01). After the treatment with liraglutide,the above mentioned parameters were all improved(P < 0.01). Exendin(9-39),an antagonist of GLP-1R,attenuated the protective effect of liraglutide. Conclusion:Liraglutide has a protective effect on cardiomyocytes by down-regulating adaptor protein p66Shc expression and reducing ROS formation.
7.Predictive value of amplitude integration with electroencephalogram combined with the Scandinavian Stroke Scale in patients with acute cerebral infarction

Objective To study the value of amplitude integrated electroencephalogram (aEEG) combined with the Scandinavian Stroke Scale (SSS) in evaluating the prognosis of acute infarction.Methods One hundred and twenty-four patients with acute cerebral infarction admitted to our hospital from February 2019 to July 2021 were selected as the research objects.After admission,the amplitude integrated EEG and SSS scores were measured.After 28 days,the prognosis of the patients was assessed by the activity of daily living scale (Barthel Index).According to the scores,the patients were divided into the good prognosis group (Barthel index score>45) and the poor prognosis group (Barthel index score≤45).Logistic regression analysis was used to analyze the risk factors of poor prognosis.Draw the receiver operating characteristic (ROC) curve,and analyze the predictive value of AEEG and SSS scores for poor prognosis.Results The AEEG upper and lower boundaries,SSS score and GCS score in the poor prognosis group were lower than those in the good prognosis group (P<0.05),the proportion of brain hernia formation and multiple organ failure,and the levels of hs CRP and D-D in the poor prognosis group were higher than those in the good prognosis group (P<0.05).Logistic regression analysis showed that AEEG upper and lower bounds,brain hernia formation,multiple organ failure,SSS score at admission and GCS score at admission were all risk factors for poor prognosis (P<0.05).ROC curve analysis showed that the sensitivity,accuracy and AUC of aEEG upper and lower boundaries combined with SSS score in predicting poor prognosis of patients with acute cerebral infarction at admission were 92.35%,89.57% and 0.829 respectively.Conclusion aEEG and SSS scores are independent risk factors for poor prognosis in patients with acute cerebral infarction,and the combination of the two has a high predictive value for the prognosis of acute cerebral infarction.

According to the pathological characteristics of symptomatic chronic thoracic and lumbar osteoporotic vertebral fracture (SCOVF), the different clinical treatment methods are selected, including vertebral augmentation, anterior-posterior fixation and fusion, posterior decompression fixation and fusion, and posterior correction osteotomy. However, there is still a lack of a unified understanding on how to choose appropriate treatment method for SCOVF. In order to reflect the new treatment concept and the evidence-based medicine progress of SCOVF in a timely manner and standardize its treatment, the clinical guideline for surgical treatment of SCOVF is formulated in compliance with the principle of scientificity, practicability and advancement and based on the level of evidence-based medicine.
9.Study on protective effect and mechanism of Arisaema Cum Bile on acute liver injury induced by CCl4 in mice

OBJECTIVE To investigate the protective effect and potential mechanism of Arisaema Cum Bile on acute liver injury induced by carbon tetrachloride (CCl4) in mice. METHODS Fifty mice were randomly divided into normal group, model group, positive control group (Biphenyl diester dropping pills, 150 mg/kg), Arisaema Cum Bile low-dose and high-dose groups (0.78, 2.34 g/kg), with 10 mice in each group. The mice in each group were given relevant medicine intragastrically, once a day, for 7 consecutive days. Two hours after the last administration, those groups were given intraperitoneal injection of 0.2% CCl4-olive oil solution to induce acute liver injury model except for normal group. Seventeen hours after intraperitoneal injection, the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), interleukin-6 (IL-6), tumor necrosis factor-α(TNF- α), and malondialdehyde (MDA), superoxide dismutase (SOD) in liver tissue were measured with kit method. The hepatic index was detected. The pathological changes of liver tissue were observed by HE staining, and the degree of liver injury was scored quantitatively. The mRNA expressions of TNF-α and IL-6 in liver tissue were detected by real-time fluorescence quantitative PCR; the protein expressions of Janus kinase 2 (JAK2), signal transducer and activator of transcription protein 3 (STAT3) and nuclear factor-κB p65 (NF-κB p65) in liver tissue were detected by Western blot assay. RESULTS Compared with normal group,the levels of ALT, AST, IL-6, TNF-α and MDA, the hepatic index were increased significantly (P<0.05), while the level of SOD was decreased significantly (P<0.05); the mRNA E-mail:qhwang668@sina.com expressions of IL-6 and TNF-α, and the protein expressions of JAK2, STAT3 and NF-κB p65 were up-regulated significantly (P<0.05); the pathological observation of liver tissue showed that the structure of hepatic cord was seriously disordered, there were many inflammatory cells infiltration of liver cells, and the liver injury score was significantly increased (P<0.05). Compared with model group, pathological changes and above indexes in mice were improved significantly in Arisaema Cum Bile low-dose and high-dose groups (P<0.05). CONCLUSIONS Arisaema Cum Bile has a protective effect on CCl4-induced acute liver injury in mice, which may be related to the inhibition of inflammatory response mediated by JAK2/STAT3/NF-κB signal pathway and antioxidant stress.
