1.Effects of different time courses and depths of acute normovolemic hemodilution on injury to mucous membrane of small intestines in rabbits

Objective To evaluate the effects of different time courses and depths of acute normovolemic hemodilution (ANH) with hematocrit (Hct) on injury to the mucous membrane of small intestines in rabbits.Methods Thirty-two adult rabbits,weighing 2.0-2.5 kg,were randomly divided into 4 groups (n=8 each) using a random number table:control group (group C),Hct 18% group (H1 group),Hct 15% group (H2 group) and Hct 12% group (H3 group).The animals were anesthetized with iv 20% urethane 4 ml/kg,tracheostomized and mechanically ventilated.Blood samples were taken from the left femoral artery.Blood withdrawn from the femoral artery =2 ×body weight× body fluid per kilogram × (initial Hct-target Hct) ÷ (initial Hct+target Hct),and was simultaneously replaced with the equal volume of 6% hydroxyethyl starch 130/0.4 infused over 30 min via the left femoral vein until the target Hct was achieved in H1,H2 and H3 groups.At 30 min of stabilization after puncture (T0) and 2,4 and 8 h after ANH (T1-3),blood samples were obtained from the superior mesenteric artery and vein for blood gas analysis and for determination of plasma concentrations of tumor necrosis factor-α (TNF-α) in the superior mesenteric vein.Oxygen extraction rate (ERO2) was calculated.Small intestinal mucosal tissues were obtained at T3 to observe the pathological changes with light microscope.Results Compared with those at T0,the plasma concentrations of TNF-α at T3 and ERO2 at T1-3 were significantly increased in H1 group,and the plasma concentrations of TNF-α at T3 and ERO2 at T1-3 were increased in H2 and H3 groups.The injury to the mucous membrane of small intestine was mild in group H1,and was severe in H2 and H3 groups.Conclusion ANH performed with 6% hydroxyethyl starch causes injury to the mucous membrane of small intestine when Hct is 18% and the time course ≥ 8 h,and when Hct ≤ 15% and the time course ≥ 2 h in the rabbits.

Objective To evaluate whether ?G-455A gene polymorphism increases the risk of coronary heart disease(CHD) due to elevated plasma fibrinogen.Methods A total of 1 485 patients who had received coronary angiography due to chest pain or suspected cardiac ischemia by non-invasive examination were included in the study.According to the angiographic results,all the patients were divided into the control group(n=466) and coronary heart disease group(n=1 019).Patients in the coronary heart disease group were further divided into stable angina pectoris group(SAP,n=674) and acute coronary syndrome group(ACS,n=345) according to their clinical presentation.We investigated G-455A polymorphism of ? fibrinogen gene and plasma fibrinogen level in all the patients.Results Increased plasma fibrinogen levels were observed in CHD groups compared with controls(ACS: 380.92?92.35 mg/dL,SAP: 352.49?94.89 mg/dL,control: 311.72?87.09 mg/dL,P
3.Home-based Care Service Package for Urban Disabled Elderly in Zhengzhou, China

Objective To construct a home-care service package for the urban disabled elderly that satisfy their demands and improve the efficiency of service. Methods From August to October, 2014, a total of 285 disabled (mild, medium and severe) elderly from a cluster sampling were investigated with the self-designed questionnaire about home-care demands among them. An initiatory service package was drawn to meet the demands reported by 40% of them or above. The package was consulted by a group of 15 experts using Delphi method. Results The final service package included 11, 19, 20 items of care for the mild, medium and severe disabled elderly, respectively. Conclu-sion The disabled elderly home-care package in Zhengzhou has been created based on the demands among disabled elderly.

Heterotopic ossification is a common complication after acetabular fractures and fractures of the elbow.Heterotopic ossification often leads to severe joint movement disorder,which brings great pain to the patient.This paper reviewed the clinical research,including pathogenesis,clinical diagnosis,prevention,treatment and future directions of heterotopic ossification to investigate the effective method in prevention and treatment of heterotopic ossification.

6.Comparison of immediate therapeutic effect and safety between esmolol and lanatoside in controlling ventricular rate for patients with rapid atrial fibrillation

Objective:To compare the immediate therapeutic effect and safety between esmolol and lanatoside in con-trolling ventricular rate for patients with rapid atrial fibrillation.Methods:A total of 72 patients with rapid atrial fi-brillation (ventricular rate ≥120 beats/min)were randomly and equally divided into esmolol group and lanatoside group.Esmolol group received first dosage of 0.5 mg/kg intravenously for 1 min,then were observed for 5 min,if ventricular rate still >100 beats/min or decreased <20%,the 0.5 mg/kg esmolol was appended,meanwhile main-tained by 0.05mg·kg-1 ·min-1 via micro pump at first,maintenance dose can increase to a maximum 0.3mg· kg-1 ·min-1;lanatoside group received first dosage of 0.4mg or 0.2mg slow intravenous injection,if ventricular rate still >100 beats/min or decreased<20%,the another 0.2mg was appended.The onset time and adverse reac-tions were recorded after administration.Results:Compared with lanatoside group,there was significant reduction in mean onset time [(39.2±8.7)min vs.(5.6±3.1)min]in esmolol group,P<0.01;compared with before adminis-tration,there was significant decrease in ventricular rate in two groups after administration,P<0.05 or<0.01. On 2h af-ter administration,compared with lanatoside group,there was significant increase in decreasing range of ventricular rate (29% vs.39%)in esmolol group,P<0.05. The total effective rate and adverse reaction rate was no significant difference (P>0.05)between two groups.Conclusion:Intravenous using esmolol is efficient and safe,and it can be regarded as pre-ferred therapy to control ventricular rate in patients with rapid atrial fibrillation.

Objective To investigate the effect of tripterygium on the grow th ,invasion and angiogenesis of melanoma A375 and its action mechanisms .Methods MTS was used to test the effect of tripterygium on proliferation of A375 melanoma;the nude mouse subcutaneous melanoma xenograft model was used to detect the effect of tripterygium on tumor growth ;the Transwell ex‐periment was used to determine the effect of tripterygium on invasion of A 375 melanoma;the tubule formation experiment was used to determine the effect of tripterygium on tumor angiogenesis ;ELISA was used to detect the influence of tripterygium on A 375 cel‐lular secretion factors ,such as VEGF ,bFGF ,TGF‐βand IL‐8 .Results The in vitro proliferation and in vivo growth of A375 mela‐noma cells after tripterygium treatment were significant inhibited ,the invasion ability of A375 melanoma cells was significant weak‐ened compared with the control group ;tripterygium could inhibit tumor cell‐induced vessel formation by down‐regulating the ex‐pression of VEGF ,bFGF and IL‐8 proteins ,but it had no influence on expression of TGF‐βprotein .Conclusion Tripterygium has anti‐growth and anti‐invasion effects on A375 melanoma ,its potential mechanisms may associated with the inhibition of tumor an‐giogenesis of A375 melanoma .

Objective To study the relationship between the changing law of cytokine gene expression profile and tumor-related fatigue and to verify the preliminary theory conclusion that the pathogenesis of tumor-related fatigue may be due to the up-regulation of the TNF,TGF-?,etc.Methods The changing characteristics of gene expression profile of cytokines were studied in peripheral blood mononuclear cells samples with gene chip.Results The gene expression level of TNF-? and TGF-?1 was up regulated.Conclusions The experimental results conform to the theory research conclusion and show that the pathogenesis of tumor-related fatigue due to the up-regulation of the gene expression level of TNF and TGF-?.
9.Effects of different doses of gabapentin on streptozotocin-induced diabetic neuropathic pain

Objective To investigate the effects of different doses of gabapentin on streptozotocin (STZ)-induced diabetic neuropathic pain in rats.Methods Male SD rats aged 6 weeks weighing 180-200 g were used in this study. Diabetes ntellitus ( DM) was induced by intraperitoneal STZ 60 mg/kg and confirmed one week later by blood glucose =16.7 mmol/L before breakfast. The DM rats were randomly divided into 4 groups ( n = 6 each) : gabapentin groups received intraperitoneal gabapentin 30, 60 and 120 mg/kg twice a day (at 9:00 am and 3:00 pm) for 3 weeks respectively and control group received intraperitoneal normal saline 0.6 ml instead of gabapentin. The paw withdrawal threshold to von Frey filament stimulation was measured before and at 30, 60, 120, 180, 240 min after first gabapentin injection and once a week for 3 weeks. Results After gabapentin 60 and 120 mg/kg, the paw withdrawal threshold to mechanical stimuli was significantly increased and lasted for about 4 h. The analgesic effect peaked at 60 min after IP gabapentin injection. Normal saline and gabapentin 30 mg/kg had no significant analgesic effect. The degree of analgesia was significantly decreased at day 14 and 21 of treatment with gabapentin 60 and 120 mg/kg as compared with that at 60 min after gabapentin injection. Conclusion The hyperalgesia and allodynia in rats with diabetes mellitus can be effectively reversed by gabapentin 60 and 120 mg/kg,while long-term use of gabapentin can induce drug tolerance.
