1.The mechanism of the cerebral protective effect of erythropoietin and angiotensin receptor blocker

Objective To study the effects of erythropoietin(EPO) and angiotensin receptor blocker on the expression of eNOS and Bcl-2 after cerebral ischemia.Methods The model of focal cerebral ischemia was made by occluding middle cerebral artery(MCA) for 2h and reperfusing for 48h in healthy Sprague-Dawley rats.The rats received EPO 3 000u · kg-1 · d-1 or valsartan 40mg · kg-1 · d-1 by intraperitoneal injection just before the beginning of reperfusion and after reperfusion.The expression of eNOS and Bcl-2 was detected.Results Compared with ischemia group,Bcl-2 and eNOS expressions in EPO-treated group and EPO-ARB treated group were significantly increased (P < 0.05).eNOS expression in ARB-treated group was significantly increased (P < 0.05),Bcl-2 expression was up-regulated,but it had no significant difference.Conclusion EPO had protective effects on cerebral ischemia-reperfu-sion injury in rats,which partially mediated by up-regulating of Bcl-2 and eNOS expression.Angiotensin receptor blocker had protective effects on cerebral ischemia-reperfusion injury in rats,which partially mediated by up-regulating of eNOS expression.
2.The effects of 4℃ hypertonic saline on S100 protein in serum and brain tissue of rats after cardiac arrest

Objective To investigate the effects of 4 °C hypertonic saline (HTS) on S100 protein in serum and brain tissues of rats after cardiac arrest (SCA). Method Thirty SD male rats were randomly divided into shame-operated group (A), NS group (B) ,4℃ NS group (C), HTS group (D) and 4℃. HTS group (E), in e-qual number ( n = 6). Drugs were given to the rats of all groups at the initiation of CPR except group A. The rat model of CA was induced by asphyxia. Over 24 hours after restoration of spontanous circulation ( ROSC), venous blood sample was drawn to detcect the concentration of serum S100 protein in each group, and the rats were sacrificed and their brain tissues were taken for comparing the expressions of S100 protein in hippocampus. One-way analysis of variance and q -test were used for comparison among groups. P < 0.05 was considered significant. Results Compared with group A, the concentration of serum S100 protein in other groups were much higher ( P < 0.01). Compared with group B,the concentrations of serum S100 protein in groups C, D and E were also much lower ( P < 0.01). Compared with groups D and E, the concentration of serum S100 protein in group C was much higher ( P < 0.01). Compared with group D, the concentration of serum S100 protein in group D was higher ( P < 0.05). Compared with group A, the expressions of S100 protein in rats brain tissues of groups B,C and D were much higher ( P < 0.01). The expression of S100 protein in brain tissue of rats in group E was also higher than that in rats of group A ( P < 0.05). Compared with group B, the expressions of S100 protein in brain tissues of rats in groups C,D and E were lower (P < 0.05 and P < 0.01). Compared with group C, the expressions of S100 protein in brain tissues of rats in groups D and E were lower (P < 0.01). Compared with group D, the expression of S100 protein in brain tissue of rats in group E was lower (P < 0.05). Conclusions After CA the 4℃ HTS can decrease serum S100 protein level and inhibit the expression of S100 protein in hippocampus, then protecting the brain tissue.

4.Influence of users' cognition on information retrieval behaviors in ubiquitous knowledge environment

Users' cognition and information retrieval are faced with great challenges in ubiquitous knowledge environ-ment. The influence of cognitive factors such as personality traits, cognitive style and ability, knowledge structure and ascribed methods on users' information retrieval behaviors was thus analyzed. It was pointed out that different cognitive factors could influence the retrieval, judgment and absorption of its results,users' satisfaction and continuous use in ubiquitous knowledge environment,with suggestions put forward from the aspects of information retrieval education, information service and information users for improving the users ' knowledge system in knowledge assimilating process by working out effective retrieval strategies using the advantages of cognition.
5.Docetaxel combined with intraperitoneal hyperthermic perfusion chemotherapy and hyperthermia to cure advanced ovarian cancer

Objective:This study aimed to observe the curative effect and adverse reaction of docetaxel combined with intraperitoneal cisplatin chemotherapy and hyperthermia treatment of advanced ovarian cancer. Methods:A total of 83 patients with inoperable and recurrent advanced ovarian cancer were randomly divided into two groups:hyperthermia group and control group. The hyperthermia group consisted of 42 cases of docetaxel chemotherapy immediately treated with intraperitoneal cisplatin chemotherapy combined with abdominal local hyperthermia. The control group included 41 cases of docetaxel chemotherapy and intraperitoneal cisplatin chemotherapy treatment only. Results:The total efficiencies of the hyperthermia treatment group and the control group were 81%and 58.1%, respectively, which showed that the total efficiency significantly improved (P<0.05). The ascite control rates were 78.3%and 66.7%and CA125 decreased by 84.2%and 61.5%for the hyperthermia and control groups, respectively. The main adverse reactions were gastrointestinal reaction and bone marrow suppression. However, differences were not statistically significant. Conclusion:Docetaxel combined with cisplatin intraperitoneal perfusion hyperthermia significantly improved the curative effect on advanced ovarian cancer without increasing toxicity, which indicates that it is a treatment worth popularizing.

Objective To investigate the expression of protein interacting with Cα kinase 1 (PICK1) and its significance in colorectal adenocarcinoma.Methods The expression of PICK1 were detected by immunohistochemistry and Western blot methods in tissues of colorectal adenocarcinoma,colorectal adenoma,colorectal polyp and adjacent tissues.The correlation between PICK1 and clinical pathological data was explored.Results Immunohistochemical assay showed that the positive ratio of PICK1 protein was 72.5 % (74/102),and overexpressed in 31 cases (30.4 %,31/102) with colorectal adenocarcinoma.The ratio of high expression level of PICK1 in colorectal adenoma tissues (22.2 %,8/36) was significantly higher than that in the adjacent tissues (0,0/40) (P < 0.05).The ratio of high expression level of PICK1 in colorectal polyp tissues (5.6 %,2/36) was no statistically difference compared with that of the adjacent tissues (P > 0.05).Western blot analysis revealed that the expression of PICK1 in colorectal adenocarcinoma (1.10±0.04) was significantly higher than that in the adjacent tissues (0.75±0.07) (P < 0.05).The result showed significant correlation with the tumor location,the degree of differentiation,depth of invasion,TNM stages and lymph metastasis (all P < 0.05).However,there is no correlation was revealed between PICK1 expression and the patients age,gender (both P > 0.05).Conclusion The expression of PICK1 may be associated with the tumorgenesis,progression,invasion and metastasis of colorectal adenocarcinoma,which contributes to the prognosis of patients.
8.The effects of Telbivudine for the prevention of perinatal transmission of hepatitis B virus infection from pregnant women to newborn on the expression of IL-17A,IL-10,IL-6 in the maternal plasma,umbilical plasma,neonatal plasma

Objective To explore the expression of interleukin -17A(IL-17A),interleukin-10(IL-10), interleukin-6 ( IL-6 ) in the maternal plasma , umbilical plasma , neonatal plasma from the pregnant women who accepted the Telbivudine for the prevention of perinatal transmission of hepatitis B virus infection .Methods 85 preg-nant women who were delivered in the hospital were chosen [ Telbivudine group 25 cases,HBVDNA high viral level group,HBVDNA negative group,non-infection group(health pregnant),each group included 20 cases].The level of the serum of IL-17A,IL-10,IL-6 in the maternal plasma,umbilical plasma,neonatal plasma from the different group was prospectively analyzed by the CBA method .Results The levels of IL-17A in the maternal plasma,um-bilical plasma,neonatal plasma of the Telbivudine group [(15.29 ±4.80)pg/mL,(19.45 ±4.80)pg/mL,(17.32 ± 4.18)pg/mL]were lower than the HBVDNA high viral level group [(26.03 ±4.88)pg/mL,(34.07 ±5.59)pg/mL, (24.12 ±4.76)pg/mL],there were significant differences(P=0.000,0.001,0.019);The level of IL -10 in the maternal plasma,umbilical plasma,neonatal plasma of the Telbivudine group [(6.08 ±1.32) pg/mL,(2.33 ± 0.68)pg/mL,(3.74 ±0.61)pg/mL]were higher than the HBVDNA high viral level group [(3.95 ±1.21)pg/mL, (1.71 ±0.53)pg/mL,(2.22 ±0.55)pg/mL],there were significant differences(P=0.023,0.015,0.012);The level of IL-6 in the maternal plasma ,umbilical plasma ,neonatal plasma of the Telbivudine group were lower than the HBVDNA high viral level group,there were significant differences (all P <0.05).Conclusion Telbivudine can cause the inhibition of the replication of HBV virus , partially reverse the immunologic dissonance of the pregnant women with HBV virus ,which is conducive to maintaining pregnancy .
9.Genotyping and Homology Comparison of Meticillinresistant Staphylococcus aureus Isolated from Earthquake Victims

OBJECTIVE To investigate the genetic feature of meticillin-resistant Staphylococcus aures isolated from earthquake victims and compare them with those of common patients.METHODS The MRSA isolates underwent SCCmec(Staphylococcal Cassette Chromosome mec) multi-PCR and PVL gene detection.Diversilab method was used for homology comparison and antibiotic resistance was also analyzed.RESULTS All the MRSA isolates were confirmed to be type Ⅲ SCCmec with PVL gene negative.Homology analysis showed relatively consistent with community-acquired MRSA confirmed to be ST8 by MLST typing.CONCLUSIONS There may be cross-spread of MRSA between hospital and community.We should pay more attation even in emergent time.
10.Effects of acarbose versus fenofibrate on insulin sensitivity and β cell secretion in impaired glucose tolerance with hypertrigtyceridemia

Objective To investigate the effects of acarbose versus fenofibrate on insulin secretion and insulin resistance in the subjects with impaired glucose tolerance (IGT) and hypertriglyceridemia. Methods Eighty subjects were allocated to acarbose group (28 cases),fenofibrate group (30 cases),and control group (22 cases) without intervention for 3 months,and also divided into elderly (46 cases) and younger groups (34 cases). Fasting blood samples were collected for measuring fasting plasma glucose and lipid.Oral glucose tolerance test (OGTT) were carried out with measurement of plasma insulin and glucose before ad after treatment.Early insulin secretion indexes(△I30/△G30),insulin secretion indexes (HOMA -β) and insulin resistance indexes (HOMA-IR)were calculated. Results After 3-months of treatment,the lipid profile was evidently improved in fenofibrate group. Levels of triglyceride (TG) and total cholesterol (TC) were significantly reduced ( both P < 0.01 ),△I30/△G30 was significantly increased (P < 0.05) and HOMA-IR was decreased (P<<0.01).In acarbose group,levels of fasting plasma glucose (FPG),2hours postprandial plasma glucose (2 hPG) and HOMA-IR were reduced (all P<0.01),△I30/△G30 and HOMA-β were enhanced (P<0.01 or P<0.05).No change of above indicators was found in the control group.Compared with fenofibrate group,acarbose group had higher △ I30/△G30 (P< 0.05),HOMA-β (P< 0.01 ) and lower HOMA-IR ( P < 0.01 ).The improvement of △△I30/△G30 was correlated with the decreasing of plasma FPG,2 hPG and TG(r=0.5812,0.6327,0.3872,P<0.01),while HOMA-1R was related with the decreasing of plasma 2 hPG,TG and TC(r=0.8126,0.4671,0.2895,P< 0.01). HOMA-IR,△I30/△G30 and HOMA-β were lower (P<0.01) and acarbose improved insulin resistance better (P<0.05) in the elderly than in the younger with type 2 diabetes. Conclusions Pancreatic β-cell function declines gradually with aging,acarbose may have advantage over fenofibrate in improving early insulin secretion and resistance which was more relieved by diminishing glucotoxity in comparison with lipotoxity.
