ObjectiveTo explore the clinical significance and characteristics about terminal sialyl residues linked to N-glycans in the tissue and bile glycoproteins from bile duct carcinoma. MethodsThe mRNA expression of sialyltransferases, including ST3Gal-Ⅲ and ST6Gal-Ⅰ, was detected in the tissue extracts of 35 cases with bile duct carcinoma(BDC) and 35 cases of benign biliary disease (BBD) by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR). With the lectin of WGA(Triticum valgaris agglutinin), which can recognise the terminal sialyl residues in N-glycans, all the specimens of BDC and BBD were subject to lectin histochemical staining, followed by computer-aided digital analysis. Lectin dot-blotting with horseradish peroxidase-labeled WGA was performed for the bile samples from each of the 35 cases of BDC and BBD. Objective:To observe the therapeutic effect of recombinant human brain natriuretic peptide (rhBNP)on patients with cardiorenal syndrome.Methods:The data of 75 patients,who were diagnosed as cardiorenal syndrome and hospitalized in our hospital,were retrospectively analyzed.They were randomly divided into routine treatment group (n=40,received routine treatment)and rhBNP group (n=35,received rhBNP based on routine treatment) according to number table method.The rhBNP was pumped with 0.0075μg·kg-1 ·min-1 using micropump intrave-nously,once/day,about 10h/time and 7d was regarded as a course of treatment.Changes of 24h urine volume,N terminal pro brain natriuretic peptide (NT-proBNP),glomerular filtration rate (GFR)and echocardiograph were recorded in all patients before and 7d after treatment.Results:Compared with routine treatment group after treat-ment,there were significant increase in total effective rate (62.5% vs.94.3%),24h urine volume [(785.2 ± 143.4)ml vs.(965.34±171.8)ml],GFR [(34.1±2.6)ml/min vs.(45.2±5.6)ml/min]and left ventricular e-jection fraction [(35.6±5.5)% vs.(45.9±6.8)%],and significant reduction in NT-proBNP level [(3451.1± 1314.2)pg/ml vs.(1516.43 ± 431.52)pg/ml]in rhBNP group,P<0.01 all.Conclusion:Recombinant human brain natriuretic peptide is safe,effective and can improve renal function in treating patients with cardiorenal syn-drome.
5.An animal experimental study on access to mediastinum with flexible endoscope via suprasternal fossa

Objective To evaluate the feasibility and safety of mediastinum examination with flexible endoscope via suprasternal fossa.Methods A total of 20 healthy pigs were enrolled.Mediastinocosopy was performed via a tunnel to thoracix through a 1.5 cm incision on suprastemal fossa.Mediastinum was observed and complications were recorded.Results Mediastinocosopy was successfully achieved in all pigs.No severe bleeding or death occurred during the procedure.Five pigs got fever after the procedure with elevated white blood cell count.Four pigs had pneumothorax and three of them died in 1-6 days.Conclusion Mediastinocosopy with flexible endoscope through suprastemal fossa is feasible to diagnose mediastinal diseases,which can be important experimental evidence for the mini-invasive therapy of mediastimum diseases.
6.THE RELATIONSHIP BETWEEN THE N-GLYCOSYLATION OF ACETYLGLUCOSAMINYLTRANSFERASE V AND ITS ACTIVITY

Objective. The goal of this paper is to investigate the relationship between the N-glycosylation of acetylglucosaminyltransferase V(Glc NAcT-V) and its activity and to know which site among the 6 N-glycosylation sites in the GlcNAcT-V gene is the most important.Methods. Wild type of GlcNAcT-V was transfected into COS-7 cells and its activity was measured 48 h later. The first site (Asn 110) was mutated with site-directed mutagenesis and transfected into COS-7 cells.Results. It was found that after the cells were added tunicamycin(TM, 1 μ g/ml), the activity was 11.7% of the wild type. The activity of the cells with mutating GlcNAcT-V was about 12.0% of the wild type. RT-PCR showed that there was no significant change in mRNA expression among the three groups.Conclusion. The N-glycosylation is important for its activity. Our results suggest that the N-linked carbohydrates on GlcNAcT-V are required for the posttranscriptional activity of the enzyme.
7.The drug sensitivity of human lung adenocarcinoma stem cells to cisplatin and carboplatin

Objective:To analyze the drug sensitivity of human lung adenocarcinoma stem cells (LASC) to cisplatin (DDP) and carboplatin (CBP). Methods:Human lung adenocarcinomaic cells SPC-A1,AG,and CPA-Y2 were treated with DDP and CBP. The cell viability of cells was detected by CCK-8 assay. The phenotypic characteristics of drug surviving cells(DSCs)were determined by immunofluorescence staining. The LASC population was then separated by magnetic-activated cell sorting method. The LASC in DSCs was traced by using green fluorescence protein (GFP). The drug sensitivity of DSCs to DDP and CBP was analyzed.Results:The LASC exhibited the phenotypes of bronchioalveolar stem cells (BASC, OCT4~+CCSP~+SP-C~+). After mixture of CD221~+LASC with CD221~-lung adenocarcinoma differentiated cells, the DSC population showed OCT4~+BASC phenotypes. These DSCs were significantly resistant to DDP and CBP.Conclusion:LASC has a high resistance to DDP and CBP. This may be the reason for tumor recurrence after chemotherapy.
8.The study of interleukin-6 and interleukin-18 concentration in patients with metabolic syndrome

Objective To investigate the association of serum interleukin-6 and interleukin-18 concentration in patients with meta-bolic syndrome (MS), and explore the role of inflammtory markers in these patients. Methods 83 patients with MS were enrolled into study group, among of them, 47 were male and 36 were female, their average was 48.95±10. 17 years. The control group was made of 85 normal volunteers. The levels of fasting blood glucose (FBG), fasting insulin, plasma lipids, interleukin-6 and interleukin-18 were meas-ured in all the patients and normal subjects. The insulin sensitive index was calculated. The levels of the fasting blood inflammatory markers were compared between the two groups. Results Compared with the control group, the HOMA-βof the MS group was significantly de-creased, but the levels of IL-18 and IL-6 were significantly increased. The serum IL-18 levels were positive correlated with FPG, FINS, TG, HOMA-IR, IL-6. Conclusion There are IR and abnormal serum level of IL-18 and IL-6 in patients with MS. It has an intimate association between abnormal serum IL-18 and IR. The inflammatory reaction may play an important role in the development of IR in patients with MS.
9.Investigation and analysis of iodine nutritional statusin healthy adults in Urumqi of Xinjiang Uyghur Autonomous Region

Objective To investigate iodine nutrition status in healthy adults in Xinjiang Urumqi city and their relationship to ethnicity, gender and age. Method A cross-sectional survey was performed in 2 100 residents of Xinjiang Urumqi 2 communities in May 2013, of which 1 835 healthy adults aged 18-84 years, mean age 46.3 ± 14.2 years were enrolled. Urine iodine with arsenic-cerium catalytic spectrophotometry and salt iodine and water iodine of the residents were measured. Result The water iodine content was 3.14 μg/L, salt iodine was 27.75 mg/kg, 1 835 urine samples were collected, the urinary iodine median(MUI) was133.4 μg/L, iodine deficiency accounted for 32.37% (595 cases), iodine sufficient 42.94% (788 cases), iodine super-sufficient 14.44%(265 cases), iodine excess 10.19%(187 cases). The urine iodine level in Han ethnic group was higher than those in Uygur adults, no significant difference was observed between men and women (P>0.05). Urinary iodine levels among different age groups had no statistical difference, but in the older groups the level was lower than that in young or middle age adults. The urinary iodine level decreased gradually with the age increasing. Conclusion There was a sufficient pattern of iodine nutrition levels in healthy adults in Xinjiang Urumqi; there was a correlation between the level of iodine nutrition and the ethnicity but not with gender;iodine nutrition level in older age group was lower than those in younger age groups. Monitoring the urinary iodine continually and decreasing iodine intake may be necessary for part of iodine excess population.
10.A longitudinal study regarding the gestational changes in iodine nutrition and thyroid function among pregnant women in the iodine deficient areas of Henan province

Objective To characterize the gestational changes of iodine nutrition and thyroid function and to explore the factors associated with the thyroid function in pregnant women.Methods A longitudinal survey was conducted in 130 pregnant women in Luohe city of Henan province from October 2012 to May 2013.Samples of fasting blood and urine were collected in each trimester to test on thyroid function and urinary iodine.Data regarding social demography and lifestyle behavior were collected through questionnaire in the first trimester.Results The medians of urinary iodine (MUI) for pregnant women were 238.9,150.8 and 306.4 μg/L in the first,second and third trimesters,respectively (P<0.05).With the increase of gestational age,the level of free triiodothyronine (FT3) showed no significant change (P>0.05) but the level of free thyroxine(FT4) decreased (P<0.05),with the level of thyroid stimulating hormone (TSH) increased and then declined (P<0.05).A U-shaped curve were seen between iodine nutrition and thyroid function.With the increase of iodine level,the level of TSH first increased and then decreased while the levels of FT3 and FT4 showed the opposite trend.The level of TSH was influenced by factors as education level,history of chronic diseases,history of CT and X-ray examination,and intake of pickled food etc.The level of FT4 was associated with residence (urban or rural),stressful events in the previous year,daily means of transportation,and the hours of sedentariness,working and sleeping.Conclusion Significant differences were noticed in iodine nutrition and thyroid function of pregnant women during the three trimesters.It was essential to establish specific reference ranges for different trimesters.Thyroid functions of pregnant women seemed to be associated with iodine level and lifestyle.
