1.Effects of Hedysari radix polysaccharide on the behavior and central neurotransmitters in SAMP8

Objective To investigate the improving effect of Hedysari radix polysaccharide on the behavior and its influence on cerebral central neurotransmitters in senescence accelerated mouse prone 8 (SAMP 8).Methods The senescence accelerated mice 8 were randomly divided into a model group,a Hedysari radix polysaccharide (HRP) group and a aricept group.SAMRl were served as the control group.After intragastrical treatment for 3 months,the behavior changes and the levels of norepinephrine (NE),dopamine (DA),5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were observed respectively by applying Morris water maze and high performance liquid chromatography (HPLC).Results In the SAMP8 model group,the hidden platform test showed that the latency to find the hidden platform was remarkably prolonged,the latency range from third day to fifth day was (55.31 ±9.62) s,(58.67± 10.89) s,(58.45± 10.53) s,respectively.The level ofNE、DA、5-HT and 5-HIAA was (4.97± 1.74) mmol/L,(1.63±0.57)mmol/L,(4.29±0.94)mmol/L,(2.91±0.85)mmol/L respectively,significantly reduced (P<0.01).Compared with model group,the latency of hidden platform test was (44.73 ± 9.83) s,(40.53±8.37)s,(44.75±9.16)s respectively,which were remarkably shortened.The expression level ofNE,DA,5-HT and 5-HIAA in the HRP treatment group was (7.58± 1.62) mmol/L,(3.25 ±0.70) mmol/L,(7.04±0.95) mmol/L,(4.62±0.79) mmol/L respectively,which obviously increased (P< 0.05).Conclusion HRP could improve learning and memory ability of SAMP8 and the effect may related to up-regulate the expression level of cerebral Central neurotransmitters.
2.Influence of Hedysari Radix Concentrations Fluid on Learning and Memory and Monoamine Neurotransmitters in Senescence Accelerated Mice

Objective To explore the influence of Hedysari radix concentrations fluid (HRCF) on learning and memory and its effect on cerebral monoamine neurotransmitters in senescence accelerated mice. Methods The senescence accelerated mice were randomly divided into the senescence accelerated mouse/resistance 1 (SAMR1) control group, senescence accelerated mouse/prone-8 (SAMP8) model group, HRCF treatment group and aricept treatment group. After 3 months of intragastrical treatment, the learning and memory ability changes and content of norepinephrine, dopamine, 5-hydroxytryptamine and 5-hydroxyindoleacetic acid were detected respectively using Morris water maze and high performance liquid chromatography. Results In the model group, the hidden platform test indicated that the latency to find the hidden platform was remarkably prolonged, and the content of norepinephrine, dopamine, 5-hydroxytryptamine as well as 5-hydroxyindoleacetic acid were significantly reduced (P<0.01). Compared with the model group, the latency of hidden platform test was remarkably shortened (P<0.05) and the content of norepinephrine, dopamine, 5-hydroxytryptamine as well as 5-hydroxyindoleacetic acid were obviously increased in HRCF treatment group (P<0.01). Conclusion HRCF could improve learning and memory ability of SAMP8 and the influence may related to increasing the content of cerebral monoamine neurotransmitters.
3.Effect of modified Wenjing Decoction on reproductive hormones, 5-HT and ?-EP in patients with menopathy of excess-cold syndrome

AIM: To observe the treatment effectiveness of menopathy of excessive-cold syndrome by Modified Wenjing Decoction, and to explore the relationship among excessive-cold syndrome of menopathy and 5-HT,?-EP, and reproduetive hormones. METHODS: Sixty patients with menopathy of excess-cold syndrome were successively treated with Modified Wenjing Decoction for three menstrual cycles. The efficacy and the changes of serum 5-HT, Bendorphine(?-EP) and reproductive hormones before and after treatment were observed. RESULTS: 22 patients(36.7%) were short-term therapeutic effect, 28 patients(45.7%) were effective and 10 patients(17.6%) were ineffective. 4.Therapeutic Effect of External Application of Shuangbai Powder for Patients with Wounded Limb Injured by Venomous Snake and Nursing Experience

Objective To compare the therapeutic effect of conventional treatment and conventional treatment plus external application of Shuangbai Powder for patients with wounded limb injured by venomous snake. Methods One hundred patients bitten by venomous snake were randomized into treatment group and control group, 50 cases in each group. The control group was given conventional treatment including repeatedly washing the wound with hydrogen peroxide, debriding the wound, letting blood and draining toxicity, local blocking with chymotrypsin, wet packing the wounded limb with magnesium sulfate, and injection with anti-venomous serum, tetanus antitoxin, antibiotics, furosemide and energy mixture. The treatment group was given external application of Shuangbai Powder on the basis of the treatment for the control group. Both groups were given the comprehensive nursing of psychological nursing, wound nursing, dietary nursing, defecation nursing and functional nursing. The swelling-subsiding time for the wounded limb and pain scores of visual analogue scale (VAS) in the two groups were compared. Results(1) After treatment, swelling-subsiding time for the wounded limb of the treatment group was shorter than that of the control group, the difference being statistically significant (P < 0.05). (2) After treatment for 4 days and at the end of the treatment, VAS scores of the two groups were decreased compared with those before treatment(P < 0.05), and the decrease of VAS scores in the treatment group after treatment for 4 days was superior to that in the control group (P < 0.05). Conclusion The conventional treatment plus external application of Shuangbai Powder is an effective therapy for patients with wounded limb injured by venomous snake by shortening swelling-subsiding time and relieving pain for the wounded limb .

OBJECTIVE: To evaluate the clinical efficiency of recombinant adenovirus p53 injection (rAd-p53) combining chemotherapy in the treatment of malignant body cavity effusion. METHODS: 50 cases with malignant body cavity effusion were randomly divided into 2 groups. The treatment group were given intracavitary administration of rAd-p53 1 ? 1012VP after puncture drainage, 48h later which were given intracavitary administration of 60mg/m2 cisplatin once a week for 3~4 weeks. 6.Expression of EB virus in peripheral blood mononuclear cells in children with systemic lupus erythematosus

Objective To discuss the role of EB virus (EBV)in the pathogenesis of systemic lupus erythematosus(SLE) in children through investigating the copies of EBV DNA and expression of EBV genes in peripheral blood mononuclear cells(PBMCs).Methods (1)PBMCs were isolated from 30 patients with SLE and 12 healthy normal controls respectively and DNA was extracted from PBMCs.(2) PBMCs were co-cultured with EBV for 12 days and RNA was extracted from PBMCs.(3)Real-time fluorescence quantitative PCR(Real-time PCR) was applied to detect the copies of EBV DNA in PBMCs.(4)Reverse transcription PCR was applied to detect expression of EBV genes.Results (1) Compared with the healthy control group [(40.1 ± 11.6) copies/μg],a significant increase of EBV DNA copies was observed in SLE group[(658.6 ± 183.6) copies/μg] (P <0.05).The EBV DNA copies in the active SLE group [(785.2 ± 179.2) copies/μg] were significantly higher than those in the non-active SLE group [(586.0 ± 193.1) copies/μg] (P < 0.05).(2)There was no correlation between EBV DNA copies and systemic lupus erythematosus disease activity index (r =0.03,P > 0.05).(3) After PBMCs got co-cultured with EBV,expression of latent EBV genes and lytic genes were both increased in the patients and healthy controls.The latent EBV genes including latent membrane protein 1 (LMP1),LMP2,EBV nuclear antigen 1 and the lytic genes including BCRF1,BLLF1 were all increased significantly in the patients compared with the healthy controls (all P < 0.05).Conclusions There is a significant increase of EBV DNA copies and aberrant expression of EBV genes in SLE patients,which suggests that EBV may contribute to the pathogenesis of SLE.
7.Aberrant expression of Epstein-Barr virus genes in children with systemic lupus erythematosus

Objective To investigate the expression and significance of Epstein-Barr virus (EBV) genes in children with systemic lupus erythematosus (SLE).Methods Peripheral blood mononuclear cells (PBMCs) were isolated from 20 children with SLE and 12 healthy human controls.Enzyme-linked immunosorbent assay (ELISA) was conducted to detect anti-EBV viral capsid antigen (VCA) IgG/IgM antibodies.The culture supernatants of cells from patients with anti-EBV VCA IgG/IgM antibodies were collected,and PBMCs from the patients and controls were co-cultured with the supernatants respectively for 12 days.RNA was extracted from PBMCs before and after the coculture,and reverse transcription-PCR was performed to detect the expression of EBV genes,including LMP1,LMP2,EBNA1,BCRF1,BLLF1 and BILF1 genes.Results LMP1 gene was detected in fresh PBMCs from 10 out of 20 patients and 1 out of 12 controls (P < 0.05).No significant differences were observed between the patients and controls in the detection rate of LMP2 gene (4/20 vs.1/12),EBNA1 gene (13/20 vs.3/12),BCRF1 gene (3/20 vs.1/12) or BLLF1 gene (5/20 vs.2/12) in fresh PBMCs.After co-culture with the supernatants of cells from patients with anti-EBV VCA IgG/IgM antibodies,the expressions of EBV genes in these PBMCs were increased to different degrees,and there was a significant difference in the expressions of EBV latent genes LMP1,LMP2 and EBNA-1 as well as EBV replicative genes BCRF1 and BLLF1 between the patient-derived and control-derived PBMCs (all P < 0.05).Conclusions There is an aberrant expression of EBV genes in children with SLE,and EBV genes may contribute to the development of SLE.
8.Risk Factors for Osteoporosis in Patients with Hemiplegia

Objective To identify the risk factors for osteoporosis in patients with hemiplegia. Methods The total body bone mineral density (BMD) of 366 patients with hemiplegia in the stage of rehabilitation was measured with dual-energy X-ray absorptiometry. The data of the patients were analyzed with multiple logistic regressions. Results The incidence of osteoporosis was 15.3 % (56/366) in the cohort. ≥60 years old (OR=5.434, 95% CI:2.734~10.801), duration of disease (OR=1.046, 95% CI:1.013~1.080), body mass index (BMI) (OR=0.746,95% CI:0.622~0.840),ADL level (OR=0.618, 95% CI: 0.444~0.858) were independently correlated with osteoporosis in patients with hemiplegia. Conclusion Patients with hemiplegia in the stage of rehabilitation were at a high risk of osteoporosis. Aged over 60 years old, lower BMI, longer duration of disease and ADL level were independent risk factors for osteoporosis, which called for supervision and prophylaxis on osteoporosis.
9.Changes of Total Body Bone Mass and Bone Mineral Density in Patients with Hemiplegia

Objective To investigate the changes of bone mineral density (BMD) and bone mass and the influencing factors in patients with hemiplegia.Methods The total BMD, bone mass, lean mass and fat mass of 366 patients (313 stroke cases and 53 TBI cases) with hemiplegia were tested by dual-energy X-ray absorptiometry, as well as the clinical features of all patients, were studied.Results The bone masses in the paretic side (upper limb, lower limb and trunk) decreased significantly compared with nonparetic side ( P<0.01); the differences of bone masses (%) in upper limb, lower limb and trunk between paretic and nonparetic side decreased with duration of disease or trauma ( P<0.01). The multivariable stepwise regression analyses showed that the total bone mass and total BMD were negatively correlated with age and duration of disease significantly (all P<0.01), and positively correlated with body mass index (BMI) significantly ( P<0.01), the total BMD was also positively correlated with lean mass ( P<0.01) and ADL level ( P<0.05).Conclusion The bone mass deceases in the paretic side significantly, and gets worse with the duration of disease. Age, duration of disease or trauma and BMI influence total BMD and bone mass; in addition, total BMD is correlated with total lean mass and ADL level.

Objective To investigate the effect of idarubiein to imensify the MAC (IMAC) as preparative regimen for autologous peripheral blood stem cell transplantation(APBSCT)in acute myeioid leukemia.Methods Fourteen patients with acute myeloid leukemia who underwent APBSCT were analysed. IMAC was used as preparative regimens.Results All patients were engrafted successfully.The disease-free survival could be Ben in 8 cases(57.1%),the median disease-free survival duration were 26(8-72)months and no treatment-related mortality was present.Conclusion This study suggested that addition of Idarubicin to the MAC preparative regimens Was safe,effective and feasible for patients with acute myeloid leukemia,and may improve disease-free survival and overall survival.
