2.Establishment of FSP27 Gene Knockdown Preadipocyte Cell Line

Obesity and its related metabolic diseases become major health problems in the world.Adipose tissue plays an important role in the development of obesity.FSP27,a member of the CIDE family proteins,is expressed at high levels in white adipose tissue and differentiated 3T3L1 cells.The objective of current study is to establish a FSP27 knockdown preadipocyte cell line to investigate the in vivo function of mouse FSP27.The double strand siRNA of mouse FSP27 corresponding to nucleotides 270 to 291 was synthesized and inserted into pSilencer2.1.pSilencer-siFSP27 was co-transfected into 293T cells with the HA-mFSP27 expression vector to test its knock-down efficiency.The FSP27 siRNA was then transferred to a lentiviral vector.Lentivirus were generated and used to infect 3T3-L1 cells.It was shown here that lentivirus containing FSP27siRNA can effectively knockdown FSP27 expression in 3T3-L1 cells.Establishment of FSP27 knock-down cell line provides a useful tool for the study of in vivo function FSP27.

This paper detailed the Coleman concept and practice of coach-led transitional care of elderlies,team members and their responsibilities,intervention aspects and main tasks,as well as the intervention program and its effect analysis.Then the authors analyzed the present transitional care development in China,and recommended on development of a likewise care model for senior chronic disease patients in the country,such as building multi-disciplinary transitional care teams to provide uninterrupted care and service,encourage patients and caregivers to embrace self-management of diseases,and build an appraisal mechanism to ensure quality of care.

7.Clinical Feature and Glycoprotein D Gene Sequence Analysis of a Wild Strain of HSV-2 Isolated from One Relapsed Patient with Genital Herpes

Objective To study the relation between the clinical feature and glycoprotein D gene sequence analysis of a wild strain of HSV 2 isolated from one relapsed patient with genital herpes. Methods The partial glycoprotein D gene sequence of the above mentioned strain was amplified and cloned with PCR. Results The comparison of the amino acid sequence of gD gene between the wild strain and HSV 2G strain published showed that there was a mutation at site80 and site159. Conclusion In order to develop effective vaccine which is suitable for China, it is necessary to investigate the differences of the gene structure and function of gD among HSV isolated from China and other countries.
8.Corrosion resistance of orthodontic magnet coated with titanium nitride in artificial saliva

Objective: To study the corrosion resistance of orthodontic magnet with titanium nitride coating in artificial saliva. Methods: 6 samples of magnet in the size 3 mm?5 mm?7 mm were prepared.3 of them were coated with titanium nitride by plasma enhanced chemical vapor deposition,another 3 without coating were used as controls.The samples were immersed into artificial saliva at 37 ℃ for one month. Then the surface characters of two kinds of magnets were observed with the naked eye and by electron-probe microanalysis (EPMA). Results: The surface of the magnets with titanium nitride coating was fine and close in texture and there were no cleft and disintegrated area. However, that of the magnets without titanium nitride coating was uneven and coarse in texture,and there were evident clefts and disintegrated areas. Conclusion: The corrosion resistance of orthodontic magnets with titanium nitride coating is stronger than that without coating.
