1.Investigation of gene expression profiles in patients with blood stasis syndrome

OBJECTIVE: To investigate the differential gene expression profiles in patients with blood stasis syndrome by oligonucleotide microarray technique. METHODS: Sixteen patients with blood stasis syndrome were divided into patients with coronary heart disease (CAD) (n=8) and non-CAD patients (n=8) by using coronary angiography. The sex- and age-matched eight healthy persons were enrolled as control group. Venous bloods were collected for extracting RNA. Test-3 chip was first employed to examine the quality of samples. Then the samples were hybridized with Affymetrix U133 Plus 2.0 array to compare the gene expression profiles among the three groups. Gene-array scanner and gene chip operating software were applied to screen hybridization signals and analyze gene expression respectively. Based on the comparison of the three groups of samples, the differential genes related with blood stasis syndrome were analyzed by Gene Ontology (GO) and pathway, and confirmed by real-time reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Forty-eight differential genes were found being associated with blood stasis syndrome, including 26 up-regulated genes and 22 down-regulated genes. Five of the forty-eight genes (10.4%) were related to inflammatory reaction and immune response through the GO analysis. In the pathway analysis, five of ten significant pathways were referred to inflammation and immune response. The results of real-time RT-PCR proved the accuracy of the gene chip. CONCLUSION: Inflammatory- and immune-related genes have a remarkable predominance in blood stasis syndrome gene expression profiles, which may explain the function of inflammation and immune response in the occurrence and development of blood stasis syndrome.
2.Development of Correspondence Adapter Based on CAN

Objective To transform the equipment with traditional RS232 interface into that with CAN connection interface.Methods On the CAN main line,node is the station for information receiving and transmission in the network.The intelligent node can establish the work way ID,the address and the baud rate through the programming.It is mainly composed by the singlechip and the programmable CAN correspondence controller.Results The hardware and software were designed.SJA1000 initialization,transmission and receiving subroutines are including in the software design.Conclusion CAN is a kind of excellent field bus with high reliability,low cost and has been applied in many fields.It has already formed the international standard and been viewed as one of the most widely used field buses in future.Lots of traditional equipment with RS232 interface are facing the problem of CAN interface transformation.
3.Ultrastructural Characteristics of Amelanotic Melanocytes from Human Hair Follicles

Objective To investigate the ultrastructural characteristics of amelanotic melanocytes (AMMCs). Methods Individual hair follicles from normal human scalp were digested with collagenase type V, then washed in phosphate buffer saline. Hair-follicle cell suspensions were prepared by trypsin and cultured in a medium suitable for melanocyte growth. The keratinocytes were removed by differential trypsinization. Geneticin (100?g/mL) was used to eliminate contaminating fibroblasts. After 3 passages the cells were trypsinized, washed in phosphate buffer saline, and finally processed for transmission electron microscopy. Results Under transmission electron microscope, the cultured cells were round or oval-shaped with a single large nucleus and double-layered karyotheca. Abundant euchromosome but sparse heterochromosome was observed within the nucleus. There were various organelles in the cytoplasm, including mitochondria, rough endoplasmic reticulum (RER), ribosomes and abundant melanosomes of nearly uniform size. The electronic density granules distributed in a concentric pattern in most of the melanosomes. Colgi complexes were inconspicuous in the cells. Conclusions Compared to epidermal melanocytes, AMMCs from human hair follicles have different ultrastructural characteristics which implies their functional immaturity. AMMCs may serve as the depot for mature melanocytes.
4.Effects of Ellagic Acid on Melanogenesis and Melansome Transfer

Objective To investigate the effect of ellagic acid on human epidermal melanocyte melanogenesis and melanin transfer, and the mechanism thereof. Methods The human melanocytes and and keratinocytes were co-cultured and purified. After passing the second generation, cells of 1∶10 ratio were inoculated into the small dish (3 cm × 3 cm). The changes of melanin content and tyrosinase activity in melanocytes were detected before and after intervention with ellagic acid (100, 10 and 1 mg/L) for 48 h. The melanin transfer in cultured cells was detected by flow cytometry method. The 10 nmol/L arbutin was used as the positive control. Results The tyrosinase activity was down-regulated by ellagic acid in a dose-dependent manner. The ellagic acid can reduce the melanin content except for the 1 mg/L of ellagic acid. The melanin transfer was also inhibited by ellagic acid in a dose-dependence manner. Conclusion Ellagic acid can be used for skin-whitening cosmetic and the depigmenting effect might be due to the down-regulation of melanogenesis and melanin transfer.
5.Metrology management software for hospital metrology room

7.EFFECT OF IODINE DEFICIENCY AND HYPOTHYROIDISM ON NO CONTENTS IN DEFFERENT PARTS OF BRAIN IN RATS

Objective: This experiment was designed to study the change of nitric oxide (NO) content in different parts of brain in rats suffering from iodine deficiency(ID) and hypothyroidism since their fetal period. Methods: Iodine deficiency animal model was made and NO content in hippocampus(HPC), cerebellum (CE) and cortex(CO) were measured by using NO test kit. Results: Urinary iodine and serum T 3?T 4 concentrations significantly decreased and learning-memory capacity was impaired in ID rats. NO content was significantly lower in HPC and CE in ID rats than in control rats.(HPC:11.61?2.31/2.3?1 18 ?mol/g Prot;CE:10.77?5.16/4.70?1.95 ?mol/g Prot).No difference was found in cortex. Conclusion: Iodine deficiency and hypothyroidism since fetal period could affect NO level in rat brain. Different parts of brain have different sensitivity to ID and hypothyroidism.The changes of NO content in hippocampus may contribute to the learning-memory impairment in ID rats.
8.Effects of low frequency repetitive transcranial magnetic stimulation on motor function and affective disorder in patients with Parkinson's disease

Objective To investigate the therapeutic effects of low frequency repetitive transcranial magnetic stimulation (rTMS) on motor function and affective disorder in patients with Parkinson's disease (PD). Methods Twenty PD patients were performed by 1 Hz rTMS therapy for 15 days and 10 matched patients were performed sham stimulation. Hamilton Depression Scale (HAMD), Hamilton Anxiety Scale (HAMA) and Unified Parkinson' s Disease Rating Scale (UPDRS) were assessed before and 15 days after rTMS treatment. Results After treatment with rTMS, the motor function was improved, and UPDRS total score and UPDRS-Ⅰ , UPDRS-Ⅱ, UPDRS-Ⅲ scores were decreased. UPDRS score was decreased from (38.45±17.33) to (30.95± 17.00) (t=6.780,P<0.01). At the same time, HAMD score was decreased from (12.15±7.62) to (8.75±7.31 ) (t = 5.101, P<0.01 ). The scores of somatization, blockage and sleep disorders were lowered after treatment, but the HAMA score had no significant change(t=1.757, P>0.05). The rigidity of PD patients was improved obviously. All indexes had no improvement in sham stimulation group. Conclusions Low frequency rTMS may improve motor dysfunction and affective disorder in PD patients .
9.Manifestation study on depression in comprehensive hospitals

Objective To study and analyze clinical manifestation of depression in comprehensive hospitals in China.Methods Retrospective study was performed.3322 objects included in this research were measured by Minnesota Multiphasic Personality Inventory(MMPI).Results In Chinese comprehensive hospitals,the most common accompanying symptoms of depression were anxiety,fatigue,suicide inclination,apprehension,somatic discomfort,impotence or asexuality,hypochondriasis,somato-complaints,insomnia,self-accusmion and self-reproach,sense of dependence and so on.Depression was positively correlated with suicide risks,self-accusation,delusion of self-accusation,somatic discomfort,somato-complaints,impotence and asexuality,insomnia,headache,fatigue,apprehension,and family problems.Depression had negative correlations with self-strength,social status and economic status.The results had statistical significance.Conclusion Decreased self-strength,increased sense of dependence,low social status and economic status and family problems were the most common pathogenesis basis for depression.Depression can be manifested as various somatic disorders,especially insomnia.
10.Preparation of Polyacrylamide Resin and Its Adsorption Properties on 2,4-Dichlorophenoxyacetic Acid

Polyacrylamide resin was synthesized via surface-initiated atom transfer radical polymerization ( SI-ATRP) method. Acrylic amide ( AM) was grafted onto the surface of the chloromethyl polystyrene resin via SI-ATRP in the CuBr/2, 2'-bipyridine ( Bpy) system as catalyst at room temperature. The compositions of polyacrylamide resin were determined by means of elementary analysis, FT-IR analysis and scanning electron microscopy ( SEM) . The adsorption properties, the parameters of kinetics and the thermodynamics of the resin were evaluated in details, respectively. As the results, adsorption capacity of 2,4-dichlorophenoxyacetic acid (2,4-D) increased with the initial concentrations of solution increasing at room temperature, and its highest adsorption capacity was 111. 0 mg/g with solution concentration of 8 mmol/L. Adsorption isotherm at room temperature was determined and modeled with Langmuir and Freundlich equations. The thermodynamic equilibrium functions were calculated to be ΔG<0,ΔH=268. 2 kJ/mol, ΔS>0, hence, the adsorption was spontaneous, endothermic and entropy increasing. The kinetics fitted the pseudo-second-order well. The polyacrylamide-chloromethyl polystyrene (PAM-CMCPS) resin was used for the adsorption of 2,4-D in orange sample, and good results were obtained.
