Objective:To compare the anti-obesity effect of green tea and black tea polyphenols and investigate their molecular mechanisms. Method:Rats were divided randomly into four groups:control group,high-fat group,high-fat diet with green tea polyphenols(GTP) supplement group, and high-fat diet with black tea polyphenols(BTP) supplement group. Body weight was determined every 2 w. After 3 months, the changes of epididymal fat tissues weight and serum lipids were observed. Expressions of those genes associated with adipocyte differentiation in epididymal fat tissues of rats were measured by real-time transcription-polymerase chain reaction, including pref-1,aP2,TNF-?,leptin,PPAR-?,C/EBP-? . Results:Both GTP and BTP prevented the increase of body weight and fat induced by high-fat diet and profoundly down-regulated those adipocyte-specific genes, including aP2,TNF-?and leptin. In addition,GTP also up-regulated the pre-adipocye marker — pref-1 and reduced the expression of transcription factor, peroxisome proliferators-activated receptor(PPAR-?) . Conclusion:Tea polyphenols could prevent obesity by reversing the adipocyte differentiation, and GTP possessed stronger inhibitory effect than BTP.

BACKGROUND: Two-dimensional electrophoresis (2-DE) is precise and effective for the isolation and analysis of complex protein mixtures, has become one of valuable key techniques for proteome. Mice models were as research subjects to model human disease status, one of important methods for cardiovascular disease. The establishment and optimization of 2-DE for proteomes of mice myocardium will provide basis for heart disease.OBJECTIVE: To establish and optimize a stable technique of 2-DE for proteomic analysis of mice myocardium.DESIGN: Mice were used as research subjects, observational comparative study.SETTING: Department of Pathology, Nanfang Hospital Affiliated to Southern Medical University.PARTICIPANTS: The experiment was performed at the Department of Pathology, Nanfang Hospital Affiliated to Southern Medical University between February and September 2004. Totally 12 male BABL/c mice of 4-5 weeks without special-pathogen free with the body mass of 12-15 g were selected, which was provided by the Experimental Animal Center of Southern Medical University.METHODS: The mice were put to death by dislocation of cervical vertebra under drugged state to obtain heart. Various protein extraction method, loading sampling buffer, strip range of IPG, sampling volume of protein, staining method and so on were compared, analyzed, scanned and digilized, and then performing image analysis. Each link in the study was established and optimized.MAIN OUTCOME MEASURES: Resolution of protein and reproducibility of the trial.RESULTS: A method that was optimal for cardiac tissue can isolate about a total of 910 protein spots on pH 4-7 gel strips. 200 μg was fit for silver stain while 1000 μg was for Coomassie stains. It was found that in the myocardium of normal mice about (920±30) protein spots were obtained in 17 em pH 3-10 L gel strip, while around (880±30) were gained in the pH 4-7 L gel strip. The mean matching rate achieves to 86. 9%.CONCLUSION: The findings of the trial showed that 2-DE technique can be effectively applied for proteomics of myocardium of mice to regulate and optimize, and relatively stable separation method of myocardial protein was established.

OBJECTIVE:To optimize the placement of extra-machine medicines in the automated pharmacy and establish a ra-pid dispensing area to shorten medicine dispensing time. METHODS:12 588 prescriptions made within one week were collected from the outpatient pharmacy of our hospital,in which 29 kinds of extra-machine medicines used frequently were clustered on the basis of Pearson correlation coefficients. The extra-machine medicines used at a higher frequency were arranged in the storage posi-tions near to the medicine dispensing window, and those related to the above-mentioned medicines were stored in the positions adja-cent to them, thereby a rapid dispensing area was established. The effect of the above-said method was evaluated by comparing the time it took for the pharmacist to dispense medicines one week before and after the establishment of the rapid dispensing area. RE-SULTS:29 kinds of extra-machine medicines were clustered into 4 major categories, and stored in 4 positions in the rapid med-icine dispensing area respectively, and then the position codes were assigned. After the rapid dispensing area was established, the average single prescription dispensing time for the pharmacist decreased by about 9 s (40.43 s vs. 31.43 s,P<0.01). CONCLU-SIONS:The establishment of the rapid dispensing area based on cluster analysis method has increased the efficiency of dispensing medicines in the outpatient pharmacy of our hospital.
