Medical Journal of Chinese People's Liberation Army 2001;0(07):-
Objective To investigate the correlation between HDL-C/TG ratio and cardiovascular risk in elderly patients with hypertension, in order to set up an evaluation criterion which may objectively assess the cardiovascular risk in elderly population. Methods A survey was carried out among 2011 subjects aged 60 or over, who received health examination in the General Hospital of PLA in May 2008. Hypertension was diagnosed by using the definition recommended by WHO/ISH. The subjects were divided into 4 groups (male hypertesion group, male control group, female hypertension group and female control group) according to their gender and with or without hypertension. The parameters of hematology (systolic pressure, diastolic pressure, pulse pressure and mean arterial pressure) and the indicators of lipidemia [total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C)] were measured, and HDL-C/TG ratio was calculated. Logistic regression analysis was used to analyze the correlation between the indicators of lipidemia and cardiovascular risk. Results In both male and female hypertension groups, systolic pressure, diastolic pressure, pulse pressure and mean arterial pressure showed significant differences compared with those of the control groups gender-wise (P0.05). Other indicators of lipidemia showed no significant differences between hypertension group and control group. Logistic regression analysis showed that HDL-C/TG ratio was significantly correlated to the cardiovascular risk (P

Objective To identify the possible mutations other than A1555G in mitochondrial 12S and 16S rRNA genes that responsible for aminoglycoside-induced deafness, and to provide the basis for genetic diagnosis for this disease. Methods A total of twenty-seven blood samples were obtained from five families with aminoglycoside-induced deafness for screening the A1555G mutation and other possible mutations by PCR- Alw26 I digestion and sequence analysis. Results All samples examined in four families (A, C, D and E) carried the same homoplasmic A1555G mutation, but no A1555G mutation was found in family B. Sequencing of the DNA samples from this family displayed a rare insertion of "AA" at nucleotide 2227 in 16S rRNA gene. Conclusions Our findings suggested that the 1555 G mutation was not the only genetic defect associated with aminoglycoside-induced deafness since we did not find the A1555G mutation in one family, in which the typical maternal inheritance pattern of the aminoglycoside-induced deafness was seen. It is not enough to identify prospectively patients who are likely to be hypersensitive to aminoglycoside ototoxicity by screening A1555G mutation only. Other possible mutations in mitochondrial DNA that associated with aminoglycoside -induced deafness should be tested also.

Objective:To compare the anti-obesity effect of green tea and black tea polyphenols and investigate their molecular mechanisms. Method:Rats were divided randomly into four groups:control group,high-fat group,high-fat diet with green tea polyphenols(GTP) supplement group, and high-fat diet with black tea polyphenols(BTP) supplement group. Body weight was determined every 2 w. After 3 months, the changes of epididymal fat tissues weight and serum lipids were observed. Expressions of those genes associated with adipocyte differentiation in epididymal fat tissues of rats were measured by real-time transcription-polymerase chain reaction, including pref-1,aP2,TNF-?,leptin,PPAR-?,C/EBP-? . Results:Both GTP and BTP prevented the increase of body weight and fat induced by high-fat diet and profoundly down-regulated those adipocyte-specific genes, including aP2,TNF-?and leptin. In addition,GTP also up-regulated the pre-adipocye marker — pref-1 and reduced the expression of transcription factor, peroxisome proliferators-activated receptor(PPAR-?) . Conclusion:Tea polyphenols could prevent obesity by reversing the adipocyte differentiation, and GTP possessed stronger inhibitory effect than BTP.

Medical Journal of Chinese People's Liberation Army 2001;0(08):-
To investigate a huge family with autosomal dominant hereditary non syndromic hearing loss. In this family, sixty six of 113 family members and 8 spouses have been conducted physical examination, pure tone audiometry, immittance testing and auditory brainstem response testing (ABR). The results indicated that 37 of 66 tested family members have sensorineural hearing loss in various degrees. No associated visible abnormalities in other systems were found in this family. The mode of inheritance of this family should be autosomal dominant according to its pedigree. The full collections of both blood samples and physiological hearing assessments of this family have provided the solid basis for future study on identifying disease causing gene.

AIM: To study the effect of high fat diet on the expression of sterol regulatory element biding protein-1 (SREBP-1) and transforming growth factor β_1 (TGF-β_1) in renal tubular cells and rosiglitazone intervention. METHODS: Wistar rats were treated with high fat diet and rosiglitazone for 3 months. The serum glucose, serum insulin and serum triglyceride were detected. Oil Red O staining was used to observe the renal lipid deposit and Masson staining was for the detection of ECM accumulation. SREBP-1, TGF-β_1 and FN protein were determined by the methods of immunohistochemistry and Western blotting. SREBP-1 mRNA was detected by in situ hybridization. RESULTS: Rosiglitazone prevented effectively the increase in serum glucose, serum insulin and serum triglyceride resulted from high fat diet. High fat diet led to lipid droplet formation in renal tubular cells and interstitial ECM accumulation, which was decreased by rosiglitazone treatment. Compared to normal rats, SREBP-1 protein and SREBP-1 mRNA showed high expressions in high fat diet rats that were lowered by rosiglitazone. The precursor segment and mature segment of SREBP-1 protein were decreased by 27.39% and 27.32%. Similarly, the high expressions of TGF-β_1 and FN protein in kidney of high fat diet rats were also prevented by rosiglitazone intervention. Compared to high fat diet rats, the expression of TGF-β_1 in rosiglitazone treatment rats was lowered by 19.14%. CONCLUSION: Rosiglitazone prevents effectively the over-expression of SREBP-1 and TGF-β_1 in renal tubular cells, and decreases lipid accumulation and ECM production in rats fed with high fat diet.
