1.Research progress on the molecular mechanism of breast cancer bone metastasis

Breast cancer is one of the most common malignant tumors in women. Bone is commonly affected in the context of metastatic breast cancer.Once bone metastasis happens, patient would experience poor prognosis and impaired quality of life.However, there is a lack of approaches for more sensitive and specific diagnosis and treatments for breast cancer bone metastasis.Thus, it is im-perative to find new treatment target from molecular mechanism.In this paper, we review the current research progress on the molecular mechanism from several levels including gene profile, proteins and microRNAs.We also review the establishment of animal models of breast cancer bone metastasis.With the achievements acquired in the completed or undergoing researches on breast cancer bone metasta-sis, we hope the finding of the optimal diagnostic and therapeutic targets could lead the breast cancer research into a new era.

Activated protein kinase ( AMPK) , an important energy receptor , plays a very important role in regulating organ-ism and the energy metabolism of the cell .AMPK has complex relationship with survival of different types of breast cancer cells .Ac-cording to different conditions , AMPK may exist both anti or promoting effect on tumor .In this review , we summarize the relationship between AMPK and breast cancer related signal pathways , AMP and breast cancer endocrine therapy , breast cancer chemotherapy , ra-diotherapy for breast cancer , treatment of triple negative breast cancer and multi drug resistance of the relationship , we also expound some drugs related to AMPK and used in clinical setting .

Sox-2(SRY related HMG box-2)is an essential embryonic stem cell gene and a necessary transcription factor for maintaining the proliferation and self-renewal capability of embryonic stem cell .Sox-2 is involved in complex cell signal transduction pathway network and adjusting biological behavior of cells .Expression of Sox-2 is adjusted by different factors in transduction , transla-tion and post-translation levels .In recent years , increasing evidences indicate Sox-2 has an important role in tumor development , growth, proliferation, and metastasis, which is involved in various biological behavior of tumor cells in many types of tumors .This pa-per reviews the role of Sox-2 in adjusting tissue and tumor development and complex cell signal transduction pathway network of Sox -2 and clinical values of Sox-2 in tumor diagnosis and treatment until now .
4.Analysis of Amino Acids and Acylcarnitines in the Blood of Patients First Diagnosed with Uremia Using Tandem Mass Spectrometry

Objective Analyze the amino acids and acylcarnitines in the blood of patients first diagnosed with uremia using tandem mass spectrometry in order to provide a basis for clinical diagnosis and treatment. Methods 30 patients with uremia were selected as the research objects,while 15 cases of healthy person were served as the control. The values of 11 kinds of amino acids and 2 kinds of acylcarnitines in the two groups were detected using tandem mass spectrometry,and the data were analyzed and compared. Results The levels of alanine(Ala)and tyrosine(Tyr)in uremia group were significantly lower than those in control group(P < 0.05),while the levels of citrulline(Cit),glycine(Gly)and proline(Pro)were significantly higher in uremia group than in control group (P < 0.05). The differences were mainly in non-essential amino acids between the two groups. The levels of free carnitines and total carnitines in uremia group were significantly higher than those in normal group(P < 0.05). Conclusion For the patients first diagnosed with uremia(not treated with replacement therapy),the disturbance of amino acid metabolism mainly in non-essential amino acids were found in their blood,and the carnitine levels were higher than normal levels. The results could provide the basis for clinical nutrition therapy for patients with uremia.
5.Observation of ankle pump motor in the prevention of deep vein thrombosis after surgical operation

Objective To evaluate the influence of ankle pump motor on deep vein thrombosis after surgical operation.Methods 138 cases of patients after surgical operation who were assessed as high risk patients,were randomized into the study group (70 cases)and the control group (68 cases),routine nursing was adopted in the control group,while in the study group,ankle pump motor was adopted in addition.The incidence of deep vein thrombosis was compared between two groups.Results No deep vein thrombosis was found in the study group,while 1 case of thrombosis was detected in the control group,but there was no statistic difference.Conclusions Ankle pump motor was easy and convenient,and can prevent deep vein thrombosis by promoting the reflux of the lower limbs,improving the recovery of patients after operation,and increasing nursing quality as well.
6.Ethics management of clinical research application and self-evaluation of ethics from applicant in 2007 Capital Medical Development Scientific Research Fund

Objective To evaluate the ethical management status quo of Capital Medical Research Fund in 2007.Methods Cross-sectional study was applied to analyze the ethics management and ethics self-evaluation in applications.Results There were 652 applications,covering with 14 districts and 2 counties in Beijing,from 128 Hospitals.Applicants have some knowledge of the ethical issues in clinical research accounted for 88.7% ; informed consent of subjects considered in 72.5% ;the potential risk and protection involved in application accounted for 62.0%; personal privacy protection of subject accounted for 49.4%.Unfortunately,the benefit vs risk assessment was only involved in 28.7% applications.Applicants from general hospitals had more ethical issue knowledge (90.2%) than those from community hospitals (84.6%).Applicants from university hospitals had more ethical issue knowledge (93.9%) than those from military hospitals (80.2%).Applicants aged 55 0r over had less ethical issue knowledge (70.0%) than other applicants (89.2%).The applicants to study on descriptive research and etiological research had less ethical issue knowledge than others.578 applications filled out the approval from ethics committee or research management department.62.6% of the written were acceptable.Conclusion The applicants for clinical research had preliminary understanding for ethical issues in Beijing in 2007.Hospital ethics committees or research management departments had conducted clinical research ethical review applications for funds management.It has taken place the external conditions to carry out the ethical management in clinical research fund management.
7.EXPERIMENTAL STUDIES ON THE RENAL VESSELS LIGATION IN RABBITS

36 rabbits were divided into control and experimental groups.In the experimen-tal group,the left renal artery was ligated or the left renal artery and vein wereligated simultaneously.All the animals were alive.After 3—8 months of the opera-tion,the rabbits were killed.The establishment of collateral circulation and theanatomical,histological and histochemical changes of the kidny were observed.The resultsobtained were as follows:1.Ligation of the left renal vessels causes atrophy of theleft kidney on account of the failure of collateral circulation establishment.Theright kidney of the experimental group showed obviously compensatory hypertrophy.The weight of the compensatory hypertrophied kidneys obviously increased.2.Thecompensatory hypertrophic growth of kidneys is due to the hypertrophy of existingn(?)phrons and the hyperplasia connective tissue.3.comparison of the controls,inthe JGI,PAS and AlP reactions with the compensatory hypertrophied kidneys,there were no striking differences.The results suggest that the compensatory hyper-trophied kidneys were normal at least on the renin secretion and reabsorption func-tion.
8.Combinatorial protective effects of α-melanocyte-stimulating hormone and carboxymethylcellulose on ocular surface in dry eye rat models

Background Reasearches showed that α-melanocyte-stimulating hormone (α-MSH) inhibits inflammation and ameliorates the ocular surface abnormalities in a scopolamine-induced dry eye rat model,and the managing effect of sodium carboxymethylcellulose (CMC) on dry eyes also has been determined.However,whether α-MSH can enhance the therapeutic effects of CMC remains to be investigated.Objective This study was to investigate the protective effects of α-MSH combined with CMC on ocular surface in a scopolamine-induced dry eye rat model.Methods Sixty clean female Wistar rats were randomly divided into normal control group,model control group,NaCl group,CMC group,α-MSH group and α-MSH+CMC group,and 10 rats for each group.The dry eye models were established by subcutaneous injection of scopolamine hydrobromide at 9:00,12:00,15:00 and 18:00 per day for 28 days.0.9% NaCl solution,1×10 3 mg/ml α-MSH solution,0.5% CMC eye drop,and 1 ×10-3 mg/ml α-MSH+0.5% CMC solution were topically administered twice a day (8:00,17:00) since the initial day of modeling according to grouping.Shirmer Ⅰ test (S Ⅰ t),breakup time of tear film (BUT) and corneal fluorescence staining were performed before and 7,14,21,28 days after the application of drugs.At 28 days following the administration of drugs,the eyeballs of the rats were collected.Hemotoxylin and eosin staining was employed to examine the morphology of corneas,and periodic acid schiff (PAS) staining was used to count the conjunctival goblet cells.This study protocol was approved by Experimental Animal Ethic Committee of Tianjin Medical University (SYXK 2009-0001),and the use and care of the rats complied with ARVO Statement.Results The S Ⅰ t and BUT values were significantly reduced,and the corneal fluorescence staining scores were significantly increased over time following modeling in the model control group (all at P<0.01).No significant differences were found in the S Ⅰ t,BUT and corneal fluorescence staining scores between model control group and NaCl group at various time points (all at P>0.05).At 7,14 and 21 days after intervention,the S Ⅰ t values were (4.800±0.789),(4.100±0.516) and (4.300±0.856) mm in the α-MSH+CMC group,which were considerably higher than (2.875 ±0.719),(2.375 ±0.619) and (2.532±0.957)mm in the NaCl group (all at P<0.01).At 7 days after intervention,the BUT values were (4.938± 1.843) seconds and (5.000±1.491) seconds in the α-MSH group and α-MSH+CMC group,which were significantly higher than (3.250±1.000) seconds in the NaCl group (both at P<0.01).The corneal fluorescence staining scores in the CMC group,α-MSH group and α-MSH+CMC group were significantly lower than that in the NaCl group,with the lowest score in the α-MSH +CMC group (all at P<0.05).The thickening of corneal epithelial layer,corneal edema and arrangement disorder of corneal stroma were found in the model control group and NaCl group;while slight corneal edema and epithelial cell proliferation were exhibited in the α-MSH+CMC group by hemotoxylin and eosin staining.PAS staining showed that the number of goblet cells was much more in the CMC group,α-MSH group and α-MSH+ CMC group than that in the model control group and NaCl group (all at P < 0.01).Conclusions The sole application of α-MSH or CMC alleviates ocular surface damage and morphological abnormality to certain extent,and the combination of α-MSH and CMC generates more effective protection in comparison with sole administration of α-MSH or CMC.The early application of the drugs plays an improvement role in tear secretion and tear film stability in dry eyes.

The transcription activity of ectogenic human carcinoembryonic antigen (CEA) promoter in lung adenocarcinoma cells A549 was investigated for the further gene-targeting therapy. The reporter gene green fluorescent protein (GFP) driven by CEA promoter and human cytomegalovirus (CMV) promoter were relatively constructed and named plasmid pCEA-EGFP and pCMV-GFP respectively. The intensity of fluorescence was detected by fluorescence microscope and flow cytometry analysis after the pCEA-GFP and pSNAV-GFP plasmids were transfected into A549 cells through liposome respectively. The results showed (4.08+/-0.63) % of the A549 cells transfected with pCEA-AFP plasmid expressed, significantly lower than that of the A549 cells transfected with pCMV-GFP [(43.27+/-3.54) %]. It was suggested that ectogenic human CEA promoter in lung adenocarcinoma cells A549 was weakly expressed. The distinct specificity of CEA promoter in CEA high expression cells was regarded as a tool in selective gene therapy, but the transcription activity of ectogenic human CEA promoter was needed to increase in the future.
10.The role of PI3/Akt signaling pathway in the protective effect of morphine postconditioning on myocardium against ischemiareperfusion injury in rats

Objective To investigate the effect of morphine postconditioning on myocardial ischemiarepedusion(I/R)injury and the role of PI3K/Akt signaling pathway in the effect.Methods Seventy male SD rats weighing 280-330 g aged 16-17 weeks Were randomly divided into 5 groups(n=14 each):group Ⅰ sham operation(S);group Ⅱ I/R;group Ⅲ morphine postconditioning(M);group Ⅳ morphine postconditioning+ wortmannin(W+M);groupV wortmannin(W).Myocardial I/R injury wa.g produced by occlusion of anterior descending branch of left coronary artery for 45 min followed by 120 min reperfusion.In group M and W+M (groupⅢ,Ⅳ)morphine 1.25 mCkg was given iv at 3 min before and 2 min after reperfusion.In group W+M and W(groupⅣ,Ⅴ)wortmannin(a specific PDK inhibitor)15μ/gkg Was given iv at 20 min before ischemia. The animals were sacrificed at the end of 120 min repedusion for assessment of ischemic and infarct area and determination of total and phosphorylated Akt expression in myocardium by Western blot.Results There were no significant differences in the size of ischemic area and total Akt expression among the 5 groups. The infarct area was significantly smaller in group M than in group I/R. The were no significant differenees in the size of infarct area between group 1/R, W + M and W (group Ⅱ , Ⅳ,Ⅴ ). The phosphorylated Akt expression was significantly upregulated in group I/R and M as compared with group S, and was significantly higher in group M than in group I/R.Conclusion The PI3K/Akt signaling pathway activation is involved in the protective effect of morphine posteondifioning on myocardium against I/R injury.
