1.Evaluation of Medical Instruments Cleaning Effect of Fluorescence Detection Technique.

OBJECTIVETo compare the cleaning effect of automatic cleaning machine and manual cleaning on coupling type surgical instruments.
METHODSA total of 32 cleaned medical instruments were randomly sampled from medical institutions in Putuo District medical institutions disinfection supply center. Hygiena System SUREII ATP was used to monitor the ATP value, and the cleaning effect was evaluated.
RESULTSThe surface ATP values of the medical instrument of manual cleaning were higher than that of the automatic cleaning machine.
CONCLUSIONCoupling type surgical instruments has better cleaning effect of automatic cleaning machine before disinfection, the application is recommended.

2.Preparation and in vitro and in vivo release of ALA and HMME hydrogel suppository

Objective To prepare 5-aminolevulinic acid (ALA) and hematoporphyrin monomethyl ether (HMME) hydrogel suppositories and to evaluate their photosensitizer transfer efficiencies in rectal tumor tissue.Methods The BALB/c mice implanted SW837 rectal cancer cells subcutaneously were randomly divided into four groups:intrarectal suppository administration group,cutaneous administration group,intratumoral injection group and intravenous injection group.Fluorescence spectrophotometry was used to measure the concentration of protoporphyrin Ⅸ (PpⅨ) and HMME in rectal wall,skin and tumor tissue.The distribution of photosensitizer was determined by a fluorescence spectroscopy system.Results The concentration of PpⅨ in the ALA suppository administration group was 9.76 times (1 h) and 5.80 times (3 h) higher than that in the cutaneous administration group,and the differences were statistically significant (all P<0.05).The maximal penetration depth of ALA in tumor tissue was about 3-6 mm at 2 h after the cutaneous administration.After the HMME suppository administration,the concentration of HMME in the rectal wall was very low.The maximal penetration depth of HMME in tumor tissue was less than 2 mm after the cutaneous administration.Conclusions ALA is more likely to penetrate mucosal barrier compared to skin tissue.The hydrogel suppository based rectal administration is expected to be a new administration method for the rectal cancer photodynamic therapy using ALA.
3.Apoptosis of leukemia cells induced by dielectric barrier discharge plasma and its mechanism

Objective To explore the killing effect of dielectric barrier discharge (DBD) plasma on tumor cells and to analyze the DBD-induced apoptosis mechanism.Methods Thiazolyl blue tetrazolium bromide (MTT) assay method was used to detect the killing effect of low temperature plasma on the cytotoxicity of normal spleen leukocytes and acute promyelocytic leukemia cells (LT-12) at different doses.The changes of reactive oxygen species (ROS) level were measured after plasma treatment.The cell apoptosis rate was detected by Annexin V/PI double staining at different doses.The expression of apoptosis-related genes and proteins was detected by qRT-PCR and Western Blot.Results MTT results showed that the killing effect of plasma treatment was dose-dependent and time-dependent.The cell survival rate after 8 hours of treatment decreased from 98% to 63% with the dose increasing from 30 s to 240 s.The survival rate decreased from 78% (2 h) to 39% (24 h) after the treatment with a same dose (e.g.240 s).Annexin V/PI double staining results demonstrated that the plasma effect can induce apoptosis,and the apoptosis rate was not only positively correlated with the plasma dose,but also with the post-plasma time.The longer the post-plasma time,the higher was the apoptosis rate.The apoptotic rate of the 60 s dose treatment after 12 h was 48% that increased to 55.3% with the dose of 120 s.The production of reactive oxygen species (ROS) detected by flow cytometry also showed a time correlation of the plasma treatment.After the plasma treatment,the ROS level immediately increased to 1.24 times,and sharply increased to 5.39 times after 20 h post-plasma.The experimental results of qRT-PCR and Western blot showed that the expression of the genes and proteins of Caspase family and Bcl-2 family was very active at 8 to 12 h post-plasma treatment.Conclusions Low-temperature plasma can effectively kill tumor cells,and apoptosis is the main mechanism of death.The molecular mechanism of apoptosis of tumor cells induced by low temperature plasma was preliminary confirmed.
4.Prokaryotic expression, purification and activity analysis of recombinant human serine protease inhibitor Hespintor Kazal Domain.

Hespintor is an unknown function protein that was got from hepatoblastoma cell lines HepG2 by suppression subtractive hybridization technique (SSH), sequence analysis showed that the protein is a new member of secretory type of Kazal type serine protease inhibitor (Serpin) family, and has high homology with esophageal cancer related gene 2 (ECRG2). The coding sequence of Hespintor's Kazal domain was subcloned into prokaryotic expression vector pET-40b(+), then transformed into Rosetta (DE3). A recombinant protein about 42 kDa in the form of inclusion body was optimization expressed by inducing with 0.25 mmol/L IPTG, 30 degrees C for 5 h. and its specificity was confirmed via Western blotting. The recombinant protein was purified by metal chelate affinity chromatography (MCAC) and anion-exchange chromatography. The preliminary experimental result showed that the recombinant protein can inhibit trysin hydrolysis activity specifically. The result clearly demonstrated that Hespintor, as a novel member of Serpin, would be valuable in developing anti-tumor agents.

5.Endogenous testosterone levels and visuospatial ability

To describe the effects of endogenous testosterone levels on the visuospatial ability.Thevisuospatial ability,testosteronewere included as the key words,retrieving 46 papers in the database.35 relevant papers have been reviewed in the paper,as the results,we have found that the visuospatial ability of male was better than female.The performances of the visuospatial tasks in males were associated with the lev-els of endogenous testosterone,and the effects comprised two aspects:the organizational function and activa-tional function of the testosterone.The latter have received more attention from researchers.Until now,most studies of activational effects of testosterone on visuospatial ability remained on the level of behavioral re-search.The mental rotation task,navigation task and virtual reality scenes tasks were implied in these stud-ies.A nonlinear relationship has aroused a great controversy.Latest research suggests that further investiga-tion of the neural mechanisms related to visual-spatial ability is needed,using ERP and MRI techniques,to assess the relationship of endogenous testosterone levels with the relevant neural basis of visuospatial ability.
6.The treatment effect and adverse reactions of PDT on rabbits with rectal cancer in-situ

Objective To evaluate the influence of dosage,operation method,adverse reaction of endoscopic photodynamic therapy (EPDT) on its therapeutic efficacy in rabbit models of in-situ rectal cancer,so as to provide preclinical basis of photodynamic therapy for rectal cancer.Methods 20 rabbits with in-situ VX2 rectal cancer were randomly divided into control group,PDT low dose group,intermediate dose group,and high dose group.At 24 h before PDT,photosensitizer (hermimether) was intravenously injected into rabbits.630 nm semiconductor laser was used as light source.The growth of the tumor was observed by conventional endoscopy and endoscopic ultrasonography,and the survival time,general conditions and adverse reactions were recorded.The histopathological changes were observed by hematoxylin-eosin staining.Results At 7 d after PDT,the total response rates of low dose,intermediate dose and high dose group respectively were 40% (slight),80% (60% remarkable and 20% slight),100% (20% remarkable and 80% slight).The average survival times of the three groups were 14 d,10 d and 5 d,respectively.The main adverse reactions were inflammation,intestinal obstruction,intestinal peristalsis loss and death.Conclusions The dosage of PDT is an important factor to influence the curative effect.The appropriate dose of PDT will have a better effect on the treatment of rectal cancer.A thorough study of these problems is helpful to the clinical application of PDT in the treatment of rectal cancer.
7.Effects of applied course reform of emergency critical care nursing on self-directed learning ability of nursing undergraduates

Objective To explore the effects of applied course reform of emergency critical care nursing on self-directed learning ability of nursing students. Methods From September 2016 to January 2017, a total of 180 nursing undergraduates from Xi'an Peihua University of grade 2014 were selected as the research subjects. 86 students from 3 classes were randomly recruited into control group, and 94 students from other 3 classes to observation group. Control group was treated with traditional teaching methods, while experimental group adopted the applied course reform of emergency critical nursing. Nursing students' self-directed learning ability was compared between the two groups before and after the course. Results After the whole course, the total score of students' self-directed learning, learning motivation,planning and implementation,self management and interpersonal communication dimensions were(78.31±8.15), (23.94±3.06),(23.09±2.77), (15.51±2.13) and (15.78±2.04) , which were all higher than (73.37±9.91), (22.79±3.61), (21.72±3.38), (14.28±2.38), (14.58±2.59) in the control group respectively. The differences were statistically significant(t=-3.611, -2.301, -2.970, -3.661, -3.458; P< 0.05). The final exam score of the experimental group was (84.57±8.21), which was higher than that in the control group (81.40±8.07), with a statistically significant difference (t=2.616, P=0.007).Conclusions Applied course reform of emergency critical nursing care can enhance nursing undergraduates' self-directed learning ability.
8.Analysis of cognitive domains among community-dwelling elders with normal cognitive function

Objective To assess the characteristics of the community-dwelling elderly through a variety of objective evaluation and subjective evaluation tools and provide basis for cognitive function intervention in the community-dwelling elderly. Methods This study evaluated the global cognitive function, memory, executive function and attention and subjective memory feeling of 74 community-dwelling healthy elderly in Beijing. Results The study included 74 community-dwelling healthy elderly. The score of overall cognitive function was (27.28 ± 1.30) points. Less than 80% of participator did not acquire the total score in abstract, language, visual space and executive function. And only 9.8% acquired the total score in the delayed memory .The average score of the memory satisfaction questionnaire was (38.28 ± 9.40) points.In addition, there was no significant correlation between the scores of memory satisfaction of the elderly and the scores of cognitive function scale(r =0.038, P>0.05). The community elderly with normal general cognitive function also showed a relative decline in various cognitive areas, especially in delayed memory. In addition, there was no significant correlation between subjective memory perception and cognitive function scale (r=-0.116-0.157, P>0.05). Conclusions The community-dwelling elderly with normal global cognitive function have cognitive decline in some cognitive function domains. And the cognitive function training needs to be carried out in community. There is no relationship between subjective memory assessment and objective cognitive function. More researches about the relationship between subjective memory assessment and objective cognitive function should be carried out in the future.
9.Risk factors of cerebral microbleeds in patients with cerebrovascular disease

Objective To investigate the related risk factors of cerebral microbleeds (CMBs) and their relations with leukoaraiosis and lacunar infarction in patients with cerebrovascular disease.Methods One hundred and sixty-four patients with cerebrovascular disease,admitted to our hospital from January 2015 and January 2017 were chosen;susceptibility weighted imaging (SWI) was performed,and the distribution and number of CMBs were recorded by Microbleed Anatomical Rating Scale (MARS).The demographic data,personal history,previous medical history,biochemical indices and imageological examination results of the patients were collected and multivariate Logistic regression analysis was carried out on the basis of these data.Spearman dependence analysis was used to analyze the relations of CMBs with leukoaraiosis and lacunar infarction.Results Age (OR=1.071,P=0.024,95% CI:1.009-1.137),hypertension (OR=3.875,P=0.012,95%CI:1.347-11.148),leukoaraiosis(OR=2.080,P=0.005,95% CI:1.245-3.475),lacunar infarction (OR=2.326,P=0.003,95% CI:1.336-4.050) were independently associated with CMBs.And number of CMBs had positive correlation with severity of leukoaraiosis and number of lacunar cerebral infarcts (r=0.564,P=0.000;r=0.762,P=0.000).Conclusion Age,hypertension,leukoaraiosis,and lacunar infarction are the independent risk factors of CMBs;the severity of CMBs is positively correlated with severity of leukoaraiosis and lacunar infarction.
10.Protective effects of Lycium ruthenicum Murr. on hydrogen peroxide-induced cellular oxidative stress injury in heart-derived H9c2 cells

Objective To explore the protective effects of Lycium ruthenicum Murr. (Lrm) on hydrogen peroxide (H2O2)-induced cellular oxidative stress injury in heart-derived H9c2 cells. Methods In this study,H2O2-induced injury in heart-derived H9c2 cells was established as cellular oxidative stress injury model. The H9c2 cells were divided into blank group,model group(H2O2400 μmol/L) and experimental group(Lrm 1.00 g/L +H2O2400 μmol/L). The Lrm group was pretreated with Lrm for 2 h before treated with H2O2for 6 h. Cellular morphology of all groups was observed. Cell viability(MTT assay),apoptosis rate (TUNEL assay),Bcl-2 and Bcl-2 relating x gene protein (Bax) expression were evaluated (Western Blotting). Results Compared with the blank group, abnormal cellular morphology, decreased cell activity,and increased apoptosis were observed in the model group(P<0.05). Anti-apoptotic Bcl-2 gene was less expressed while pro-apoptotic Bax protein level was increased. Compared with the model group, cellular morphology and activity was greatly improved in the experimental group with reduced apoptosis rate (P <0.05). The increased expression of Bcl-2 and reduction of Bax were also significantly different. Conclusion Our findings suggest that Lrm tended to improve the H9c2 myocardial cellular morphology, enhance cell activity and reduce apoptosis rate. Its mechanism of action in reducing oxidative stress injury may be related to the up-regulation of Bcl-2 as well as down-regulation of Bax protein expression.
