1.Research progress in molecular targeted therapy for gynecological cancer

Molecular targeted therapy (MTT) refers to a new therapy designed to interfere with a specific molecular target or signaling pathway that may have a significant influence in tumor growth or progression. MTT can inhibit cell proliferation and induce apoptosis by specific interventions in the signaling pathways of tumor cells. MTT exhibits potential as an effective treatment against cancer. This approach can improve the ability to kill cancer cells and reduce damage to normal tissues. MTT has gradually been used clinically in treating cancer and shown advantages over traditional methods. This paper reviews research progress in this targeted therapy to treat gynecological cancer.
2.Effects of sleep deprivation at different time during night on serum cortisol and emotion of healthy men

Objective To investigate the effects of short-term sleep deprivation(SD)at different time during night on serum cortisol level and emotion state of healthy men,and to find out the influence of work shift onto human circadian rhythm.Methods Two periods of sleep-time were deprived in the present study:the late-night SD(permitted to sleep from 0:00 to 3:00 each day)and the early-night SD(permitted to sleep from 3:00 to 6:00 each day).Ten healthy male adults were chosen to undergo the late-night SD first,followed by normal sleeping for eight nights for washout,and then underwent early-night SD.Each SD period lasted 6 days,i.e.1 day before SD,4 days of SD and 1 day after SD for recovery.Fasting blood samples,for detection of serum cortisol,were collected at 7:00 am every SD day,and the state anxiety inventory(S-AI)scoring was done at the same time.Results The cortisol levels were lowered after 2 types of SD(P0.05).Dependability analysis showed that the cortisol level during the late-night SD period was negatively related to the SD days(r=-0.954 7,P0.05).Both types of SD could elevate anxiety scores,which positively correlated with the SD days(late-night SD:r=0.990,P
3.Barrier function of porcine retinal pigment epithelial cells cultured in vitro

Objective To investigate the blood-retinal barrier(BRB)function of porcine retinal pigment epithelial(RPE)cells cultured in vitro. Methods Primary porcine RPE cells were cultured,and the third generation were inoculated in a microporous filter with the filter membrane of polyvinylpyrrolidone(PVP)-free polycarbonate membrane.After 1,2,3 and 4 weeks of culture,the surface of filter membrane was observed by light microscope,and after 2 weeks of culture,the section of filter membrane was observed by light microscope and transmission electron microscope.Transepithelial electrical resistance(TER)was detected and the permeability was measured with fluorescein sodium(FS)and horseradish peroxidase(HRP). Results Primary porcine RPE cells were cultured successfully.RPE cells converged1week after inoculation; 2 and 3 weeks after inoculation,the density of RPE cells did not changed obviously; 4 weeks after inoculation,the density of RPE cells decreased.The characteristics of polarized growth of monolayer were found in RPE cells on the surface of filter membrane; 2 weeks after inoculation,the TER of RPE cells was(97.44?11.36)?/cm~2 which maintained till the 3rd week after inocubation.After incubated for 30 minutes,only 0.27% of FS and 0.17% HRP reached the inferior filter membrane,and the permeability rate of SF with low molecular weight was higher than which of HRP with high molecular weight. Conclusions The filter with PVP-free polycarbonate membrane may be used to set up the model of RPE cells with polarized growth of monolayer and investigate the barrier function of RPE cells.

5.Clinical analysis of 12 cases of nasal surgery in patients with coronary heart disease after accepted PCI.

OBJECTIVE:
To investigate the perioperative treatments of endoscopic sinus surgery for nasal diseases in patients with coronary heart disease after PCI.
METHOD:
Clinical data of 12 cases of endoscopie-assised surgery such as nasal tumors resection,functional sinus surgery,correction of deviated nasal septum,low-temperature plasma hemostasis in patients with coronary heart disease after PCI were analyzed retrospectively.
RESULT:
Serious bleeding did not take place with the 12 cases during surgery, and surgery progressed smoothly; one of patients had heavy nosebleed after surgery, however her condition was stable when received active treatment. Follow-up 3 months to 2 years, nasal diseases of 12 patients recovered well and symptoms were relieved; cardiovascular events such as hemorrhage, thrombosis and so on did not occur.
CONCLUSION
Due to physiological function of the heart dysfunction in patients with coronary heart disease after PCI,they often accompany a number of trouble issues such as medical disorders, oral antiplatelet drugs, surgery affordability loss and increase surgical risk. Correct and effective perioperative treatments, strictly surgical indications are really necessary which can keep patients safe through perioperative period.

6.Effects of smoking cessation intervention combined with salmeterol and fluticasone propionate powder for inhalation in patients with COPD

Objective To observe the effect of smoking cessation intervention combined with salmeterol and fluticasone pro‐pionate powder for inhalation on chronic obstructive pulmonary disease (COPD) patients with clinical symptoms and pulmonary function .Methods Totally 78 male long‐term smoking cases were randomly divided into control group(n= 40)and observation group(n=38) .The control group was treated with the ophylline sustained‐release tablets ,shah mette lo fluticasone;observation group received smoking cessation intervention .The smoking rate ,symptoms ,acute exacerbation ,life score and lung function (FEV1 ,FEV1/FVC) ,adverse reactions of two groups were observed .Results Two groups of symptoms were all improved ,but the observation group was better (P<0 .05) .FEV1 ,FEV1/FVC ,acute exacerbation ,scores were improved ,but the observation group was better(all P<0 .05);quit rates in the observation group was better than that of the control group(P<0 .05);and did not found adverse reactions .Conclusion The effect of smoking cessation intervention combined with salmeterol and fluticasone propionate powder for inhalation efficacy in the treatment of COPD is distinct ,can improve the pulmonary function and symptoms .

8.Genistein inhibits the time coursed expression changes of ceruloplasmin in rats of focal cerebral ischemia

Aim To define the effect of genistein (GST) on ceruloplasmin (CP) expression in cerebral cortex of focal cerebral ischemia male rats.Methods The effect of GST on CP expression were investigated by means of immunohistochemistry and Western blot in cerebral cortex of focal cerebral ischemia male rats.Results The expression of CP was time coursed during brain ischemia. According to the sham group,CP expression decreased in 6 h-,12 h-and 24 h-ischemia groups,and then increased gradually in 48 h-and 72 h-ischemia rats. GST weakened the ischemia-induced CP change.Pretreatment with an antiestrogen tamoxifen (TAM) could completely abolish the effect of GST.Conclusion The present results indicate that GST may inhibit the time coursed expression changes of CP and then provide neuroprotective effects by inhibiting iron related oxidative stress during brain ischemia via estrogen receptor.
9.Overexpression of trophoblastic stem cell transcription factor,forkhead boxD3,contributes to malignancy of human choriocarcinoma JAR cells

BACKGROUND: Choriocarcinoma is a kind of trophoblastic neoplasm with highly aggressive phenotypes. Forkhead box D3 (FoxD3) is an embryonic and trophoblastic stem cel transcription factor. It plays important roles in different physical and pathological situations such as embryogenesis, carcinogenesis and tumor progression.
OBJECTIVE:To investigate the role of FoxD3 in choriocarcinoma malignancy and the possible mechanism.
METHODS:The human choriocarcinoma JAR cel line was employed in this study. The mRNA and protein expressions of genes were measured by quantitative RT-PCR (qRT-PCR) and western blot, respectively. The FoxD3 specific short hair RNA was applied to down-regulate gene expression. The cel proliferation was evaluatedin vitro by cel counting assay andin vivo by tumor growth. The migration/invasion was determined by transwel assay. The profile of FoxD3 targeted genes was investigated with an Agilent microarray and verified by qRT-PCR.
RESULTS AND CONCLUSION: The FoxD3 mRNA and protein expressions in JAR cells were significantly higher than those in primarily cultured normal trophoblastic cells. Knockdown of FoxD3 by short hair RNA in JAR cells could inhibit cell proliferation and migration/invasionin vitro, and suppress thetumor growth with decreased β-human chorionic gonadotropin secretionin vivo. A profile of seven focal adhesion molecules (ITGA5, ITGB6, THBS4, COL6A3, VTN, NRXN3 and NLGN1) was verified to be targeted by FoxD3. Furthermore, knockdown of FoxD3 by short hair RNA could decrease the activation of focal adhesion kinase. All these findings suggest the overexpression of FoxD3 can contribute to the aggressive phenotype of choriocarcinoma JAR cells by regulating the profile of focal adhesion molecules and focal adhesion kinase.
10.Urinary Nosocomial Infection in Intensive Care Unit:A Clinical Analysis

OBJECTIVE To investigate the characters of urinary noscomial infection in intensive care unit(ICU)of our hospital in order to reduce its incidence. METHODS The 661 patients in ICU of our hospital from Jan 2005 to Jun 2006 were analyzed,and in which 63 nosocomial urinary infection patients(9.53%) were found.We analyzed the 63 patients retrospectively. RESULTS Nosocomial urinary infection occurred in 9.53% patients admitted to the ICU.Nosocomial urinary infection in ICU was correlated with ages,using of antibiotics and hormones,duration of catheterization and APACHEⅡ score,but not with gender.The most prevalent pathogens of nosocomial urinary infection in ICU were fungi(40.66%),gram-negative bacteria(31.87%) and Gram-positive bacteria(27.46%).The clinical manifestation of nosocomial urinary infection in ICU was atypical. CONCLUSIONS Nosocomial urinary infection in ICU has its own characters which clinicians should pay more attention to.To reduce its incidence,we should avoid abusing of antibiotics and hormones,and make the duration of catheterization as short as possible.
