Background Oxidative stress is a pathophysiological process of retina,so it is very important to explore a protective way against retinal oxidative stress.Studies determined that extract of ginkgo biloba (EGb) has antioxidant,anti-apoptosis,anti-thrombosis and anti-inflammatory effects, however, the effect of EGb on human Müller cells in oxidative stress is still below understood. Objective This study was to investigate the protection of EGb against oxidative stress of human retinal Müller cells induced by As2O3 in vitro. Methods Human retinal Müllercell line was cultured in DMEM/F12 with 10% fetal bovine serum (FBS),2 μmol/L glutamine and 1% antibiotics.As2O3 solution at the final concentration 5 mg/L was added in the medium for 24 hours to establish oxidative models, and then the EGb with the final concentrations of 5,10 and 20 mg/L was used to cell models for 24 hours, respectively.Cell viability was detected by MTT assay, and reactive oxygen species (ROS) levels in the cells were detected with CM-H2DCFDA fluorescent probe. The relative expression levels of caspase-3 mRNA in cytoplasm and cell nuclei were assayed by reverse-transcription PCR (RT-PCR),and the expressions of Nrf2 protein were quantitatively detected by Western blot. Results Müller cells adhered well 24 hours after cultured. At 6-7 days after culture,Müller cell body was large with abundant cytoplasm and mosaic-like arrangement. However, floating cells were seen after As2 O3 treatment.Cell viability (absorbance) was significantly different among the normal culture group,As2 O3-treated group,As2 O3 + 5 mg/L EGb group,As2 O3 + 10 mg/L EGb group and As2 O3 + 20 mg/L EGb group, with the strongest viability in the normal culture group and the weakest viability in the As2 O3-treated groups (F=163.57,P =0.00).The fluorescence intensity of ROS was the weakest in the normal culture group and the strongest in the As2 O3-treated group and was gradually weakened with the increase of EGb doses, showing a remarkable difference among the groups (F =4 013.61,P =0.00).The relative expression level of caspase-3 mRNA in the cells was gradually reduced with the increase of EGb doses, with a statistically significant difference among the groups (F =2 199.72,P =0.00).In addition,no considerable difference was seen in the expression level of Nrf2 protein (grey scale) in cytoplasm among the groups (F=15.42,P=0.40);while in the nuclei, the expression levels of Nrf2 protein were 100.01 ±0.04,46.59±0.63,54.51 ±0.62,59.93 ±0.17 and 67.60±0.24 in the normal culture group,As2 O3-treated group and As2O3+5 mg/L EGb group,As2O3+10 mg/L EGb group,As2O3+20 mg/L EGb group respectively, with a significant difference among them (F=7 271.72,P=0.00).Conclusions EGb can protect human retinal Müller cells against As2O3-induced damage in a dose-dependant manner by antioxidant and antiapoptotic effects in vitro, and the activities occur primarily in cell nucleus.

Objective To investigate the effect of probiotics preconditioning on intestinal mucosal barrier function in a rat model of cardiopulmonary bypass (CPB) . Methods Twenty-four adult male SD rats weighing 350-450 g were randomly divided into 3 groups ( n = 8 each) : group Ⅰ sham operation ( group S); group Ⅱ CPB and group Ⅲ probiotics + CPB. CPB was established between left carotid artery and right jugular vein and maintained for 60 min. Croup Ⅲ received intragastric instillation of probiotics 2 ml once a day for 7 consecutive days before CPB. Blood samples were collected at 2 h after CPB was terminated for determination of plasma D-lactate, TNF-and IL-6 concentrations and diamine oxidase (DAO) activity. Venous blood was obtained from portal vein for determination of LPS concentration and cultured. Liver, pancreas, spleen, kidney and mesenteric lymphnode (MLN) specimens were obtained under sterile condition and cultured for bacterial growth. The mucous membrane of small intestine was examined with electron microscope. Results CPB significantly increased plasma DAO activity, and D-lactate, IPS, TNF-α and IL-6 concentrations and the rate of bacteria-positive culture of portal venous blood, liver, MLN, lung and kidney in group Ⅱ as compared with group S. Probiotics preconditioning significantly attenuated the above-mentioned CPB-induced changes. Microscopic examination showed that probiotics preconditioning significantly ameliorated CPB-induced damage to the epithelium of the small intestine. Conclusion Probiotics preconditioning can significantly attenuate CPB-induced inflammatory response and protect the intestinal mucosal barrier function.

Objective To Study the effect of efflux pump-inhibitors(EPI)-Verapamil in photodynamic therapy (PDT) using hematoporphyrin monomethylether (HMME) as photosensitizer on the cariogenic bacteria in dental plaque biofilms. Methods According to the administrator order of the verapamil and photosensitizer in PDT,streptococcus mutans, streptococcus sanguis, eosinophilic lactobacillus and actinomyces viscosus were used to establish the dental plaque biofilm model. The experiment was divided into five groups, group A was incubated with the photosensitizer and verapamil group, group B using verapamil before incubated the photosensitize, group C suing photosensitizer before incubated verapamil, group D with PDT only, group E was control group. After laser treatment, the influence of the dental plaque biofilms was observed by confocal laser scanning microscope. Results As saline-treated group is a group of normal the dental plaque biofilms. In PDT only group, compared with the saline group, red fluorescence increased significantly, the bacteria lose accumulation capacity, and were isolated and scattered in dispersed state. In PDT plus verapamil group, compared with only PDT group, green staining increased, bacterial activity increased. In group B,cells were incubated with verapamil before incubated the photosensitizer group, green staining increased significantly, red fluorescence reduced, indicating live bacteria increased, and bacteria activity was significantly increased. Conclusion PDT is an effective method in eliminating cariogenic bacteria of dental plaque biofilms.Bacterial efflux pump inhibitors can lower HMME-PDT inhibition cariogenic bacteria in dental plaque biofilm, and pre-verapamil administration could significantly inhibit the effect of PDT treatment of dental caries.

Abstract: Neurotensin receptor-1 (NTR1), which can stimulate the intracellular cascade signal pathway, belongs to the large superfamily of G-protein coupled receptors. NTR1 is related to the occurrence and development of several kinds of diseases. In order to screen the inhibitors for the cancers associated with NTR1 protein, we established a CHO (Chinese hamster ovary) cell line in which human neurotensin receptor-1 was highly expressed. The method is to construct the recombinant plasmid which was lysed with the hNTR1 gene and transfect it into CHO cells. After selected with G418, the cell line was evaluated by Western blotting analysis and calcium flux assays. Through the calcium flux assays on FlexStation 3, we got the EC50 value of neurotensin peptide which is the natural NTR1 agonist, and the IC 50 value of SR48692 which is the known NTR1 antagonist. The established human CHO/NTR1 cell line can be used to study the profile of NTR1 biological activity and further screen of NTR1 antagonists and agonists.
