1.Revision of Research Capacity Self-rating Scales of Nursing Staff and its reliability and validity

Objective To revise the Research Capacity Self-Rating Scales of Nursing Staff and test its reliability and validity.Methods Based on the definition of scientific research capacity,the initial dimensions and items of Research Capacity Self-Rating Scales of Nursing Staff was formed by literature review,semi-structured interview and experts evaluation.The content validity was evaluated by experts,and the face validity by nursing staffs.600 nurses were investigated by convenient sampling.The reliability and validity and discrimination of the scale were tested.Results Research Capacity Self-Rating Scales of Nursing Staff were selected in 7 dimensions including the capacity of find problem,research design,scientific research practice,innovation,information retrieval,data processing and writing.Scale-level Content validity index was 0.94.The Cronbach alpha coefficient of Research Capacity Self-rating Scales of Nursing Staff was 0.964,the Cronbach alpha coefficients of the seven dimensions were 0.771-0.975,split-half reliability was 0.885.Seven factors were extracted by using principal factor analysis,cumulative contribution was 71.002%.The correlation coefficient between the Scientific Research Capacity Self-rating Scales of Nursing Staff and General Self-Efficacy Scale was 0.649 (P=0.000).The discrimination was better.Conclusions The Research Capacity Self-rating Scales of Nursing Staff revised shows good reliability and validity.It can objectively evaluate scientific research capacity of nursing staff and provide scientific reference for further improving the research capacity of the nursing staff.
2.Effect of Qige San Ethyl Acetate Extract on Human Esophageal Carcinoma Eca109 Cells Apoptosis by Inhibiting Signal Transducers and Activators of Transcription 3 Signaling Pathway

Objective To study the effect of Qige San ethyl acetate extract on the proliferation of human esophageal carcinoma Eca109 cells and to explore its possible mechanism. Methods Inhibitory effects of Qige San ethyl acetate extract on the proliferation of esophageal carcinoma Eca109 cells were detected by MTT assay, cell apoptosis was detected by flow cytometer, and the expression of protein STAT3, Bcl-2 and Caspase 9 was detected by Western blotting method. Results In the range of 10~100 μg/mL, Qige San ethyl acetate extract inhibited the proliferation of Eca109 cells effectively (P<0.05) . Compared with the control group, Qige San ethyl acetate extract in the concentrations of 1.47, 33.26, 75.52 μg/mL significantly increased the apoptotic rate of Eca109 cells within 48h ( P<0.01). And Western blotting results showed that the ex pression levels of STAT3 and Bcl-2 were reduced, and Caspase 9 was increased with the increase of drug concentration. Conclusion Qige San ethyl acetate extract could significantly inhibit the proliferation of Eca109 cells and induce cell apoptosis, and its mechanism is probably associated with the inhibition of signal transducers and activators of transcription 3 (STAT3) signaling pathway.
3.CT and MRI features of pancreatic serous micro-cystic cystadenoma

Objective To analyze the CT and MRI features of pancreatic serous micro-cystic cystadenoma (SMCA).Methods CT and MRI data of 17 patients with SMCA confirmed by pathology were analyzed retrospectively.Tumor's location,diameter,septation,contrast-enhancement features,calcification,central scar, and the extent of pancreaticobiliary duct dilatation were recorded.Results Seventeen patients were enrolled.10 underwent CT,6 underwent MR(2 patients also underwent MRCP), and 1 underwent both CT and MR.Tumors were located at pancreatic head in 2 cases,neck in 2 cases,and body/tail in 13 cases.Lesion size ranged from 1.7 cm to 14 cm with an average of 5.2 cm.Septation was seen in 16 cases.Central scar was seen in 9 cases Septation and scar were enhanced and cystic component was not enhanced on contrast enhancement.6 patients presented the main pancreaticduct dilation.Conclusion Pancreas SMCA has certain imaging features.It is always manifested as lobulated mass with multiple small capsules(<2 cm) and fiber separations.The fiber separation enhance significantly,but lower than normal pancreatic tissue.Central scar is the characteristic manifestation of SMCA, and it manifests iso or slightly lower signal on T1WI and T2WI.Typical SMCA can be diagnosed according to the CT and MR imaging manifestations.
4.Abnormal status of tumor markers detected among healthy physical examinees: clinical significance of early tumor screening at molecular level

Purpose The aim of this study is to define molecular sub-healthy status among individuals attending routine physical examination.Methods This study examined 8 blood tumor markers among 5 825 healthy individuals.Results (1) Differences in levels of tumor markers were observed between males and females.(2) Elevated CA72-4 levels were seen in 16.05% of individuals in both sexes.(3) More males had elevated levels of CEA and CYFRA21-1.Among the individuals with elevated levels of tumor markers,most were clustered in the prime age group of 41 to 50 years.Conclusion This is the first large-sample survey in healthy Han population in Xinjiang,which found elevated levels of all 8 markers tested with varying rates.Accordingly,this study proposes for the first time a concept of molecular sub-healthy status and a strategy of early tumor screening at molecular level.The molecular sub-healthy status may be an early warning sign for potential cancers,which should draw attention by preventive public health policies.
5.Resting state functional magnetic resonance imaging changes in spasmodic torticollis patients based on fractional amplitude of low frequency fluctuation

Objective To observe the differences of cerebral activation pattern with resting state functional magnetic resonance imaging (rs-fMRI) between patients with spasmodic torticollis (ST) and healthy controls,thus to investigate the pathogenesis of ST.Methods Nineteen ST patients and 21 age,sex and education-matched healthy controls,recruited from the Department of Neurology,First Affiliated Hospital of Guangxi Medical University between November 2012 and January 2016,were included in this study.rs-fMRI and factional amplitude of low frequency fluctuation (fALFF) were used to obtain differences between patients with ST and healthy controls,and correlative analysis was made on fALFF values of abnormal brain regions and ST patients' symptom severity (Tsui scores).Results Compared with healthy controls,patients with ST had significantly increased fALFF in the left cerebellum and significantly decreased fALFF in the left posterior cingulate cortex/precuneus,right posterior cingulate cortex/precuneus,left middle temporal gyrus,right angular gyrus,left post-central gyrus,right supplementary motor area (t =-5.714-5.920,P <0.01),and abnormal brain regions' fALFF values had no correlation with patients' age of onset,disease course,symptom severity (P > 0.05).Conclusion Abnormal sensorimotor area,default mode network and cerebellum dysfunction may play a role in the pathophysiology of ST.
6.Clinical characteristics of hospital-acquired venous thromboembolism: a single-center cross-sectional study

Objective:To observe the characteristics and outcomes of hospital-acquired venous thromboembolism (HA-VTE), and to guide the clinic to take preventive measures for high-risk patients to reduce the incidence of HA-VTE.Methods:The clinical data of 1 570 hospitalized patients with HA-VTE from December 2013 to December 2019 in Xuanwu Hospital, Capital Medical University were retrospectively analyzed, including the basic information, department, risk factor evaluation of venous thromboembolism (VTE), prevention and outcomes, etc.Results:During the period, the total incidence of HA-VTE was 0.50% (1 570/317 047). The annual incidence of HA-VTE increased year by year, from 0.19% (85/44 737) in 2014 to 0.82% (564/68 780) in 2019. The incidence of HA-VTE in elderly patients (age ≥65 years old) was significantly higher than that in young and middle-aged patients (age form 18 to 64 years old): 0.96% (970/100 768) vs. 0.28% (600/216 279), and there was statistical difference ( χ2 = 654.96, P<0.01). There was no statistical difference in the incidence of HA-VTE between male and female: 0.51% (780/151 617) vs. 0.48% (790/165 430), χ2 = 2.19, P>0.05. HA-VTE patients were mainly distributed in the neurology department, emergency department, neurosurgery department, orthopedics department, vascular surgery department, general surgery department, etc, with the highest proportion of 27.83% (437/1 570) in neurology department. The departments with high incidence of HA-VTE were intensive care unit, emergency department, stroke center, orthopedics department, rehabilitation department and neurology department, with the highest incidence of 7.69% (7/91) in intensive care unit. The hospital stay in patients with HA-VTE was significantly longer than that in patients without HA-VTE: 14 (9, 20) d vs. 7 (3, 11) d, and there was statistical difference ( Z = - 39.75, P<0.01). During hospitalization, 94 patients died, and 7 cases (0.45%, 7/1 570) were directly caused by HA-VTE. Only 0.13% (2/1 570) of the patients underwent the risk factor evaluation of VTE. Conclusions:The annual incidence of HA-VTE has a clear upward trend, and the incidence of critical illness and elderly patients is the highest. HA-VTE significantly prolonged the average hospital stay of patients and increased the risk of death. Screening and evaluation should be strengthened, high-risk groups should be identified, and active preventive measures should be taken to reduce the risk of HA-VTE.
7.A preliminary study on the MATP gene knockout in a mouse melanoma cell line using CRISPR-Cas9 system

Objective To knockout the MATP gene of mouse melanoma cell line B16F10 using CRISPR/Cas9 system,and to lay foundation for the functional study of MATP gene.Methods Specific primers of MATP were designed according to the report in http://crispr.mit.edu/ website.The primers were linked to pCAS9/gRNA1 vector.Then the positive vector was transfected into mouse melanoma B16F10 cells,and monoclonal cell lines were obtained by the infinite dilution method.After the genomes of different monoclonal cell lines were extracted and sequenced,the cell lines with MATP gene cleavage were screened,and the expression of MATP in these cell lines was verified by Western-blot analysis.Results Three MATP gene knockout cell lines were successfully obtained.The western-blot results showed that the cell lines did not express MATP protein.Conclusions The knockout of MATP gene in B16F10 cell line can be successfully achieved using the pCAS9/gRNA1 vector.
8.Interleukin-18 and post-stroke depression

Post-stroke depression (PSD) can seriously affect the outcome and quality of life of patients with stroke. However, due to the lower diagnostic rate, a large number of patients with PSD have not been treated in time and have a poor outcome. Therefore, only early detection, early diagnosis and early treatment of PSD can improve the prognosis and quality of life of patients with stroke. In recent years, a large number of studies on serum biomarkers have tried to find objective indicators related to PSD. Among them, interleukin-18 is associated with the pathogenesis of stroke and PSD, which may be attributed to its polymorphism and constitutive expression in the brain, especially the role of serum level changes in the risk stratification and outcome of acute ischemic stroke. This article reviews the role of interleukin-18 in the pathogenesis of PSD.
9.Relationship between PDE4C gene methylation in peripheral blood leukocytes, interaction of environmental factors and pathogenesis of breast cancer

Objective The objective of this study was to investigate the relationship between the methylation of phosphodies-terase 4C(PDE4C)gene in peripheral blood leukocytes,and its interaction with environmental factors and the pathogenesis of breast cancer. Methods A case-control study was conducted to select 402 cases of breast cancer and 470 cancer-free controls from Octo-ber 2010 to December 2014. The specific DNA methylation-modified quantitative PCR method was used to detect the methylation of PDE4C gene. The use of crossover analysis and multivariate logistic regression was used to analyze the relationship between the level of gene methylation and its interaction with environmental factors and the pathogenesis of breast cancer. Results PDE4C methylation was not found to be significantly associated with the onset of breast cancer. High-frequency/high-intake of whole grains,vegetables, onion-based plants,poultry,milk,and regular exercise could reduce the risk of breast cancer;high-frequency/high-intake of pork, irregular menstrual cycle,and a high psychological stress index could increase the risk of breast cancer. PDE4C hypermethylation had a significant combining effect with edible onion-producing plants greater than 3 times/week,with an OR of 0. 373(95% CI:0. 208~0. 668,P=0. 001). Conclusion Hypermethylation of PDE4C is not an independent risk factor for breast cancer,and its combination with environmental factors affects the risk of breast cancer.
10.A consensus on prenatal diagnosis and genetic counseling for chromosomal mosaicism.

With the extensive application of highly sensitive genetic techniques in the field of prenatal diagnosis, prenatal chromosomal mosaicisms including true fetal mosaicisms and confined placental mosaicisms are frequently identified in clinical settings, and the diagnostic criteria and principle of genetic counseling and clinical management for such cases may vary significantly among healthcare centers across the country. This not only has brought challenges to laboratory technician, genetic counselor and fetal medicine doctor, but can also cause confusion and anxiety of the pregnant woman and their family members. In this regard, we have formulated a consensus over the prenatal diagnosis and genetic counseling for chromosomal mosaicisms with the aim to promote more accurate and rational evaluation for fetal chromosomal mosaicisms in prenatal clinics.
