1.A study on Huoxue Tongmai capsules for regulation of small molecule metabolites in plasma of patients with coronary heart disease and blood stasis syndrome

Objective To observe traditional Chinese medicine (TCM) Huoxue Tongmai capsules (HXTMC) for the regulation of small molecule metabolites in plasma of patients with coronary heart disease and TCM blood stasis syndrome. Methods A prospective study was conducted in which 136 patients with coronary heart disease and blood stasis syndrome treated in Dagang district in Tianjin from January 2009 to December 2013 were selected and randomly divided into two groups:control group (68 cases) and HXTMC group (68 cases). The control group received the conventional oral therapy with nitrates, β receptor blockers, calcium antagonists, enteric coated aspirin, etc, and the HXTMC group received the conventional therapy and additionally oral administration of HXTMC, 4 capsules each time (each capsule 0.25 g), trice a day, 30 days constituting a therapeutic course, 3 courses in total. According to the standard of curative effect evaluation of angina pectoris in coronary heart disease, the clinic total curative effects of angina pectoris and electrocardiogram (ECG) were evaluated. The concentrations of copeptin, tumor necrosis factor-α(TNF-α) and interleukin-10 (IL-10) in the plasma were tested by enzyme-linked immunosorbent assay (ELISA), and the TNF-α/IL-10 ratio was calculated. Results The total clinical effective rates of patients and the ECG in HXTMC group were significantly higher than those of the control group [clinic total effective rates:89.71%(61/68) vs. 80.88%(55/68), ECG total effective rates:57.35%(39/68) vs. 48.53%(33/68), both P<0.05]. Concentration of copeptin and TNF-α/IL-10 ratio in the plasma after treatment of the both groups were significantly lower than those before treatment, and the changes were more obvious in HXTMC group [copeptin (ng/L): 0.22±0.04 vs. 0.31±0.05, TNF-α/IL-10 ratio:0.49±0.11 vs. 0.65±0.09, both P<0.05]. Conclusion Addition of HXTMC to conventional therapy for treatment of patients with coronary heart disease accompanied by blood stasis syndrome can lower the plasma level of copeptin and TNF-α/IL-10 ratio, thus it may improve the clinical curative effects of the patients.
2.Study on the Genotoxicity of Bromate

Objective To study the genotoxicity of bromate,a chief by-product of ozonated drinking water.Methods Genotoxicity at gene,DNA and chromosome levels induced by bromate DBP was determined using Ames test,unscheduled DNA synthesis(UDS)assay and micronucleus test(MNT)of mice bone marrow polychromatic erythrocytes respectively.Results In Ames test,bromate showed no mutagenic effect on salmonella typhimurium TA98and TA100strains with or without S9(rat liv-er metabolic activation system)compared with the negative control group,but the results of UDS assay showed that bromate could induce unscheduled DNA synthesis in rat primary hepatocytes and the results of MNT indicated that bromate increased the micronucleus rate of bone marrow polychromatic erythrocytes of mice.Conclusion It was suggested that bromate DBP might have genotoxicity at DNA and chromosome levels.
3.The correlations between the different pathological types gastritis and the expressions of AQP3 and AQP4

Objective To explore the correlations of expressions of gastric mucosa water channel aquaporin AQP3 and AQP4 and different gastritis types. Methods The gastric mucosa was mounted under gastroscope. The types of gastric mucosa pathology and activity were tested by the common pathohistology. The expressions of AQP3 and AQP4 were determined by immunohistology. Results The expressions of AQP3 and AQP4 of chronic superficial gastritis were significantly higher than those in the non-gastritis group and chronic atrophic gastritis group (P < 0.01), especially in the activity period of chronic superficial gastritis. The expressions of AQP3 and AQP4 of chronic atrophic gastritis group were reduced when compared to those in the non-gastritis group, in spite of no statistical differences between them. While compared to the non-gastritis group , the expression of AQP3 of chronic atrophic gastritis group during the active stage was remarkably decreased (P < 0.05). Conclusion The expressions of AQP3 and AQP4 of gastric mucosa in chronic gastritis in various pathological types are different. AQP3 and AQP4 may be the targeted point , which could be used for the differential diagnosis and treatment of chronic gastritis of different pathological types.
4.Hsa-miR-218 inhibits growth of cervical cancer HeLa cells by targeting to LASP1

AIM:To study the effect of hsa-miR-218 on cervical cancer HeLa cell growth and the underlying molecular mechanism .METHODS:The lentivirus expression vector pmiR-218 targeting to hsa-miR-218 was constructed . pmiR-218 was transfected into HeLa cells .The number of viable HeLa cells was counted by the method of Trypan blue ex-clusion.The inhibitory rate of cell activity was detected by WST-8 assay.The expression of LIM and SH3 protein 1 (LASP1) at mRNA and protein levels was determined by real-time PCR and Western blot.The interaction between miR-218 and LASP1 was examined using a luciferase reporter assay .RESULTS:The lentivirus expression vector pmiR-218 tar-geting to hsa-miR-218 was constructed successfully and confirmed by DNA sequencing .Over-expression of miR-218 inhibi-ted the activity of HeLa cells with the inhibitory rates of 15%, 26%and 65%at 24 h, 48 h and 72 h, respectively .The difference between transfection group and blank control /negative control group was statistically significant .The luciferase activity was reduced when co-transfection with miR-218 mimics and LASP1-3’ UTR plasmid.The relative expression of miR-218 was increased after transfection with pmiR-218.Over-expression of miR-218 down-regulated the LASP1 expression at mRNA and protein levels by 25%and 75%respectively.Compared with blank control group and negative control group , the difference was statistically significant (P<0.05).CONCLUSION:pmiR-218 effectively inhibits the growth of HeLa cells in a time-dependent manner.miR-218 targets to the 3’UTR of LASP1, thus down-regulating the expression of LASP1 in HeLa cells .
5.Detection of Neuron-specific Enolase and Soluble Intercellar Adhesion Molecule-1 in Serum and Cerebrospinal Fluid in the Patients with Viral Encephalitis

Objective To investigate the changes of the neuron-specific enolase(NSE) and soluble intercellar adhesion molecule-1(CAM-1) in serum and cerebrospinal fluid(CSF) in the patients with viral encephhalites (VE). Methods The levels of NSE and sICAM-1 in serum and CSF were determined before and after treatment using ELISA in 58 patients with VE and 20 normal persons. Results The levels of NSE and sICAM-1 in serum before treatment were obviously higher than those of control group, and their differences were significant (P0.05). Conclusions NSE and sICAM-1 may contribute to pathologic course in infection of VE and the levels of NSE and sICAM-1 in serum and CSF may serve as markers of differential diagnosis and clinical significance.
6.Investigating predictors of the 641 elders' willingness to enroll in nursing homes in Shanghai

Objective · To survey the willingness to enroll in nursing homes among the elders in Shanghai and explore its predictors. Methods · A field survey was performed to collect data from 641 elders aged 60 years old and above in six community health centers in Shanghai. Sociodemographic variables, willingness to use nursing home, perceived barriers and perceived benefits were measured. Results · The questionnaire is with good structural validity and good intrinsic reliability. KMO index, P value of Bartlett's test of sphericity, and the range of Cronbach's α coefficients were 0.862, 0.000,and 0.739-0.824, respectively. Among the respondents, 46% were willing to enroll in a nursing home in this survey. Age had significant influence on their willingness (P=0.045). After adjusting for age, the multivariate Logistic regression analysis showed that shame (OR=0.76, 95% CI=0.62-0.94), adaptability (OR=0.61, 95% CI=0.50-0.74) and perceived benefits (OR=1.63, 95% CI=1.32-2.03) were related to willingness (P<0.05).Conclusion · Elders in Shanghai have a relatively high level of willingness to enroll in nursing homes, especially the elders aged from 60 to 69. Shame and adaptability of elders, and the services provided by nursing homes were the predictors of the elders' willingness.
7.Follow-up study of risk factors of white matter hyperintensity and cognitive impairment in elderly patients with type 2 diabetes mellitus

Objective To follow up the progression of white matter hyperintensity(WMH)and cognitive impairment in elderly patients with type 2 diabetes mellitus,and to investigate the risk factors affecting the progression of white matter hyperintensity(WMH)in elderly patients with type 2 diabetes mellitus.Methods Seventy-six elderly patients with type 2 diabetes were followed up for 2 years.General clinical data were collected at baseline and 2 years after follow-up,brain MRI was improved,WMH severity was assessed by Fazekas scale.The Hamilton anxiety scale(HAMA)and the Hamilton depression scale(HAMD)assessed the mental state of the subjects.A series of cognitive function tests were performed.The cognitive function assessment tools used included MMSE,number span test(attention),word immediate and delayed memory(memory),Stroop color word interference test(executive function inhibitory control),verbal fluency test(cognitive flexibility),and clock drawing test(visuospatial structure ability).Results At baseline and after 2 years of follow-up,subcortical WMH(SC-WMH)[1.5(1,2)vs.2(1,3)]and total WMH score[3(2,4)vs.3(2,4)],number span inversion(6.32±1.59 vs.5.37±1.43),total number span score(14.71±2.61 vs.13.71±2.32),word delay memory(2.86±0.86 vs.2.14±0.99),Stroop color word interference test color response time[0.58(0.43,0.72)vs.0.61(0.53,0.79)],word response time[0.47(0.37,0.55)vs.0.51(0.42,0.65)],color word response time[0.98(0.95,1.05)vs.1.01(0.97,1.13)],clock test(8.09±1.48 vs.7.76±1.74)and verbal fluency test(4.42±0.82 vs.4.29±0.75)were compared,and the difference was significant(P<0.05).Age(OR=1.383,95%CI:1.094~1.748)and creatinine(OR=1.100,95%CI:1.004~1.205)were significantly associated with WMH progression(P<0.05).Conclusions After 2 years of follow-up,WMH and SC-WMH in the elderly patients with type 2 diabetes have exhibited certain progression.Attention,delayed memory,executive function,visuospatial structure,and cognitive flexibility are all impaired to varying degrees.Patients'age and creatinine are significantly correlated with the progression of WMH.
