1.Effect of metabolic syndrome on patients with impaired glucose regulation in regard to cardiovascular and cerebrovascular diseases

The effect of metabolic syndrome on patients with pre-diabetes in regard to cardiovascular and cerebrovascular diseases (CCVD) was investigated.The subjets with pre-diabetes were divided into metabolic syndrome (MS) group and non-MS.Follow-up after the baseline survey,the new-onset cardiovascular events of patients were collected.The incidence of CCVD in the MS group was 5.66%,being higher than 1.88% in non-MS group (P<0.05).After factors being corrected,compared with the non-MS group,the relative risk (RR) of CCVD in MS group was 2.616 (95% CI 1.328-5.153).Compared to only 1 component of MS patients,the risks of CCVD in MS component 2,≥3 patients were increased by 29% (HR =1.29) and 62% (HR =1.62).Metabolic syndrome may increase the risk of cardiovascular and cerebrovascular diseases in pre-diabetes populations.The occurrence of CCVD and MS content showed a dose-dependent relationship.
2.Xenotransplantation of agarose-microcapsulated porcine islets for the treatment of diabetes mellitus

BACKGROUND: Previous studies have shown that agarose-microcapsulated adult porcine islets had some effective biology activities in vitro. However, the biologic effects of such microcapsule materials in transplanted animal need to be further evaluated. OBJECTIVE: To observe the effect of intraperitoneal transplantation of agarose-microcapsulated adult porcine islets in experimental diabetic rats. DESIGN, TIME AND SETTING:A controlled observational experiment was performed at the Central Laboratory of Zhengzhou University from December 2007 to December 2008. MATERIALS: Agarose was used to make microencapsules which were used to coat adult porcine islets by phase-separation method. METHODS: The islets with or without agarose were cultured in RPMI1640, and the culture liquid was changed every two days. The supernatant was collected on the first and fifth days to determine insulin content. The islets placed in different solutions, including 5.6 mmol/L glucose, 16.6 mmol/L glucose, and 10.0 mmol/L alkali, were used for insulin stimulated test by radioimmunoassay. A total of 27 diabetic rats were randomly divided into three groups: control group, unencapsulated islets transplantation group and encapsulated islets transplantation group, injecting saline, (0.9-1.6)?103 unencapsulated islets, and (0.8-1.7)?103 encapsulated islets, respectively. MAIN OUTCOME MEASURES: the difference of basic insulin of microencapsulated islets with agarose; the activity of microencapsulated islets responded to glucose and alkali; the subsistence time of rats accepted microencapsulated islets transplantation. RESULTS: The islets could secrete insulin consecutively for a month, and the cells in capsules survived very well without autolysis and breakage. At day 2 after culture, the islets had marked reactions to the stimulation from glucose in high concentration and theophylline. 4.Association of serum visceral adipose tissue-derived serine protease inhibitor levels with carotid atherosclerosis in elderly patients with type 2 diabetes

The serum levels of visceral adipose tissue-derived serine protease inhibitor (vaspin)were measured in 65 elderly type 2 diabetic patients with carotid atherosclerosis( T2DM + CA group), 63 elderly type 2 diabetic patients without carotid atherosclerosis (T2DM group ) and 55 normal elderly individuals( NC group)by enzyme-linked immunosorbent assay. The results showed that the serum vaspin levels in T2DM + CA group were significantly lower than those in T2DM group[(0. 23 ± 0. 13 ) μg/L vs.(0. 41±0. 16)μg/L, P <0. 01]. The serum vaspin levels in T2DM + CA group and T2DM group were significantly lower than those in NC group respectively[(0. 63 ± 0. 11 ) μg/L, P < 0. 01 and P < 0. 05].Logistic regression analysis showed that high sensitivety C-reactive protein and Homeostasis Model Assessment insulin resistance index (HOMA-IR) were negatively correlated with serum vaspin levels,which indicates that lower serum vaspin levels might be associated with the pathogenesis of carotid atherosclerosis in elderly type 2 diabetic patients.
5.The Influence of Surface Roughness of Tooth Preparation and Cement Compatibility on the Adhesive Strength

Objective To investigate the influence of tooth preparation surface roughness and different dental ce-ments on adhesive strength, and provide some reference information on the selection of dental rotary instruments and dental cements. Methods Sixty dentin samples were prepared and randomly divided into 3 groups. Samples from group A, B and C were grinded by diamond burs with grit-sizes of 125μm, 60μm and 28μm respectively. Two samples selected randomly from each group were observed under scanning electron microscope (SEM) for the surface microprofile. The remaining 18 samples from each group were evenly divided into 3 subgroups. Each subgroup was teamed with one of 3 dental cements:zinc polycarboxylate cement (ZP), glass ionomer cement (GI) and resin-modified glass ionomer cement (RMGI). Co-Cr alloy casts were cemented onto the dentin samples, and the adhesive strength was tested. Meanwhile, the types of failure were re-corded for each sample. Results The surface roughness of tooth preparation samples showed a downward trend in group A, group B and group C. Among the involved 3 dental cements, RMGI exhibited the highest adhesive strength, and there was no significant difference in the adhesive strength between ZP and GI. Among different combination subgroups, A-RMGI had the highest adhesive strength, B-RMGI run the second place, while C-ZP and C-GI were proved the lowest. However, there was no interaction between the two factors. Furthermore, group A mainly showed failure typesⅠandⅡ, group B evenly showed all the failure types and group C mainly showed failure typesⅡandⅢ. Conclusion The adhesive strength is related to the surface roughness. RMGI exhibits the highest adhesive efficiency. There is no obvious compatibility in particular surface roughness and dental cements.
6.Clinical Observation of Zhenqi Fuzheng Granules in the Adjunctive Treatment of Cervical Cancer

OBJECTIVE:To observe therapeutic efficacy and safety of Zhenqi fuzheng granules in the adjunctive treatment of cervical cancer. METHODS:86 patients with cervical cancer were randomly divided into control group(43 cases)and observation group (43 cases). Control group was given intravenous dripping of Diphenhydramine hydrochloride injection 20 mg,Granisetron hydrochloride and glucose injection 5 mg,Cimetidine injection 20 mg at 0.5 h before chemotherapy. After 0.5 h of intravenous drip-ping,the patients received chemotherapy:Docetaxel injection 40 mg/m2,d1,d8,intravenous dripping;Cisplatin for injection 25 mg/m2,d1-3,intravenous dripping;21 d as a cycle,a total of 4-6 cycles. On the basis of control group,observation group was addi-tionally given Zhenqi fuzheng granules 5 g,twice 2 day. Clinical efficacies of 2 groups were observed as well as T cell subsets in-dexes [CD3+,CD4+,CD8+,natural killer cell(NK)],KPS scores,termination,terminal time,the times of chemotherapy and the oc-currence of toxic reaction before and after treatment. RESULTS:The total remission rate of observation group was significantly higher than that of control group;the proportion of termination of chemotherapy in observation group was significantly lower than control group;terminal time of chemotherapy was significantly shorter than control group;the times of chemotherapy was signifi-cantly more than control group;the incidence of gastrointestinal reaction,anemia,thrombocytopenia and leukopenia were signifi-cantly lower than control group,with statistical significance(P<0.05). There was no statistical significance in liver function dam-age,renal function injury and stomatitis between 2 groups (P>0.05). Before treatment,there was no statistical significance in T cell subsets indexes and KPS scores between 2 groups(P>0.05). After treatment,CD3+,CD4+ and NK cells in observation group were significantly higher than before treatment and control group,while CD8+ was significantly lower than before treatment and control group;KPS scores of 2 groups were significantly higher than before,and the observation group was significantly higher than the control group,with statistical significance(P<0.05). There was no statistical significance in T cell subsets indexes of con-trol group before and after treatment(P>0.05). CONCLUSIONS:On the basis of routine treatment,Zhenqi fuzheng granules can improve therapeutic efficacy,immune function and the quality of life,moreover,reduce toxic reaction in patients with cervical can-cer.
7.Quality Standard Study on Danqi Tablets

[Objective] To set up standard method of quality to Danqi Tablets.[Method] Take HPLC to make nature authentication,take reverse HPLC to test the content of salvianolc acid B in the preparation.[Result] The authentication is strong in attribute and has good repetition,the measurement has good linear relation,the recycle rate is 101.7%,RSD 1.77%.[Conclusion] The quality standard can effectively control the quality of Danqi Tablets.
8.Correlation study of the expression of alpha-fetoprotein mRNA in the peripheral blood and postoperative survival and metastasis of patients with liver cancer

[Objective]To explore the correlation of the expression of alpha-fetoprotein(AFP)mRNA in the peripheral blood and postoperative survival and metastasis of patients with liver cancer.[Methods] A total of 66 patients with liver cancer who received radical resection surgery from January 2005to December 2006 was enrolled in this study.The cell total RNA was extracted from peripheral blood and the expression of AFP mRNA was detected by nested PCR.All the patients were followed up for 60 months after surgery.[Results]The expression rate of AFP mRNA in the peripheral blood was 40.91%(27/66).The expression of AFP mRNA in the peripheral blood in patients with liver cancer was significantly related to microvascular invasion and metastasis(P < 0.05 or < 0.01),but the expression had no relationship with sex,age,HBV infection,cirrhosis,AFP concentration,tumour size and number,and Edmondson grading(P>0.05).The overall 1,2,and > 3 years survival rates of patients with positive AFP mRNA after surgery were 66.7%(18/27),38.9%(7/18),28.6%(2/7),respectively.The overall 1,2,and ≥3 years survival rates of patients with negative AFP mRNA after surgery were 84.6%(33/39),60.6%(20/33),45.0%(9/20).There was statistical significance between the survival rates of AFP mRNA-negative patients and AFP mRNApositive patients(P < 0.01).[Conclusions] The detection of AFP mRNA in the peripheral blood may provide clue for early microscopic metastasis.It can be a prediction index for postoperative recurrence.
9.The clinical value of mammary sentinel lymphoscintigraphy in breast cancer

Purpose: To evaluate the clinical value of mammary sentinel lymphoscintigraphy in breast cancer. Methods: 91 patients with breast cancer in stage T1-2N0 were injected with 55. 5MBq/0. 5ml 99mTc- human serum albumin on the surface of the lesion subdermally or 92. 5MBq/4ml unfiltered 99mTc-sulfur colloid in four divided doses around the lesion. 63 patients underwent mammary lymphoscintigraphy and sentinel node biopsy using a hand-held r-ray detector probe were performed 2 - 16 hr postinjection in 70 patients during breast surgery . Results: The sentinel lymph node( s) (SLN) could quickly be shown by these two tracers in 81% (51/63) cases . A SLN with low activity could be seen in 13. 7% (7/5.1) cases , in which 85.7%(6/7) were proved to have a metastasis. Lymph drainage to the internal mammary nodes occurred in 38. 5% (5/13) of patients with an inner-lesion and in 26. 3% (10/38) of patients with an outer-lesion . The SLN was successfully identified in 95. 7% (67/70) of the patients and the number of the nodes ranged from 1 -5 with an average 1. 6 per person . The accuracy of the SLN with respect to the positive or negative status of the axillary nodes was 92.53% (62/67) . The sensitivity , specificity, PPV, NPV of the method was 82.75% (24/29), 100% (38/38), 100% (24/24) and 88. 37% (38/43), respectively . Conclusions: Mammary lymphoscintigraphy in breast cancer is helpful for localizing SLN correctly and identifying abnormal lymph drainage.
10.Polyvinyl alcohol-chitosan-collagen as scaffolds for tissue engineering

Objective To fabricate a novel polyvinyl alcohol (PVA)-chitosan(Cs)-collagen (Col) composite material and to confirm the feasibility of its application as a scaffold in tissue engineering.Methods PVA was blended with chitosan and collagen.The water content,swelling ratio,and tensile strength of the scaffolds were tested.SEM was used to observe the histological modality of the cross section.Results Scaffolds which al'e composite with different molecular weight and different amount of Cs and Collagen ale made,with water content ranging from 60.15% to 72.50% and swelling ratio being from 185.33% to 317.57%.The tensile strength of the composite material is 5.70MPa.The inner modality and structure of the scaffolds varied as the proportion of the chemical components changed.Conclusion PVA-Cs-Col scaffolds have high water content and proper swelling ratio and ale rich in porous structure.When the blending proportion is Cs:PVA:COI=30:15:0.20,the scaffold performs best,which shows to be a suitable structure for tissue engineering scaffold.
