1.Follow-up after treatment of CIN2~3

Treatment of CIN2~3 includes cervical ablation,cervical local excision and hysterectomy,while cervical local excision is the standard procedure for CIN2~3.The recurrence rate was 0.7%~21% after surgery treatment of CIN2~3;The high risk factors of recurring are positive margin,serious lesion and age over 40.The follow-up items for postoperation involve cytology,colposcopy,ECC and HPV testing;If there were 2 LSIL or 1 HSIL for cytology,the patients should be given colposcopy examination and ECC.Retreatment must be clinically individual one.It is the same significance for follow-up and treatment of CIN2~3.

Objective The purpose of this study was to construct a rat model of luteal phase defect ( LPD) with mifepristone ( RU486 ) and investigate the improving effect of Bushen Zhuyun Recipe ( BZR ) on the gonad axis morphology of the LPD rat . Meth ods We established rat models of LPD by giving RU486 at 1 mL/100 g followed by treatment with BZR at 0.4 (medium dose) or 1.2 mL/100g ( high dose ) .Then we observed the changes of morphology of the hypothalamus pituitary ovary uterus . Results Electron microscopy revealed hypothalamic mitochondria vacuolization , degranulation of rough endoplasmic reticulum , dissolved nuclear membrane , and decreased secretory granules , while light microscopy exhibited slightly reduced density of pituitary cells , karyopyknosis of some of the cells ,and cytoplasmic vacuolization in the model rats .Electron microscopy also showed obvious shrinkage of cell nuclei , chromatin condensation , disordered cell arrangement , swelling mitochondrial cavity , and formation of vacuoles in the pituitary anterior lobe, while light microscopy also manifested increased ovarian follicular atresia , reduced granule cell layers and corpora lutea , thinned endometrial layers , decreased uterine glands of the tube wall , reduced glandular cavity , and increased interstitial collagen fibers .After BZR treatment, the number and volume of corpora lutea were increased , the granulosa cell layer and endometrial layer thickened , the number of uterine glands increased , and the gland cavity expanded . Conclusion The axes of the hypothalamus , pituitary, uterus,and o -varian were morphologically changed in the RU 486-induced rat models of LPD, while BZR could improve these changes .
3.Culture and functional identification of macrophages from human peripheral blood

Objective:To isolate monocytes from human peripheral blood mononuclear cells( PBMC) ,induce macrophages,and identify the function of macrophages.Methods:Monocytes were isolated from PBMC using magnetic activated cell sorting( MACS) anti-CD14 microbead.Sorted CD14+and CD14-cells were checked by flow cytometer to evaluate the efficiency of sorting.The sorted CD14+cells were cultured in IMDM media with 10%human AB serum and 10 ng/ml M-CSF for 7 days to generate macrophages,which were identified by morphological features and phagocytosis function.Results:A high purity of monocytes was obtained by MACS anti-CD14 microbead.The percentage of CD14+cells was 10% and 85.8% before and after sorting, respectively.The macrophages were approximately 40-45 μm in maximum diameter and had the fried egg colony morphological features after 7 days culture.The lymphoma ( Raji) cells were efficiently engulfed by macrophages.Conclusion: The high purity of CD14+monocytes is isolated from PBMC and monocyte-derived macrophages efficiently engulfed lymphoma cells.
4.Lower motor neuron lesion caused by single level lower thoracic disc protrusion

Objective To investigate clinical features of lower motor neuron lesion (LMNL) caused by the single level lower thoracic disc protrusion (LTDP),and to observe clinical outcomes of surgical treatment.Methods Between January 1997 and December 2009,17 patients with LMNL caused by single level LTDP underwent en bloc resection of the superior articular process,Cave-in 360° circumferential decompression and internal fixation in our hospital.MRI and CT scans were taken to confirm lesion levels:T10-11 in 4 patients of whom 3 had patellar clonus and ankle clonus,T11-12 in 5 patients of whom 4 had ankle clonus,and T12L1 in 8 patients who only had positive Babinski sign.The neurologic status was assessed using the Japanese Orthopaedic Association (JOA) scoring system.The muscle strength of the tibialis anterior was assessed using the Manual Muscle Test (MMT).Sagittal Cobb angle and cross-sectional area of the dural sac at the level of maximal compression in MRI were also observed.Results All patients were followed up for 22 to 76 months (average,48.6 months).The mean JOA score increased from preoperative 5.88±1.11 to 9.53±0.94 at final follow-up (t=16.143,P<0.05).The muscle strength of the tibialis anterior recovered to more than grade 4 in all patients.Postoperative Cobb angle was unchanged compared with that before operation.MRI indicated that the cross-sectional area of the dural sac at the level of maximum compression increased from preoperative 35.8±7.3 mm2 to postoperative 132.9±6.5 mm2 (t=70.78,P<0.05).Conclusion LMNL can be caused by LTDP.The eu bloc resection of the superior articular process,Cave-in 360° circumferential decompression and internal fixation can provide a satisfactory decompression effect and marked recovery of neurological function.
5.Effect of Protocatechuic Acid on Preventing PC12 Cells from Aamyloid Beta-Peptide-induced Toxicity and Its Mechanism

Objective To investigate the protective effect of protocatechuic acid(PCA)on the PC12 cell model of Alzheimer’s disease(AD)and to explore its mechanism . Methods Amyloid beta peptide 1-42(Aβ1-42)fiber polymers were identified by immunofluorescence. After PC12 cells were stimulated with the Aβ1-42 fiber polymers, the cellular morphology was observed at different time points of hour 0, 3, 6, 9, 12, 24 , and the cellular viability was tested by methyl thiazolyl tetrazolium(MTT)assay to monitor the modeling condition. The effect of PCA on PC12 cells was detected after PC12 cells were pretreated with the different contentions of PCA. Autophagy-related marker Beclin1 protein level was detected by Western blotting method to investigate the protective mechanism of PCA. Results Aggregated white Aβ1-42 mass was stable at hour 12 and 24, and showed no significant difference between the two time points, the cell damage rate being 40%. Therefore, we defined culturing time being 12 and 24 hours as the modeling condition of AD model. The cell viability was increased with 200-800 μmol/L of PCA after culturing for 24 hours(P<0.01) , and the Western blotting results showed that the Beclin1 protein expression was up-regulated by PCA. Conclusion PCA prevents PC12 cells from Aβ1-42-induced toxicity, the mechanism being related with the increase of cellular autophagy.
6.Comparison of two asthma models in rats

Aim Toestablishanexcellentratasthmamodel from using OVA+pertussis sensitized, OVA sensitized and per-tussissensitizedrats.Methods Thethreemethodswereusedto sensitize rats;methacholine bronchial provocation tests were per-formed to determine airway hyperresponsiveness;bronchoalveolar lavage fluid ( BALF) was prepared after the animals were chal-lenged by nebulized antigen. The differential white cell count in BALF was performed, and lung tissue was detected by morpho-logicalanalysis.Results AllofOVA+pertussissensitization,OVA sensitization and pertussis sensitization could deteriorate lung function, increase inflammatory cells and cause pathological change, and OVA + pertussis sensitized rat model had better effect than OVA sensitized and pertussis sensitized rat models. Conclusion OVA+pertussissensitizationandOVAaerosolisa successful rat asthma model.
7.Preliminary preparation of cognitive assessment table in nursing work on ethics problem

Objective To prepare cognitive assessment questionnaire on nursing ethics. Methods The methods of the research applied interview and face cross-sectional survey,prelimi-nary test paper was formed on the basis of the original research papers,and formal assessment test was formed with 17 questions.Using stratified sampling,468 nurses aged 19~53 years were col-lected from 4 first-class hospitals,reliability and validity assessment were tested through factor analysis regression and other statistical analysis methods.Results Nurses in nursing work on ethics problem of cognitive assessment table eventually confirmed 17 items,internal consistency co-efficient of the questionnaire was 0.922,and internal alpha coefficients of 2 factors were 0.901 and 0.921.By factor analysis,basic nursing operation,protection of privacy and self-supervision 2 factors were extracted and they could explain that the total variance was 53.589%,and project load value was 0.517~0.758.Conclusion The nurses to ethics problems in nursing work cogni-tive assessment scale have good reliability and validity,which can be used to study the nursing ethics.
8.Growth regulation of ovarian cancer cell line HO-8910 by transforming growth factor β1 in vitro

Objective To further understand the role of growth regulation of human ovarian cancer cells by transforming growth factor (TGF) β1.Methods The cell proliferation, cAMP synthesis, gene expression, and induction of programmed cell death (PCD) in human epithelial ovarian cancer cell line HO-8910 cells exposed to TGFβ1 in vitro were studied.Results TGFβ1 inhibited cell growth and DNA synthesis, and induced G0/G1 arrest in cell cycle. It could also trigger PCD in cells. This induction of PCD may occur within G0/G1 phase. Meanwhile, the assay also showed that TGFβ1 could inhibit the mRNA expression of c-myc, EGFR and TGFβ1 genes in cells.Conclusions TGFβ1 can not only act as an autocrine to inhibit cell proliferation, but also trigger PCD in HO-8910 cells. These functions may be fulfilled through some specific signal transduction pathways.

10.Effects of social relational quality on social avoidance and distress in patients with permanent cystostomy

Objective To investigate the social relational quality and social avoidance and distress in patients with permanent Cystostomy , and explore the effects of social relational quality on social avoidance and distress.Methods A total of 203 patients with permanent cystostomy were surveyed using the social relational quality scale and the social avoidance and distress scale .The scores of social avoidance and distress were compared among patients with different clinical characteristics , and the hierarchical regression analysis was applied for exploring the effects of social relational quality on social avoidance , distress.Results The total scores of social relational quality and social avoidance and distress in patients with permanent cystostomy were (44.18 ±6.60) and (15.38 ±6.32) respectively.Patients in different genders, ages, marital statuses, occupations, catheterization times and primary diseases had significantly different scores of social avoidance and distress ( P<0.05).The hierarchical regression analysis showed that gender , occupation, time of catheterization, primary disease, family cohesion, family commitment and friendship were independent factors in predicting the social avoidance and distress of patients with permanent cystostomy ( P<0.05 ) .Conclusions The social relational quality of patients with permanent cystostomy is at low level , while the social avoidance and distress is at relatively high level .The social relational quality determines the social avoidance and distress of patients with permanent cystostomy , so improving the social relational quality is promising to avoid the social avoidance and distress of patients with permanent cystostomy .
