Objective To compare the short and long term effect of knee osteoarthritis treatment with two therapeutic methods which are simple high cut of fibula and arthroscopic lavage debridement combined with fibular osteotomy.Methods Data of 45 consecutive patients with knee osteoarthritis were retrospectively observed and selected,and then they were divided into two groups according to the operation method.23 patients with simple high cut of fibula were selected as the control group,and 22 patients with arthroscopic lavage debridement combined with fibular osteotomy were selected as the observation group.Results The VAS and AKS scores of the control group at the time of 3m,6m,1 2m were (4.1 3 ±0.76)points,(1 07.04 ±21 .53)points;(2.70 ±0.64)points,(1 41 .1 7 ±1 2.57)points;(2.43 ±0.79)points,(1 48.26 ±5.81 )points;and the scores of the observation group were (3.45 ± 0.60)points,(1 26.64 ±1 4.24)points,(2.70 ±0.73)points,(1 46.45 ±7.26)points,(2.41 ±0.85)points and (1 48.26 ±5.81 )points.The differences between the two groups were statistically significant at the time of 3 months after surgery(t =3.32,-3.62,all P <0.05),the effect of observation group was better than the control group;but the other indices had no statistically significant differences (t =0.51 ,-1 .72,0.1 1 ,-0.20,all P >0.05 ). Conclusion The simple high cut of fibula with small trauma,less cost,less surgical difficulty,and long -term clini-cal curative effect is not less than the arthroscopic lavage and debridement combined with fibular osteotomy.There-fore,it should be carried out in the primary hospitals widely.
4.Analysis of variants located in 3'UTR regions of NOTCH1 and JAG1 genes for children with conotruncal heart defects

Objective · To explore the correlation between variants located in 3' untranslated regions (3'UTR) of NOTCH1 and JAG1 genes and conotruncal heart defects (CTD). Methods · Six hundred CTD children without 22q11 deletion and three hundred healthy children were enrolled in this hospital-based case-control study. Variants located in the 3'UTR regions of NOTCH1 and JAG1 genes were detected by high-throughput sequencing. The accuracy of the variants were verified by PCR and Sanger sequencing. Online software PicTar, TargetScan and microRNA.org were used to make functional predictions. Results · One mutation and three SNPs were found in the 3'UTR of NOTCH1. Three mutations and six SNPs were found in the 3'UTR of JAG1. The genotypic distributions of two SNPs (rs3840074 and rs8708) located in JAG13'UTR between CTD group and the controls were statistically significant (both P<0.05). Results of prediction showed that all the four mutations and two meaningful SNPs could bind to microRNA. Conclusion · The variants located in 3'UTR regions of NOTCH1 and JAG1 genes may be related to the occurrence of CTD.
5.THE EXPRESSION OF PECAM-1 IN RAT COLON ADENCARCINOMA AND LYMPHATICS

Objective To observe the expression of PECAM-1 in colorectal cancer in rat and to explore the mechanism of lymphatic metastasis of tumor. Methods Wistar rats were used to make the animal model of primary colorectal cancer,by using HE staining,the cancer type and progression of cancer were devided.By immunohistochemistry staining,the expression of PECAM-1 in tumor tissue,blood vessels and lymphatic vessels were observed. Results PECAM-1 expression in blood vessels and lymphatic vessels were observed in normal tissue,while its expression on tumor tissue and lymphatic vessels decreased according to the cancer progression.Conclusion PECAM-1 may be involved in the early process of tumor cells adhering to the endothelium;decreased expresson of PECAM-1 on lymphatics may be related with lymphangiogenesis and the opening of endothelial junction of lymphatic vessels.
6.Effects of irbesartan on expression of HGF at mRNA and protein levels in rats with myocardium infarction

AIM:To investigate the effects of irbesartan on the expression of hepatocyte growth factor (HGF) at mRNA and protein levels in rats with myocardial infarction (MI), and to explore the mechanisms of irbesartan attenuating myocardial fibrosis.METHODS:The male Wistar rat model of MI was successfully established.The surviving rats 24 h after the operation were randomly divided into 3 groups:model group,irbesartan group and sham group, with 9 rats in each group.The rats in irbesartan group were treated with the solution of irbesartan (50 mg·kg-1·d-1) by intragastric administration, while the rats in model group and sham group received the equal volume of saline by the same way.The body weight and left ventricle mass (LVM) of the rats were measured at the 4th week after operation, and the pathological changes of the ischemic myocardium were observed with HE staining.Meanwhile, the expression of HGF at mRNA and protein levels was detected by RT-qPCR and Western blot.RESULTS:HE staining showed that the myocardial cells in sham group were in neat arrangement, while the cardiac structure in model group and irbesartan group was in disorder.The pathological changes in irbesartan group were less than that in model group.No difference in the body weight at the 4th week after operation was observed, while the LVM was significantly different among the 3 groups (P<0.01).The LVM in model group was higher than that in sham group (P<0.01), and that in irbesartan group was higher than that in sham group (P<0.05).The LVM in irbesartan group was lower than that in model group (P<0.05).The expression of HGF at mRNA and protein levels was detected in each group.The expression of HGF at mRNA and protein levels in irbesartan group was higher than that in sham group (P<0.05), and that in model group was higher than that in sham group (P<0.01).Moreover, the mRNA and protein levels of HGF in irbesartan group were lower than those in model group (P<0.05).CONCLUSION:The LVM of MI rats with the treatment of irbesartan was reduced obviously at the 4th week after operation, and the pathological changes were also improved.At the 4th week after the operation, the treatment of irbesartan inhibited the expression of HGF at mRNA and protein levels.

Objective To study the inhibition of Cephalanoplos(Bunge) kitam extract on human liver cancer cells.Method The inhibition of Cephalanoplos(Bunge) kitam extract on human liver cancer cells was measured with MTT colorimetic method,The human liver cancer cell apoptosis was studied by transmission electron microscope and TUNEL staining.Results The inhibition was enhanced with concentration increase of Cephalanoplos(Bunge) kitam extract and time of culture.With Cephalanoplos(Bunge) kitam extract 10,20,40,80 mg/L for 24,48,72,96h,the cell apoptosis was significant by T test.Conclusion Cephalanoplos(Bunge) kitam extract can inhibit the growth of BEL-7402 cancer cells.
8.ULTRASTRUCTURAL STUDIES ON INITIAL LYMPHATICS OF HUMAN GASTRIC CARCINOMA

Objective To study the ultrastructural features of initial lymphatics of gastric carcinoma,to discuss the metastasis mechanism of malignant cells by lpmphatics. Methods The postoperative samples of patients with gastric cancer were obtained in center,peripheral and normal region of cancer.Common Resin-embedded specimens were made.Lymphatics were located by hemithin sections under light microscope and observed by ultrathin sections under electron microscope. Results There were no lymphatics in carcinomatous centre region.Compared with normal region,there were more initial lymphatics in peripheral region of gastric carcinoma.The lumina were dilated and irregular and the endothelium were destroyed;The organelle of endothelial cells changed obviously.The distribution and morphology of the lymphatics did not changed evidently in the normal region.Conclusion These results suggest that amount,ultrastructure and morphology of lymphatics changed in peripheral region of carcinoma;Gastric cancer cells may invade lymphatics via destroyed parts of endothelium or interendothelial openings.
9.Prokaryotic expression and immunogenicity of Fba,a novel fibronectin-binding protein of group A streptococus

Objective To express the novel fibronectin-binding protein Fba of group A streptococcus(GAS)and analyze its immunogenicity,so that to evaluate the immune responses to GAS infection.Methods fba gene was amplified by polymerase chain reaction(PCR)and confirmed by sequencing.Then it was cloned into pGEX4T-2 vector and Fba protein was expressed in E.coli BL21.The protein expression was identified by enzyme-linked immunosorbent assay(ELISA)and Westernblot.The sera from mice infected with GAS and anti-streptolysin-O positive patients were detected using microtiter plates coated with purified Fba protein as antigen.Afterward Balb/C mice were immunized with this purified protein and the sera were collected after the third immunization for the detection of IgG titer.Results It was confirmed by ELISA and Western blot that the recombinant Fba protein had a specific affinity with anti-Fba sera of rabbit.The anti-serum IgG titer of mice imrnunized with Fba protein was up to 1:4800.Conclusions GAS infection or Fba protein immunization are able to induce high serum titer of anti-Fba which could react specifically with the recombinant Fba protein.It indicates that Fba protein has good immunogenicity and antigenicity.So Fba protein could be a GAS candidate vaccine and an important tool to detect anti-GAS titer in GAS infected patients.
10.Examination of gastrointestinal transmitting function with small amount of barium meal and its clinical significance

Objective To investigate the effect, preponderance and clinical significance of the examination of gastrointestinal transmitting function with small amount of barium meal. Methods Thirty cases of healthy subjects and 50 patients with constipation swallow 40 g barium meal once, then abdominal fluoroscopy or plain films were taken until barium meal being egested totally. Results The transit time of stomach, small intestine, colon, rectum and the whole digestive tract were (189?64)minutes, (330?108)minutes, (2239?659)minutes, (136?49)minutes, (2801?735)minutes in healthy subjects and (212?87)minutes, (366?94)minutes, (3665?1322)minutes, (650?219)minutes, (5973?2341)minutes in constipation patients respectively. The transit time of colon and rectum in constipation patients was obviously longer than that of healthy subjects (P0.05). Conclusion The pictures of the examination with small amount barium meal are clear, precise, reliable, and provide exact data for constipation classification.
