1.Expression and significance of telomerase in development process of scar formation

ObjectiveTo study the expression of telomerase of fibroblasts in the formation and development of scar,and to investgate the relationship between the telomerase activity and the formation and development of scar.MethodsExpression of telomerase was detected by immunohistochemical techniques in 18 specimens from granulation tissue,17 keloid,16 specimens from hypertrophic scar,28 specimens from mature scar,32 specimens from normal skin,and SPSS16.0 statistics software was used to analyze the relationship between telomerase and scar.Results It was shown that the positive expression rate of telomerase in the granulation tissue group was 94.4 %,that in the keloid group was 58.8 %,that in the hypertrophic scar group was 18.8 %, and that in the mature scar and in the normal skin was zero. comparisons between groups,in addition to the normal scar group and normal skin group,other groups were statistically significant differences.ConclnsionsScar formation is a process with multi-factor participation,while telomerase activation is an important factor. Detecting telomerase activity in the development process of scar formation may determine the prognosis.Through the control of telomerase activity in the pathogenesis of scar may become a new approach of scar treatment.
2.Diagnostic and differential diagnostic of primary plasma cell leukemia and lymphoma with increased plasma cell

Purpose To investigate the diagnosis,differential diagnosis and clinical manifestation of primary plasma cell leukemia (PPCL) and lymphoma with increased plasma cell.Methods Through clinical data and cell morphology,flow cytometry (FCM),immunofixation electrophoresis and immunohistochemistry of EliVision two-step examination were used to analyze 7 cases of PPCL and 3 cases of lymphoma with increased plasma cell.Results All patients with PPCL and lymphoma with increased plasma cell presented with anemia,thrombocytopenia,fever,liver and spleen and lymph node swelling.The proportion of plasma cells in peripheral blood morphology were larger than 20%,accompanied by morphological abnormality.FCM of peripheral blood showed all 7 cases of PPCL expressed CD38 and CD138,CD56 expression in the 2 cases and CD20 in the 2 cases.The light chain (Lamda,Kappa) showed a monoclonal restricted expression,which was consistent with the diagnosis of PPCL.CD19 and CD45 were weakly positive in 3 cases of lymphoma with increased plasma cell,CD38 and CD138 were positive,and no restricted expression was found in light chain IgL,wich belonging to the immunophenotypes of normal plasma cells.Of 3 cases of light chain (Ig) without restrictive expression,2 of them were angioimmunoblastic T-cell lymphoma (ATCL) and 1 case was CD30-positive sinusoidal large B-cell lymphoma (CD30 + SLBCL) that confirmed by lymph node biopsy and pathological examination.Conclusion The PPCL and lymphoma with increased plasma cell have the same clinical manifestations and similar morphological characteristics of blood cells.The diagnosis of PPCL should be combined with immunoelectrophoresis and FCM,and the diagnosis of lymphoma with increased plasma cell needs to be confirmed by histological examination of lymph nodes.
3.Determination of protein binding rate of oleanolic acid in human plasma and serum albumin.

A LC-MS method was established for the determination of the protein binding rates of oleanolic acid in human plasma and serum albumin. The equilibrium dialysis combined with LC-MS to determine the total concentration in plasma and free drug concentration of oleanolic acid was carried out. The human plasma protein binding rates of oleanolic acid at three concentrations were 79.6%, 81.9% and 63.3%, respectively. The human serum albumin protein binding rates of oleanolic acid at three concentrations were 53.5%, 56.6% and 47.7%, respectively. The method is shown to be simple, accurate, sensitive and specific for the determination of biological samples. The protein binding rates in human plasma and serum albumin were of high strength.
4.Construction of p300 specific siRNA vector and its effect on GATA4 expression in cardiac muscle cells

Objective To construct the p300 specific siRNA vector in mice and observe its effect on cardiac transcription factor,GATA4,for further study of the effect of p300-mediated histone acetylation on heart growth. Methods Three recombinant plasmid vectors ( p300 RNAi1,p300 RNAi2,and p300 RNAi3s) ,designed and constructed according to the conservative sequence of mice p300,were respectively tansfected into in vitro cultured cardiac muscle cells of suckling mice. 6.Comparative study on dissolution of Xianlinggubao capsules prepared by different processes.

OBJECTIVETo study the dissolution characteristics of seven active components of Xianlinggubao capsules in vitro, and compare the dissolution of Xianlinggubao capsules prepared by different processes.
METHODThe dissolution of Xianlinggubao capsules was determined by small cup method with the rotating speed of 100 r x min(-1) and phosphate buffer (pH 6.8) as dissolution medium. The different dissolution rates in vitro of seven kinds of active components (icariin, epimedin C, asperosaponin VI, psoralen, isopsoralen, salvianolic acid B and tanshinone IIA) of Xianlinggubao capsules were investigated. The contents of active components were determined by HPLC. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2).
RESULTThe results of determination showed that the similarity factor values (f2) of the dissolution curves of seven active components after different preparation processes were all less than 50.
CONCLUSIONThere is significant difference in dissolution of active components between Xianlinggubao capsules of different preparation processes. The accumulative dissolution percentages of the active components in new Xianlinggubao capsules are higher than that of Xianlinggubao capsules.

7.Dissolution of tanshinone IIA from xianling gubao capsule.

OBJECTIVETo establish a method to determine the dissolution of Tanshinone IIA in xianling gubao capsules, and compare the dissolution of Xianling Gubao capsules prepared by different processes.
METHODThe small cup method was used at a rotary speed of 100 r x min(-1), the dissolubility was inspected in the following 6 mediums: (1) water; (2) 0.1 mol x L(-1) hydrochloric acid solution; (3) pH 6.8 phosphate buffer; (4) pH 6.8 phosphate buffer solution + 0.5% cholic acid; (5) pH 6.8 phosphate buffer solution + 1.0% cholic acid; (6) pH 7.6 phosphate buffer solution + 4. 0% cholic acid. A HPLC method was used to determine the dissolution samples of tanshione IIA in different dissolution mediums. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2).
RESULTThe accumulative dissolution percentage was the highest while pH 7.6 phosphate buffer solution + 4.0% cholic acid was used as medium. The similarity factor values (f2) of tanshinone IIA dissolution curves of different preparation processes of Xianling Gubao capsules were all less than 50.
CONCLUSIONThere is significant difference in tanshinone IIA dissolution between Xianling Gubao capsules of different preparation processes. The accumulative dissolution percentages of the tanshinone IIA in new Xianling Gubao capsules is higher than that in the capsules prepared by other preparation processes.

8.Primary study on quality standard of carbonizing drug characteristic of ginger carbon.

OBJECTIVETo establish the quality standard for carbonizing drug characteristic of ginger carbon.
METHODGingers and different carbonized gingers were compared by the absorption of pigment, tannin content, pH, mouth's coagulation time and bleeding time.
RESULTThe study resulted in the recommended carbonizing standard that the absorption capacity shall not be less than 7.50 mg x g(-1) for methylene blue, the tannin content shall not be less than 2.103 mg x g(-1), the pH shall be (5.50 +/- 0.10), and coagulation time and bleeding time shall be the shorter the better.
CONCLUSIONThe established assessment standard for carbonizing drug characteristic of ginger carbon is reasonable, easily operated and feasible.

9.Effect of Tripterygium glycosides combined with Irbesartan on podocyte injury in diabetic nephropathy

Objective To discuss effect of Tripterygium glycosides combined with Irbesartan on podocyte injury in dia-betic nephropathy. Methods All 80 cases with diabetic nephropathy were selected as subjects, and were divided into group A, group B, group C and group D. Based on active control of blood sugar and other treatment,group A was treated with Tripterygium glycosides combined with Irbesartan, group B was treated with Tripterygium glycosides, and group C was treated with Irbesartan. Sufficient number of cells,and urinary CTGF,TGF-β,OPN,BMP-7 concentra-tion were compared. Results After treatment,proteinuria, podocyte number,CTGF,TGF-β, OPN of four group were decreased apparently,and BMP-7 was increased(P all约0.01 or 0.05). Proteinuria,podocyte number,CTGF,TGF-β,OPN of group A,B,C were lower than group D,and BMP-7 was higher than group D(P all<0.01). Urinary podocyte number and urine protein were positively correlated with each other, and urinary podocyte number and urine protein were positively correlated with CTGF, TGF-β,OPN, negative correlated with BMP-7 (P all <0.01). Conclusion Tripterygium glycosides combined with Irbesartan for diabetic nephropathy has a role in protecting the kidney podocytes.
10.Comparative study of absorption kinetics in intestines of rats on Xianlinggubao capsule prepared by different technologies.

OBJECTIVETo study the characteristics of intestinal absorption of psoralen and isopsoralen of Xianlinggubao capsule, and compare the absorption of Xianlinggubao capsule prepared by different technologies.
METHODNon everted gut sac method was applied to investigate the influence of absorption sites and drug concentration on psoralen and isopsoralen absorption, which were determined by HPLC.
RESULTAlthough the absorption rate constants of psoralen and isopsoralen in duodenum were more than that in jejunum and ileum, there was no significance difference between them. The absorption rate constants of psoralen kept at the same level when the concentrations of drug solution were at middle and low level, while the absorption rate constant at high level was absolutely lower than them (P < 0.05). The results of isopsoralen were the same as psoralen's.
CONCLUSIONIntestinal absorption of psoralen and isopsoralen may be affected by the dissolution. The absorption rate constants of psoralen and isopsoralen in new Xianlinggubao capsules are higher. The absorptions of active components absorption has significant difference in different preparation processes of Xianlinggubao capsule.
