Objective To prepare Mtb8.4 gene vaccine and to study the cellular immune response induced by the vaccine. Objective To establish the determination method of baicalin in the children's YifeiPingchuan mixture by RP-HPLC.Methods A Kromasil C18 column was used with methanol-(0.4％)phosphoric acid-triethylamine (45 ∶ 55 ∶ 2 drops) as mobile phase with a column temperature of 28 ℃.The flow rate was 1.0 ml/min.The detection wavelength was set at 277 nm.Results The linear range of baicalin was 0.040～1.000 μg (r=0.9999) with a average recovery of 99.82 and RSD of 1.01％.Conclusion The method for quantification of baicalin was sensitive,accurate,and convenient,which can also be used as one of basis of quality standard for Yifei-Pingchuan mixture.

Objective To investigate the different effect of positive and negative emotion induced by film clips on explicit and implicit memory,and whether there is mood congruency effect. Methods Thirty participants who were randomly assigned to three experimental groups were investigated. They were exposed in different film clips and then worked on memory task. After each emotional condition,the 5-point Self Assessment Scale was registered , Buchner's processing dissociation corrected model was employed to separate explicit and implicit memory. Results (1) The emotion arousal level of different films is significantly different, the scores of negative film and positive film were more than that of neutral film( (3.24 ± 1.02),(2.85 ± 1.35),(1.12 ± 1.42) , P＜0.01) ; the scores of emotional valence of positive,negative and neutral priming condition were (4. 10 ±0. 20), (3. 60 ± 0.22) and (3.20 ±0. 97), and there was a significant difference between emotion priming conditions (F(2,27) = 46.81, P＜ 0. 01). (2) There was a significant difference among different emotion priming conditions in R (F(2,87) = 30.129, P＜0.05) .and there was no significant difference among different emotion priming conditions in A. Conclusions Explicit memory showed mood congruency memory effect,but implicit memory,neither the positive emotion priming group nor the negative emotion priming group exhibited mood congruency memory effect.

OBJECTIVE: To discuss the method of reducing error in estimating postmortem interval (PMI). METHODS: Two hundred and fifty-six solved murder cases from 2003 January to 2013 January in Changzhou and Nanjing City were collected, The PMI of all cases was estimated by traditional method and then compared with the real PMI obtained after the cases were solved. The cases were grouped according to the PMI, the accuracy was calculated, and the reasons of suboptimal PMI were analyzed. RESULTS: The accuracies of early PMI (less than 12h and 13-24 h) were 90% and 89%, respectively; while the accuracies of late PMI (1-7 d, 1-2 weeks, 3-4 weeks, 1-6 months, 7-12 months and 1-5 years) decreased over time, being 79%, 76%, 83%, 79%, 60% and 50%, respectively. The common reasons of estimating error included improper inference methods, water submerged body, extreme temperature, lack of objective evidence, intentionally abandoned body, and changed or destroyed scene, etc. CONCLUSION The multiple index data can reduce the error in estimating PMI.

AlM: To observe the early tear film changes after trabeculectomy using Keratograph 5M.METHODS:Fourty-one patients (46 eyes) of glaucoma who underwent trabeculectomy were involved. First tear break up time ( FTBUT ) , average tear break up time (ATBUT), non-invasive tear meniscus height (NlTMH), lipid layer thickness and meibomian gland scores were measured at 1d preoperatively and at 1d, 1wk, 1mo postoperatively.RESULTS:At 1d, 1wk and 1mo postoperatively, FTBUT and ATBUT decreased greatly (P<0. 05), as well as lipid layer thickness (P<0. 017). NlTMH at 1d postoperatively increased significantly, but at 1wk and 1mo postoperatively decreased greatly (P<0. 05). There was no significantly difference in meibomian gland scores between preoperation and postoperation (P>0. 05). CONCLUSlON: Keratograph 5M could be used to evaluate tear film function rapidly and accurately. Trabeculectomy significantly alters tear film stability and tear secretion in the short term after operation.

Aim To study the role of TNF-α/NF-κB signaling in matrix metalloproteinase ( MMP)-9 expres-sion induced by lipopolysaccharide ( LPS ) in airway epithelial cells, and to investigate the effects of lenti-virus mediated RNAi targeting a disintegrin and metal-loproteinase 17 ( ADAM17 ) gene on MMP-9 expression induced by LPS. Methods The ADAM17 siRNA ex-pression vector was constructed, and packaged to re-combinant lentivirus in 293T cells. The HBE4-E6/E7 cells were pretreated for 30 min by NF-κB inhibitor ( PDTC) and a recombinant human TNFR p75-Fc fu-sion protein ( Etanercept) , or infected by the recombi-nant lentivirus for 72 h, and then stimulated for 24 h by LPS or TNF-α. The release of TNF-α was detected by ELISA. The mRNA and protein levels of MMP-9 were analyzed respectively by RT-PCR and Western blot. NF-κB activity was detected by electrophoretic mobility shift assay. Results LPS and TNF-α signifi-cantly increased MMP-9 mRNA and protein expressions and the activation of NF-κB in HBE4-E6/E7 cells ( P<0. 05 ) . Etanercept and PDTC significantly inhibited MMP-9 expression and the activation of NF-κB induced by LPS ( P<0. 05 ) . Lentivirus mediated RNAi targe-ting ADAM17 significantly decreased TNF-α produc-tion, inhibited MMP-9 mRNA and protein expressions and the activation of NF-κB induced by LPS in HBE4-E6/E7 cells ( P <0. 05 ) . Lentivirus mediated RNAi targeting ADAM17 did not inhibit MMP-9 mRNA and protein expressions and the activation of NF-κB in-duced by TNF-α ( P>0. 05 ) . And PDTC significantly inhibited MMP-9 mRNA and protein expressions and the activation of NF-κB induced by TNF-α ( P <0. 05 ) . Conclusions TNF-α/NF-κB signaling partic-ipates in the regulation of MMP-9 expression induced by LPS in airway epithelial cells, and lentivirus-media-ted RNAi targeting ADAM17 plays an important role in that signaling pathway upstream by regulating TNF-αrelease.
