Objective To investigate the effect of lipopolysaccharide (LPS) on stimulating high mobility group box-1 protein (HMGB1) mRNA expression in peritoneal macrophages in rats and its potential signaling mechanism. Methods Abdominal macrophages obtained from male Wistar rats were seeded on 24-well (1?10 6 cells/well) tissue culture plates. The cells were incubated for 3 days before they were stimulated with LPS, fludarabine (specific inhibitor of signal transducer and activator of transcription 1, STAT1), or rapamycin (specific inhibitor of signal transducer and activator of transcription 3, STAT3). After being stimulated, macrophages were denatured directly in tissue culture plates to determine HMGB1 mRNA expression. The time-dependent and dose-dependent responses between LPS stimulation and HMGB1 mRNA expression were analyzed, and the effect of treatment with fludarabine and rapamycin on HMGB1 mRNA expression was also observed. Results After being stimulated with 75-100ng/ml LPS, the HMGB1 mRNA expressions in macrophages were up-regulated markedly, peaking at 24-36 hours, and remained elevated up to 48 hours. It was found that the HMGB1 mRNA expression was significantly inhibited by treatment with either fludarabine (100?mol/L) or rapamycin (25ng/ml). Conclusions These data suggest that LPS stimulation can result in up-regulation of HMGB1 mRNA expression in peritoneal macrophages in rats. Janus kinase/STAT pathway may be involved in modulating HMGB1 mRNA expression in LPS-activated macrophages.

Objective:To study effect of tripterygium glycosides(TG)on T-lymphocyte subsets in rats with idiopathic thrombocytopenic purpura(ITP),and further to discuss the possible mechanism of TG in treating ITP.Methods:ITP animal model was established by intraperitoneal injection of exogenous anti-platelet–serum(APS).Sixty model rats were randomly divided into five groups:normal group, saline group, the high and low dosage of TG groups, prednisone group.Every rat was giving liquid medicine by intragastric administration one time per day for ten days 36 hours after the first intraperitoneal injection of exogenous APS.The changes of peripheral hemogram, the amount of megakaryocyte of bone marrow and T-lymphocyte subsets were observed. Results:In high dosage of TG group, the number of platelet count increased obviously, the number of megakaryocyte in bone marrow,CD8+and CD4+decreased,and CD4+/CD8+increased.Conclusion:High dosage of TG had obviously therapeutic effect on IPT rat, such as increasing platelet count obviously, reducing number of megakaryocyte in bone marrow obviously and ameliorating T-lymphocyte subsets. The mechanism may be related to regulating the cell immune function.

Objective To investigate the influence of lentivirus mediated short hairpin RNA(shRNA)target to human papilloma virus(HPV)16 E6 on invasive ability of cervical cancer Caski cells. Methods Lentivirus was produced after shRNA target to human papilloma virus(HPV)16 E6 and to nonsense was cloned to lentivirus work vector.Infection ratio was assessed by assay of EGFP positive cells of Caski.Total mRNA of E6 was determined by RT-PCR after Caski cells were infected by lentivirus. The change of E6 protein expression was analyzed by Western blot. The invasive ability of Caski cells was assayed employing Transwell. Results The optimal MOI (Multiplicity of infection)of lentivirus to Caski was 2.5.Total mRNA and protein of E6 were decreased (by 70%and 63%)in interfering group compared with control group. The invasive ability of Caski cells also reduced after infected by lentivirus. Conclusion shRNA mediated by lentivirus can inhibit expression of HPV16 E6 and invasive ability of cervical cancer cells.

strains that could produce raw starch-digesting glucoamylase were isolated from soil and mildewed rice. The highest raw starch-digesting glucoamylase activity strain named OR-1 was identified as Rhizopus.sp.The raw starch-digesting glucoamylase activity of the strain is 90U/mL.Through UV and NTG mutagenesis, the raw starch-digesting glucoamylase activity raised to 200U/mL and 325U/mL respectively. The RDA were 70% and 65% respectively.

Microbiology 1992;0(05):-
A bacterium that contains a chitin deacetylase has been isolated from ocean soil. It has been identified Baciluus .The best suitable condition of producing this enzyme is that pH is 4.0,35℃,and the incubate time is 80 hours in the present of Ca 2+ as well as having no chitosan. The best temperature of this enzyme when it acts is 40℃~50℃ and the best pH range is 4.5~5.0.

Objective To investigate the clinical effect and the prospect of Infliximab in treatment of intravenous immunoglobulin (IVIG)-resistant Kawasaki disease (KD) patients. Methods Clinical features, inflammatory markers and coronary changes were observed in 2 cases of IVIG-resistant KD patients hospitalized in Peking University First Hospital, who were treated effectively by Infliximab.Relevant researches on the mechanism and progress of the Infliximab treatment for IVIG-resistant KD in the last 10 years were reviewed at the same time. Results Two KD patients hospitalized in Peking University First Hospital had been treated with 2 g/kg IVIG for 2 times and followed by methylprednisolone treatment. However, fever and other clinical manifestations occurred again after 2 days and 6 days when temperature returned normal. They both defervesced and all the symptoms were improved after 1 dose of Infliximab (5 mg/kg) by laboratory examinations.Four published literatures of the basic research and 9 retrospective or prospective clinical researches of Infliximab treatment of KD showed that Infliximab alleviated the inflammatory level in the KD patients significantly.Complete remission was up to 72.73%-92.11%.Those KD patients defervesced within 12 h,with dramatic improvement of symptoms and signs.Arthralgia also disappeared in the patients with arthritis.Only 1 case was complicated with hepatitis in the acute phase and cholecystitis in recovery time.A phase 3 randomised,double-blinded,placebo-controlled trial had been done to assess the addition of Infliximab to the standard therapy. Conclusions Infliximab is a feasible choice for IVIG-resistant KD patients.Efficacy and safety of Infliximab for KD treatment have been proved in the literature. However,Infliximab for KD treatment has not been indicated in the drug instruction,so the informed consent from the guardians and Ethics Committee is needed.

2 mmx3 mm experimental windows were prepared in adamant slippery surfaces of 7 fresh uprooted permanent teeth. The teeth surfaces in the windows area were demineralized to create artificial caries mould of early stage by aciding the experimental teeth surfaces of 0, 12, 24, 48, 72, 96, 120 hours with demineralized liquid (pH 4.5) in vitro. The demineralized changes on the experimental teeth surfaces were detected by dental Optical Coherence Tomography (OCT) system, which were newly developed by our research group recently, and the detecting results were compared with clinical digital photomicrography and scanning electron microscopy on the same tooth sample, to checkout the efficacy and feasibility of dental OCT for early quantification detecting of artificial enamel demineralization in vitro. The dental OCT system can safelydetect early enamel demineralization of micron level and noninvasively obtain fine resolution quantification information both in surfaces view and sectional view; OCT could accurately detect surface demineralization changes on the experimental windows of artificial dental caries as early as after 12 hours aciding treatment, earlier than the visual inspection and clinical digital photomicrography. OCT could obtain both superficial view and sectional view of quantificational demineralization in early enamel caries homeochronously, and had high correlation to the results of ultramicromorphological changes detected by scanning electron microscopy. Dental OCT system developed by our group could accurately detect early artificial dental caries atraumaticly with high sensitivity and safety. Moreover, it can obtain quantification data in micron level without damaging the experimental teeth samples.
