1.Effects of 5-Aza-2'-deoxycytidine on biological behaviors and the RASSF1A gene expression of human lung carcinoma cell lines H460

Objective To investigate the effects of demethylating agent 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the biological behaviors and the expression of RASSF1A of lung carcinoma cells H460. Methods Lung carcinoma cell line H460 were treated with 5-Aza-CdR. Cell proliferation was determined by MTT assay and colony forming test. The methylation status of RASSF1A gene was detected by PCR. The expression of RASSF1A protein was measured by Western blotting, and the cell cycle was analyzed by flow cytometry. Results With 5-Aza-CdR treatment, the proliferation speed of H460 lung carcinoma cells was slowed down and the colony formation rate of H460 cells was decreased significantly compared with control group (38.5 %, 27.5 % and 60.5 % in 5-Aza-CdR 5 and 10 μmol/L groups and control group, respectively, P <0.05). The methylation degree in the promoter of RASSF1A gene was decreased and the expression of RASSF1A protein was detected after 5-Aza-CdR treatment. 5-Aza-CdR induced G1 phase arrest of the H460 cells. Conclusion The hypermethylation of CpG island in the promoter of RASSF1A gene results in the loss of RASSF1A protein expression in human Lung carcinoma cell line. The demethylating agent 5-Aza-CdR could restore RASSF1A gene expression.These findings provide theoretic evidence for clinical treatment of human lung carcinoma with demethylation agent 5-Aza-CdR.
2.Establishment and evaluation of indirect competitive enzyme-linked immunosorbent assay for detecting the concentration of trypsiongen activation peptides

Objective To establish an ELISA method for detection of the concentration of TAP in human urine samples.Methods TAP-BSA was used as coating antigen. The TAP was a competitor to TAP-BSA. They reacted to the limited amount of monoclonal antibody against TAP.Results The optimal concentration of the coating antigen was 250ng/ml.The dilutions of monolclonal antibody against TAP and sheep anti-mouse IgG were 25?g/ml and 1∶4 000 respectively. The optimal range was from 0.69 to 1 000ng/ml.The assay provided a sensitivity of 0.69ng/ml.The coefficients of variation of intra-assay and inter-assay were 9.10% and 10.33% respectively. The average recovery rate of TAP was 97.70%.Conclusion The indirect competitive enzyme-linked immunosorbent assay is established.

Objective To investigate the effect of EST on function of sphincter of Oddi and on the long-term complications.Methods A total of 68 patients with common bile duct stones were recruited from January 2009 to January 2013 in The First People's Hospital of Hangzhou.All patients were under follow-up to observe the incidence of late complications after EST.Oddi sphincter manometry and bile bacterial culture were performed before EST and one year later.Results With the 13-36 months follow-up,the incidence of late complications was 16.9% (11/65).The peak pressure and frequency of contraction of Oddi sphincter after EST were significantly lower than those before EST,but there were no significant differences between patients with complication and without (P > 0.05).After EST,positive rate of biliary bacteria in patients with complications was much higher than that in patients without complications [70% (7/10)vs.31.7% (13/41),P =0.026],although there were no significant differences between them before EST [54.5%(6/11) vs.46.3% (25/54),P =0.618).Logistic regression analysis showed that cholecystolithiasis,bile duct diameter (> 1.5 cm),number of stone (> 3),maximun diameter of stone (> 20 mm),large EST were independent risk factors for late complications of EST.Conclusion Importance should be attached to the late complications of EST with Oddi sphincter function declination,and biliary enteric reflux being the basic factor in late compilations after EST,which are also influenced by many risk factors.

The effects of positive end-expiratory pressure (PEEP) on pulmonary shunt were studied during gen- eral anesthesia and postoperative period.Twenty cholecystectomy patients were randomly divided into experiment group (group P) and control group (group Z). PEEP and ZEEP were used separately after induction. Objective:To evaluate the application value of diffusion tensor imaging (DTI) in guiding the postoperative radiothera-py plan of the gliomas adjacent to the corticospinal tract (CST). Methods:Thirty patients with gliomas adjacent to the CST underwent routine magnetic resonance imaging (MRI) contrast-enhanced scanning and DTI after radiotherapy. Tractography data sets were ac-quired and were fused with the images of corresponding anatomical MRI and computed tomography. The acquired data sets of radio-therapy planning system were imported to assist with the delineation of the target volume, organs at risk, and CST. Two sets of radio-therapy plan, which considered or did not consider the dose protective effect of the CST, were formulated and compared using the treat-ment technique of intensity modulated radiotherapy. Results:The protective radiotherapy and unprotected plans both achieved the thera-peutic dose to the target volume and the protection of the routine organs at risk. In the protective dose (with an optimization program that considered the dose reduction of CST), the maximum and mean radiation doses suffered by the patients' ipsilateral and contra-later-al CSTs were lower compared with the unprotected plan (P<0.05). Conclusion:DTI can identify the location and shape of CSTs, and their relationship with the postoperative radiotherapy target of gliomas. These findings contribute to the formulation of a protective ra-diotherapeutic regimen to keep the CST from the maximum and the mean radiation doses to the largest extent, thereby decreasing the possibility of nerve damage after radiotherapy.
9.Effect evaluation on employee assistance program (EAP) for medical workers

Objective To evaluate the mental health of medical workers, and develop an EAP suitable for heahhcare sector. Methods A 1-year EAP was implemented in a hospital in Beijing, in which SF-36 and JSS scales were used to study the health-related quality of life and job satisfaction of these workers. Results Work pressure is found to be the main source of pressure for medical workers, accounting for 63.7%. Scores of the group receiving EAP service are found to be significantly higher (P<0.01) in all dimensions (excepting work nature and supervisor dimension) of both SF-36 and JSS. Conclusions Medical workers are exposed to high pressure of work, having strong need for EAP service as result. EAP is an effective means to maintain mental health and improve quality of life and job performance, if applied widely.
10.Expression of PPAR-γ and its relationship with apoptosis in rectal carcinoma

Objective To investigate the expression of PPAR-γ in rectal carcinoma and the relationship between the expression of PPAR-γ and apoptosis in rectal carcinoma. Methods Tissue microarray and immunohistochemistry were used to detect the expression of PPAR-γ gene, bcl-2 and bax in 16 cases of the normal rectal tissue, 16 cases of rectal adenoma and 80 cases of the rectal carcinoma. TUNEL was used to detect of the AI in 80 cases of the rectal carcinoma. Results The positive rate of PPAR-γ expression in normal rectal tissue, rectal adenoma and adenocarcinoma were 6.3%, 25.0% and 71.3% respectively. Clinicopathological analysis showed that the in-crease of PPAR-γ expression were not associated with age, sex and histological classification (P > 0.05), but associated with differentiation grades, pathological stages, lymph nodes metastases and 3-year survival rate (P < 0.05). The expression of PPAR-γwas positively correla-ted with bcl - 2 and bax. AI was negative related with PPAR-γ (r = -0.51, P < 0.05). Conclusions PPAR-γ gene may play a role in the oncogenesis and progression of rectal carcinoma. The abnormal regulation of apoptosis may play an important role in the pathogenesis of rectal carcinoma. Detection of PPAR-γ is of great value for diagnosing colorectal carcinoma, and evaluating malignancy extent and prognosis.
