1.Study on immune mechanism of a recombinant ricin B chain protein against ricin intoxication in BALB/c mice

Objective:To explore the immunity provided to BALB/c mice by immunization with the recombinant ricin B chain protein (rRTB). Methods:Female BALB/c mice were randomly selected into rRTB-vaccinated group and PBS group. Mice were subcu-taneously (s. c. ) injected four times with 14 days immunization time interval. Changes of antibodies (IgG,IgG1and IgG2a) in serum were detected by ELISA. Meanwhile,the expression of IL-4 and IFN-γ were detected by flow cytometry. Results:The mean IgG titers reached 106 after the fourth immunization and a strong secondary response was induced in vaccinated mice when challenged with toxin. There was significant difference between rRTB-vaccinated group and PBS group ( P<0. 05 ) . The same result was shown in IgG1. However,no changed was detected in IgG2a. Meanwhile,there was significant difference for IL-4 between two groups (P<0. 05), while no significant difference for IFN-γwas observed. Conclusion:rRTB can produce higher levels of antigen-specific antibodies ( IgG and IgG1),and cytokine (IL-4) of splenocytes,which means the recombinant protein can induce the Th2-type immune response and trigger a good immune response. rRTB may be a potentially valuable vaccine candidate against human exposure to AT.
2.Mechanism of phloretin induced the apoptosis of hepatoma carcinoma cell SMMC-7721

[Abstract ] Objective To investigate effect of phloretin on apoptotic of hepatoma carcinoma cells SMMC-7721, and explore its mechanisms.Methods Logarithmic phase of hepatoma carcinoma cells SMMC-7721 were cultured separately with 30, 60, 120 mg/L phloretin, morphological alterations of apoptotic were observed by phase contrast microscopy and AO/EB double fluorescence staining method was used to observe were low, medium and high concentration trentment group, respectively.the cells treated by phloretin.Apoptotic rates, cell cycle progression, mitochondrial trans-membrane potential and intracellular calcium homeostasis were detected by flow cytometry.Results Cells appeared typical apoptosis morphological alterations.Phloretin induced SMMC-7721 cell line apoptosis in a dosage and duration dependent manner.Cell cycle was arrested at G1 phase, mitochondrial trans-membrane potential decreased, intracellular free Ca2 +increased.Conclusion Phloretin induce apoptosis of SMMC-7721 by affecting cell cycle progression, reducing mitochondrial trans-membrane potential and changing intracellular calcium homeostasis.
3.Clinical research of chlamydia infection in female genital tract

Objective To investigate the high risk factors of chlamydia (CT) infection in female.Methods Enzyme linked immunosorbent assay was used to detect 120 cases of gynecological clinic,including 60 cases asymptomatic patients before abortion and 60 cases patients with symptoms of mild genital itching,vaginal secretions slight increased.CT antigen in uterine cervix were detected,and the relationship between its symptoms,age,birth time,contraception,cervical condition,the number of sexual partners,mycoplasma and CT infection were analyzed.Results Among 120 cases,there were 28 cases of CT positive,positive rate was 23.3% (28/120).CT infection rate of symptomatic and asymptomatic patients was 16.7%,30.0% respectively,the difference was not significant(P>0.05).The CT infection rate of patients' age younger than 30 years was 27.8%,higher than older than 30 years (10.0%),the difference was significant(x2=3.975,P>0.05).The CT infection rate of patients had multiple sexual partners in nearly lyear was 62.5%,higher than that had one sexual partner(9.0%),the difference was significant(x2=37.42,P>0.05).The CT infection rate of patients didn't use contraceptive tools was 29.1%,higher than that used contraceptive tools(12.2%),the difference was significant(x2=4.319,P<0.05).Conclusion The symptoms of CT infection are no specificity,should be targeted at screening for patients have high-risk factors,such as age less than 30 years old,have multiple sexual partners nearly 1 year and don' t use contraceptive tools.
4.The clinical observation of two methods of hemostasis in the management of hemorrhage in central placenta previa during cesarean section

Objective To discuss and validate which method is more effectiveness for central placenta previa through analysing the clinical outcomes of the management of hemorrhage between uterine packing and stitching hemo-stasis during cesarean section.Methods 48 pregnant women with central placenta previa was conducted in this ret-rospective study,which were all cesarean section delivery.These patients were divided into two groups.24 patients with uterine packing with gauze in the control of massive hemorrhage during cesarean section were indetified,other-wise,the other 24 patients were conducted with sewing and ligation uterine.And then,the effectiveness and outcomes were compared.Results These all patients were alive and keep the uterus.The average estimated blood loss of the stiching group was (554 ±327)mL,obviously less than the uterine packing group,which was (828 ±584)mL,and the difference was statisatically significant (t =6.689,P<0.01 ).The rate of blood transfusion during operation was 8.33% (2/24)in the group of stiching,and was 50.0%(12/24)in the uterine packing group(P<0.05).Similar-ly,the operation time of the stiching hemostasis group,which was (48.9 ±10.0)min,was evidently shorter than (68.3 ±13.3)min of the other group(t=8.126,P<0.01).The two groups had no significant difference in therate of primary healing of incision,postoperative fever and admission time(P>0.05).Conclusion The uterine stiching is an effective,rapid and simple technique in the control of hemorrhage of central placenta previa.
5.Effects of proliferation and apoptosis of phloretin on human gastric cancer cells SGC-7901

Objective To investigate the proliferative and apoptotic effects of phloretin on gastric cancer cell andthe possible mechanisms. Methods SGC-7901 were treated with different concentrations of phloretin(40,80,160 mg/L), and the cell morphological alterations were detected by using Hoechst33258 staining, cell activity were detected by MTT assay, cell apoptosis, cell cycle progression, mitochondrial trans-membrane potential and intracellular calcium homeostasis were detected by flow cytometry.ResuIts After treated with different concentrations of phloretin at different times, SGC-7901 cell showed morphological alterations.Phloretin could inhibite the proliferation of SGC-7901 cell line, and induced its apoptosis in a dosage and duration dependent manner.Cell cycle was arrested at G1 phase, mitochondrial trans-membrane potential dropped, intracellular free Ca2 +increased.ConcIusion phloretin can induce apoptosis of SGC-7901 via arresting cell cycle progression, reducing mitochondrial trans-membrane potential and disturbing intracellular calcium homeostasis.
6.Ultrasonographic Diagnosis of Fetal Craniocerebrum in Second-trimester

Objective To explore the ultrasonic imaging features of fetal craniocerebrum, and improve the recognizition on the normal structure and deformity of fetal craniocerebrum. Methods Fetal cranium was measured during 12~40 weeks of gestation, including biaparietal diameter(BPD),lateral ventricular width(LVW),cerebellomedullary cistern(CM), and nuchal translucency(NT) or nuchal fold thickness(NF). Results 18 cases of abnormal craniocerebrum were identified,nine cases of which were accompanied with other abnormalities, and fourteen of which were found during 16~28 weeks of gestation. All cases were proved by subsequent labor or induced labor. Conclusion Second-trimester is the best period of prenatal ultrasound diagnosis of fetal craniocerebrum.
7.A case report of solitary plasmacytomas of bone located in the lower femur

The original articles in recent years about solitary plasmacytomas of bone(SPB) were reviewed in terms of etiopathogenisis,diagnosis,therapy plan and prognosis.SPB is a rare tumor characterized by the uncontrolled proliferation of plasmocytes.After it is diagnosed by local biopsy and further immunocytochemistry test,the wide excision or thorough curettage in corporation with radiotherapy is the choice of treatment.The primary tumor size,the content and lasting time of M proteins or local recurrence evidence after properly treated are all closely correlated with SPB prognosis.SPB is a rare tumor with a tendency to grow into multiple myeloma.The correct diagnosis and treatmert in time and long-term fellow up are vital factors for prognosis.
8.Effects of indole-3-carbinol on hepatic stellate cells activated by acetaldehyde in precision-cut liver slices

Aim To investigate effects of indole-3-carbinol (I3C) on hepatic stellate cells (HSCs) activated by acetaldehyde in precision-cut liver slices (PCLS). Methods PCLS were incubated with 700 ?mol?L-1 acetaldehyde and 200 ~ 800 ?mol?L-1 I3C for 6 h. The expression of ?-smooth muscle actin(?-SMA) in liver slices was analyzed by immunohistochemistry. The leakages of glutathione S-transferase (GST), lactate dehydrogenase (LDH) and content of transforming growth factor-?1 (TGF-?1) in media were assayed. Contents of malondialdehyde (MDA) and hydroxyproline (Hyp) in tissue were also determined. Expressions of matrix metalloproteinases-1 (MMP-1) and tissue inhibitor of metalloproteinase (TIMP-1) in media were analyzed by the western blot. Results The increase of activated HSCs due to acetaldehyde was inhibited by I3C (200~800 ?mol?L-1). Meanwhile, I3C treatment (200~800 ?mol?L-1) showed significant and concentration-dependent antagonistic actions on the increment of GST, LDH leakages into the media and MDA, Hyp contents in tissues induced by acetaldehyde. The increase of TGF-?1 was also remarkable inversed by I3C (200~800 ?mol?L-1). 9.Effect of total parenteral nutrition with glutamine on the plasma endotoxin in the MODS patients

Objective:To determine the effect of total parenteral nutrition(TPN) supplemented with glutamine(Gln) on the concentration of plasma lipopolysaccharide(LPS)of the multiple organ dysfunction syndrome(MODS) patients.Methods:40 MODS patients in the ICU were randomly divided into routine group(Group A,n=20),the Gln+TPN treatment Group (Group B,n=20).The two groups of patients received treatment of parenteral nutrition with equal nitrogen and equal calorie.Group A were given routine TPN,and Group B were given routine TPN and Gln 0.27 g/(kg?d)[i.e.dipeptiven 0.4 g/(kg?d)] for seven days.The concentration of plasma Gln and LPS were checked before the treatment of TPN and on the first,the third and the seventh day after the treatment of TPN.At the same time 20 healthy blood donators were chosen as the healthy control group(Group C).Results:Before treatment the concentration of plasma Gln of the two patient groups were both lower than that of Group C(P0.05).On the first,the third and the seventh day of the treatment of TPN,the concentration of plasma Gln of Group A was lower than that of Group B(P0.05).After the treatment of Gln plus TPN,the concentration of plasma LPS of Group B was lower than that of Group A(P0.05).Conclusion:The Gln(dipeptiven) can help to decrease the concentration of plasma LPS and the TPN treatment time of the MODS patients.
10.Study on Preparation Technology for Pingfenggujin Granule

Objective To optimize a preparation technique of Pingfenggujin Granule. Methods The technique was determined by the orthogonal design with content of Astragalosides, polysaccharides and the rate of dried extracts. The contents of polysaccharides and Astragalosides were determined by UV and ELSD-HPLC respectively. Results The optimum water-extraction technique was to steep 2.5 h, ten times of water as much as weigh of total drugs and extract three times, 2 h each. Conclusion The optimum technique is feasible, and the method of content determination is accurate and reliable.
