observation group than in the control group at T1 and T2 (P < 0.01). These findings indicate that dexmedetomidine for sedation after hip replacement can lessen inflammatory response and contribute to the recovery of hip function.

Objective To explore the application of bacterial community diversity profiling in forensic comparison of soil.Methods The DNA profilings of bacterial community from 5 soil samples of different source and 4 samples of same source were obtained through the specific amplification of 16S rDNA segments followed by terminal restriction fragment length polymorphism(T-RFLP)analysis,and the similarity indexes between different soil samples were calculated.Result The ranges of similarity indexes between different sources of soil samples were 0.3～0.44,and the ranges between same sources of soil samples were 0.76～0.87.Conclusion There is a great relativity between the bacterial community diversity profiling and the source of soil sample.

Objective The purpose of this paper is to study PGM1 genotyping by PCR RFLP.Method 300 unrelated individuals of Han were genotyped using PCR RFLP. The target amplificaton products of extron 4 and 8 of PGM1 gene were digested by Bgl II and Nla III respectively.The digested DNA fragments were typed by PAGE.Result This PGM1 RFLP system can discriminate 9 genotypes with Dp of 0 7450 in Han population.Compared with conventional PAGIEF, 1+2- and 1-2+ cant be differentiated and the rare genotypes also cant be detected by this method.The advantage of this method was PGM1 genotyping successfully in bloodstains stored for 25 years and with 0 1ng genomic DNA.PGM1 RFLP method is useful for forensic identification.

Objective To survey the significance of specific immune response in the relationship between Sagnation the Liver-qi syndrome and the spleen Deficiency syndrome.Methods The rats with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome were respectively corresponding and crossely treated with Radix Bupleuric dispersing the stagnated Liver-qi powder Decolion of Four Noble Drugs. Their T, B lymphocyte activity and specific immune response indexes changes (such as IgG, IgM in the serum) were respectively observed by lymphocyte transformation test and immunoturbidimetry. Results There were no notable changes about the IgG level among all kinds of groups. The IgM levels in the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome decreased, especially in the Liver-qi Stagnation syndrome (P0 05), Decolion of Four Noble Drugs could increase the IgM level of the patients with the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome (P0 05); The T lymphocyte transformation rate of the Spleen Deficiency syndrome obviously decreased, and Decolion of Four Noble Drugs could recover it to the normal level (P0 05). The rate of B lymphocyte transformation the Liver-qi Stagnation syndrome's also showed the increasing tenderly (P0 05). Conclusions The relationship between the Liver-qi Stagnation syndrome and the Spleen Deficiency syndrome could occur in the phase of antigen presentation, especially in the phase of the T H cell activation stimulated by antigen. Radix Bupleuric dispersing the stagnated Liver-qi powder could enhance T, B lymphocyte activities by reverse the Liver-qi Stagnation syndrome,and Decolion of Four Noble Drugs could enhance T lymphocyte by improving the Spleen Deficiency syndrome.

Objective To establish a new allele specific PCR method for GPT typing: and study GPT polymorphism by FLDAS-PCR and PCR-RFLP. Method 248 unrelated individuals of Han were genotyped using FLDAS-PCR and PCR-RFLP. Results Three genotypes were identified with DP of 0.631 in Han population. Typing results by FLDAS-PCR were completely consistent with those by PCR-RFLP. The allele frequencies were GPT * 1 = 0.5423 and GPT * 2 = 0.4577. The population data fitted the Hardy-Weinberg law. Conclusion GPT-FLDAS-PCR and GPT-RFLP method is useful for forensic identification.

Objective To observe the clinical efficacy of Wenshen Xiaochuan Paste Recipe (WXPR) in preventing bronchial asthma, and to explore the mechanism. Methods Eighty patients were randomly divided into treatment group and control group. Each group had 40 patients. The control group was treated by conventional western medicine such as inhalation of steroid, and the treatment group was given oral use of WXPR based on the treatment for the control group. Treatment for both groups covered 60 days, and the follow-up lasted 10 months. The clinical outcomes included the clinical efficacy and immune indexes of peripheral blood T lymphocyte subsets CD3+, CD4+, CD8+and CD4+/CD8+ratio, and the serum IgE level. Results (1) After treatment and 12 months later, the scores of asthma control test (ACT ) in the two groups were increased obviously as compared with those before treatment, and there were statistically significant differences (P<0.05 or P<0.01) . The differences of ACT scores between the two groups were insignificant after treatment ( P>0.05) , but were significant ( P<0.05) 12 months later. (2) Within one year after treatment, the frequency of acute asthma attack was decreased in both groups (P<0.05 compared with that before treatment), and the treatment group had better effect on decreasing the attack frequency than the control group (P<0.05). (3) After treatment, the serum IgE, CD4+level, and CD4+/CD8+ ratio were decreased significantly, while CD8+ level was increased in the treatment group, and there were statistically significant differences ( P<0.05 compared with those before treatment). However, the differences of immune indexes in the control group were insignificant before and after treatment ( P>0.05). The improvement of immune indexes in the treatment group was superior to that in the control group ( P<0.05). Conclusion WXPR has good clinical efficacy in preventing and treating bronchial asthma. The mechanism is related with the inhibition of airway inflammation and improvement of immune function probably through regulating T lymphocyte subsets and lowering serum IgE level in asthma patients.

Objective To investigate the effect of influenza virus on the expression of collagen triple he-lix repeat containing 1 (CTHRC1). Methods A549 cells were infected with influenza virus. mRNA and protein levels of CTHRC1 were determined by RT-PCR and Western blot , CTHRC1 levels in the cell supernatants and sera of influenza virus infected patients were determined by enzyme-linked immunosorbent assay (ELISA). The difference of CTHRC1 levels between healthy controls and HCV patients was analyzed. Results Compared with controls, mRNA and protein levels of CTHRC1 were higher in A549 cells infected with H3N2. Serum CTHRC1 levels were higher in influenza virus infected patients than in healthy controls (P < 0.05). Conclusion Influenza virus can promote the synthesis and secretion of CTHRC1.

Objective The volatile oil was further purified to get the patchouli alcohol by molecular distillation taking patchouli oil which contained 23.97% of patchouli alcohol as raw material.Methods The best process parameters,including temperature,feed rate,and scraping rate,of molecular distillation technique for separating patchouli oil were studied by orthogonal test.The content was detected by GC under the conditions of elastic quartz capillary column HP-5,detector temperature 280 ℃,gasification temperature 280 ℃,distributary refer 20∶1.The effects of evaporation temperature on content and extraction rate of the patchouli oil had been investigated and the difference between the molecular and vapor distillations was studied.Results Content and extraction rate of patchouli alcohol under the best evaporation temperature 65 ℃ by molecular distillation reached to 40.71% and 76.55%,respectively.Conclusion The molecular distillation technique that could get the product with higher purity and extracting rate than that by vapor distillation in patchouli oil purification has a very good prospect in industry.

Objective To investigate the distribution and drug resistance of pathogens of ventilator-associated pneumonia(VAP) in general intensive care unit(ICU).Methods Statistical methods were used to analyze retrospectively the data of pathogens and drug resistance of VAP in general ICU from 2004 to 2006.Results Totally 347 pathogens were isolated from deep-part secretion of lower respiratory tracts of 302 VAP in general ICU.The main pathogens included Pseudomonas aeruginosa(32.3%),Acinetobacter baumanii(31.1%),Staphylococcus aurous(16.7%),Klebsiella(7.5%) and Coagulase-negative Staphylococci(6.1%).Gram-negative bacillus showed a high resistance to piperacillin and Cefotaxime but high susceptibility to carbapenems and the enzyme inhibitor antibiotics.Gram-positive coccobacteria showed a high resistance to Penicillin,Clindamycin and Erythromycin but high susceptibility to Vancomycin and Quinolone.Conclusion Gram-negative bacillus was the main pathogen of VAP in general ICU,multidrug resistance was serious.The rate of methicillin-resistant staphylococcus aurous(MRSA) and methicillin-resistant coagulase-negative staphylococci(MRCNS) was high,with serious drug resistance.According to the data of the bacteria pathogen and antibiotics susceptibity,we can select antibiotics reasonably to control the infection and delay the emergency of new drug-resistant bacteria.
