OBJECTIVETo study the effect of Shumai Capsule (SMC) on angiogenesis and expression of relevant growth factor in rats with myocardial ischemia (MI).
METHODSModel rats of MI were duplicated and treated with SMC (SMC group), bFGF + calparine (positive control group) and normal saline (model group) respectively. Besides, a sham-operative group was set up and treated with normal saline. The rats were sacrificed in batches at the time after being medicated for 1, 2 and 4 weeks, for determining von Willebrand factor (vWF) and vascular endothelial growth factor (VEGF) protein expression in ischemic myocardium by immuno-histochemical staining, myocardial micro-vessel density (MVD) using digital analysis system, and the gene expression of VEGF by quantitative real-time PCR.
RESULTSCompared with the sham-operative group and the model group, levels of MVD, protein and gene expression of VEGF in the SMC group were higher respectively at three time segments (all P <0.01), but showed insignificant difference to those in the positive control group.
CONCLUSIONSMC could promote angiogenesis in ischemic myocardium of rats, the up-regulation on VEGF mRNA and protein expression might be one of the potential mechanisms of SMC in promoting angiogenesis.

Objective To summarize the experience of establishing the stable rat model of chronic allograft nephropathy. Methods We used Fisher rats as donors and Lewis rats as recipients. After the left kidney of the donor perfused in situ under hypothermic condition, the left renal vein, abdominal aorta and bladder flap of the donor was anastomosed with the left renal vein, renal artery and bladder of the recipient, respectively. The recipients were given cyclosporin oral solution 10 mg/kg every day by gavage for 10 days after transplantation. The blood and urine samples were collected 1 month, 2 months and 4 months after transplantation and renal function and total urine protein were examined. The pathological changes of the renal allograft were observed 2 and 4 months after transplantation. Results Forty-five rats received operation and achievement ratio was 85%. The renal transplantations were finished in 120 ± 20 min. The Scr, BUN, Cycs and total urine protein demonstrated a significant increase one month after transplantation. On the second and fourth month, with the exception of urine protein continued to increase, the other indicators did not change significantly. Two months after transplantation renal pathology demonstrated light to moderate interstitial fibrosis, infiltration of lymphocytes and plasma cells. At 4th month the renal allografts showed extensive interstitial fibrosis, a large number of infiltrating interstitial cells, thickening, hardening, occlusion of glomerular basement membrane, and renal tubular atrophy that were consistent with pathological changes of chronic allograft nephropathy. Conclusion Through adequate surgical training and improvement, and specification for rat nephrectomy, transplantation surgery, and postoperative management in every detail, the model with high success rate and stability can be achieved.

The growth and development of plant is affected by environmental factors that include light, temperature and water. Many cis-elements that are response to these elicitors interact with transcription factors, and regulate the expression of target genes. It concludes by pointing out recent studies of cis-elements and transcription factors in the plant environmental response promoters, such as light, temperature and water inducible promoter. The molecular mechanisms of gene expression regulated by environmental factors was discussed. This is important to study the mechanisms that plant adapt the environment.

In this study, the 5' -flanking proximal region of stress-induced gene encoding betaine aldehyde dehydrogenase was isolated by Adaptor-PCR and TAIL-PCR from halophyte Suaeda liaotungensis. 1993 bp sequence was obtained by sequencing. The transcription start site, which localized at 62 bases upstream of the start ATG, was predicted using TSSP-TCM program. The functional elements were analysed by PLACE programm. The SlBADH gene promoter contains the basic elements: TATA-box, CAAT-box, and stress-induced elements: salt responsed element, cold, dehydration, ABA and frozen responsed elements, WUN responsed elements and HSE. Obtaining the promoter of betaine aldehyde dehydrogenase gene from Suaeda liaotungensis provides a foundation for analyzing the stress-induced promoter elements, studying the relationship between structure and founction of the promoter, and investigating the molecular mechanism of BADH gene regulation.

OBJECTIVETo observe the protective effects of total glycosides Rubus parviflolius (TGRP) on local cerebral ischemic.
METHODThe local cerebral ischemia in rat was made by middle cerebral artery occlusion(MACO). The infraction weight was determined by TTC stain. SOD, MDA, GSH and apoptotis were determined with different method respectively.
RESULTTGRP 20, 10 mg x kg(-1) ig markedly improved the abnormal nervous symptoms, incredsed the SOD, GSH activity and reduced contentes of MDA in brain of MACO rat, TGRP 20 mg x kg(-1) ig significantly decreased the numbers of apoptotic cells in ischemic cortex.
CONCLUSIONTGRP has protective effects against cerebral infraction, and its mechanism may be related to anti-apoptotis and free radical.

OBJECTIVETo explore the effects of total saponins of Rubus parviflolius (TSRP) on brain edema and blood brain barrier in rats.
METHODThe model of local cerebral ischemia was established in rats by reversible inserting a nylon thread into the anterior cerebral artery through the internal carotid artery brain hydrated amount and content change of Evan' s blue (EB) in cortex subjected to 2h middle cererbral artery occlusion (MACO) followed by 6 h, 24 h, 48 h, 72 h reperfusion and effect of TSRP. penetrability of blood brain-barrier (BBB) the index includes brain hydrated amount and penetrability of blood brain-barrier BBB.
RESULTCom- pared with I/R group. Both brain hydrated amount and the EB content decreased significantly in TSRP groups on the 6 h, 24 h, 48 h, 72 h of reperfusion after 2 hour of cerebral ischemia induced by MACO model.
CONCLUSIONTSRP could decrease brain hydrated amount and markedly lower permeability of blood-brain barrier subjected to 2 h MACO followed by 24 h reperfusion, and this may be a mechanism of TSRP alleviating brain edema during I/R.
