Objective To study the disease pathogenesis of systemic lupus erythematosus patients with thrombocytopenia ,and discuss reticulated platelets (RP) in SLE merger thrombocytopenia disease of diagnosis and treatment in the clinical application val-ue .Methods 55 patients diagnosed with SLE patients and 50 cases of healthy check-up ,low platelet count patients of SLE patients treated before and after the inspection by Sysmex XE-5000 automatic blood cell analyzer immature platelet fraction (IPF) and other peripheral blood cell parameters .Results Group of patients with SLE and normal controls of IPF test results respectively was (5 .30 ± 3 .75)% ,(2 .74 ± 1 .05)% ,both comparative difference was statistically significant (P<0 .05);SLE associated with throm-bocytopeniagroupofIPF[(10.14±3.66)% ],platelets(PLT)[(67.2±13.5)×109/L],meanplateletvolume(MPV)[(12.64± 0 .92)fL] ,platelet distribution width(PDW)[(18 .24 ± 1 .70)fL] compared with normal control group and SLE did not reduce plate-let group ,differences had statistical significance(P<0 .05);SLE associated with thrombocytopenia in patients with IPF respectively before and after treatment was (9 .76 ± 1 .82)% and (5 .86 ± 0 .96)% ,both comparative difference was statistically significant (P<0 . 05) .Conclusion The main cause of SLE patients with thrombocytopenia may be associated with peripheral blood platelet destruc-tion ,IPF′s result can reflect the bone marrow platelet production ,Help in the auxiliary diagnosis of disease and prognosis judg-ment .

Objective To observe the intervention effect of acupuncture on mammalian target of rapamycin(mTOR)of hypoxic-ischemic encephalopathy(HIE)model in newborn rats.Methods Seven-day-old SD rats were randomly devided into six groups.Sham-surgery group:the bilateral common carotid arteries were separated,without ligation,and the rats were without hypoxia.HIE model group:the bilateral common carotid arteries were separated and ligated,and the rats were in low oxygen.In the sham-acpuncture group,four positions were acupunctured:from Baihui,Dazhui,Quchi and Yongquan by 1 cm after hypoxia ischemia.In acpuncture group,four positions were acupunctured:Baihui,Dazhui,Quchi and Yongquan after hypoxia ischemia,positioned according to the ordinary acupoints for acupuncture in rats.In the antagonists group,the HIE model rats were injected wortmannin 5 μl to the left ventricle.The acupuncture and antagonists group had the same operations of antagonists group and acupuncture group at the same time.All the groups were subdivided into four subgroups in terms of four time periods of 1 d,3 d,7 d and 21 d.The morphological changes of organizational structures of hypoxic ischemia side in brain tissue were observed by pathological methods.The levels of mTOR mRNA expression were detected by RT-PCR.Results In acupuncture group, acupuncture treatment for 21 d showed that acupuncture group had clearly organizational structure,regular arrangements of neurons,normal cell outline,relatively clear nucleolus,mitigated swell neuron and hyperplas-tic glial cells.Compared with that of the HIE model group,acupuncture treatment increased mTOR mRNA dramatically on day 1 ,day 3,day 7 and day 21 (Q =7.099 1 ,9.009 9,7.894 0,9.914 7,P ﹤0.05 respective-ly).The expression of mTOR mRNA in acupuncture and antagonists group was significantly higher than that in sham-surgery group on day 1 ,day 3 day 7 and day 21 (Q =9.868 9,15.843 4,21.007 2,P ﹤0.05 respec-tively).In day 21 ,compared with the sham operation group and the HIE model group,the expression of mTOR mRNA in acupuncture and antagonists group were significantly higher (Q =18.236 0,4.729 7, P ﹤0.05 respectively).Conclusion Electroacupuncture treatment can promote the expression of mTOR at the mRNA level and protect brain of newborn rats with HIE.

OBJECTIVE:To systematically review the efficacy and safety of lamivudine(LAM)combined with adefovir dipiv-oxil (ADV) versus entecavir (ETV)alone in the treatment of decompensated HBV-associated cirrhosis. METHODS:Retrieved from PubMed,Cochrane Library,CBM,CNKI,VIP and WangFang Database, randomized controlled trails (RCT) about LAM combined with ADV(combination group)versus ETV(control group)in the treatment of decompensated HBV-associated cirrhosis were collected. Meta-analysis was performed by using Stata 11.0 software after data extraction and quality evaluation. RESULTS:Totally 13 RCTs were included,involving 972 patients. Results of Meta-analysis showed,there were no significant differences in the serum alanine transaminase (ALT)[SMD=0.079,95%CI(-0.086,0.244),P=0.348],total bilirubin (TBIL) [SMD=0.056, 95%CI(-0.118,0.230),P=0.529],ALB[SMD=-0.020,95%CI(-0.494,0.454),P=0.935],HBV-DNA negative conversion rate [RR=1.012,95%CI(0.950,1.079),P=0.710],HBV e antigen(HBeAg)seroconversion rate [RR=1.181,95%CI(0.969,1.439), P=0.099],HBeAg negative conversion rate [RR=1.011,95%CI(0.860,1.189),P=0.893],follow-up 96 week liver function Child-Turcotte-Pugh score [SMD=-0.063,95%CI(-0.299,0.173),P=0.601],virological breakthrough rate [RR=1.562,95%CI (0.471,5.178),P=0.466],mortality rate [RR=1.198,95%CI(0.624,2.300),P=0.587] and the incidence of adverse reactions [RR=1.552,95%CI(0.618,3.900),P=0.349]in 2 groups;follow-up 48 week liver function CTP score in combination group was significantly higher than control group,the difference was statistically significant [SMD=0.352,95%CI(0.031,0.672),P=0.031]. CONCLUSIONS:LAM combined with ADV shows similar efficacy and safety to ETV in the treatment of decompensated HBV-as-sociated cirrhosis,but within short term(48 weeks),ETV is more likely to inhibit the development of liver fibrosis of the chronic HBV patients,and even can reverse liver fibrosis and cirrhosis.

Background Zero spherical aberration intraocular lenses(IOL)is designed to prevent the addition of positive spherical aberration after surgery.Research indicated that some positive spherical aberration can provide better depth distance of focus and pseudoaccommodation.Objective The present study was to compare the visual function and wavefront aberrations in pseudophakic eyes with zero spherical aberration IOL and spherical IOL.Methods A prespective case-controlled study was designed.Eighty eyes of 52 patients with age-related cataract were enrolled and divided into two matched groups based on random number table method.The regular phacoemulsification was performed on the eyes,and a zero spherical aberration IOL(Akreos AO)was implanted in the test group and a spherical IOL was used in the control group(Akreos Adapt IOL).The corrected distance visual acuity(CDVA),contrast sensitivity,depth of focus and wavefront aberrations were recorded and compared at 3 months after cataract surgery between these two groups.The trail was approved by the Ethic Committee of Eye Hospital of Wenzhou Medical College,and written informed consent was obtained from each patient prior to the program.Results The clinical demography from the two groups was matched(P ＞ 0.05).There were no significant difference in the CDVA (LogM AR)(-0.03 ±0.08 versus-0.02+0.10)(t =-0.50,P =0.61)and in depth of focus(3.48± 1.07 DS versus 3.20±0.77 DS)(t =1.15,P=0.25)between the zero spherical aberration IOL group and the spherical IOL group.The contrast sensitivities under the mesopic condition at 12.0 c/d and mesopic with glare at 3.0,6.0,18.0 c/d were 12.42 ± 13.16,42.58 ±24.96,30.19± 25.64 and 3.03 ± 5.49 in the zero spherical aberration IOL group,and those in the spherical IOL group were 5.59 ± 8.11,28.74 ± 18.69,17.07 ± 19.35 and 0.22 ± 1.15 without significant differences between these two groups(P＜0.05).Under the 5.0 mm pupil analyzing zone,the spherical aberration in zero spherical aberration IOL group was(0.13 ±0.07)μm,showing a significant reduction in comparison with spherical IOL group(0.21 + 0.07 μm)(P ＜ 0.05).No evidently differences were found in total high-order aberration,coma aberration and trefoil aberration(P＞0.05),but the sphere aberration was considerably lower in the zero spherical aberration IOL group compared with spherical IOL group(t=-4.19,P＝0.00).Conclusions The visual quality of the eyes implanted zero spherical aberration IOL is significantly better than ones implanted with spherical IOL.

Oxidation method with sodium iodide was used to synthesize immunogenic antigen (PF-BSA) and coating antigen (PF-OVA) of paeoniflorin. UV spectroscopy showed that paeoniflorin was successfully conjugated with BSA and OVA. After immunized by PF-BSA, the mice can produce anti-paeoniflorin antibodies specifically. The ELISA test results showed the high titer (1:12 800) and specificity (IC50 = 0.791 mg x L(-1)) of the antiserum from mice injected with PF-BSA. Also, the antiserum showed low cross activities against nine traditional Chinese medicine (TCM) of small molecules. These artificial antigens were successfully synthesized and the anti-paeoniflorin antibody well prepared, which provides the experimental basis for the further study of ELISA and its kit.

OBJECTIVETo seek the special plasma molecule in unstable angina (UA) patients of qi deficiency and blood stasis syndrome (QDBS) and method for explore the proteomic specificity of the disease.

METHODSLC-MS(E) analysis was performed in UA patients of QDBS or non-QDBS and in healthy persons after the 6 proteins with optimal abundance in plasma being removed by polyclonal antibody affinity column (product of Agilent Co. USA).

RESULTSActin were found only expressed, and FN, ApoH and ANXA6 were found highly expressed in plasma of patients with UA-QDBS, suggesting they might be the special molecules for the disease. Moreover, as compared with health persons, SAA, CP, MYH11 and C6 showed high expression, and the 6 proteins, e.g. A1BG, ApoA4, GSN, HBB, HBD and TF, showed low expression in the plasma of UA-QDBS patients.

CONCLUSIONUA-QDBS might belong to a kind inflammatory reaction. There are simultaneous existence of myocardial injury, blood coagulation factor abnormality, lipid metabolic disorder and oxygen transport obstacle in patients of UA-QDBS, they influence and interact mutually. The newly discovered differential proteins might provide clues for studying or discovering new protein targets of angina relieving drugs. The new technique of label free quantitative proteomics is an efficient method for bio-marker research of diseases and syndromes.

Objective To investigate the specific immune responses induced by DNA vaccine com-bined with recombinant adenovirus carrying major outer membrane protein ( MOMP) gene of Chlamydia trachom-atis (Ct) serovar E.Methods Recombinant eukaryotic expression plasmid pVAX 1-MOMP and recombinant adenovirus Ad-MOMP were constructed and purified .Four immunization strategies were designed including DNA immunization (DNA group), recombinant adenovirus immunization (Ad group), DNA prime-recombinant ade-novirus boost regimen ( DNA/Ad group ) and recombinant adenovirus prime-DNA boost regimen ( Ad/DNA group) .Two weeks after the final immunization , vaginal wash specimens , blood samples and spleens were col-lected from all mice for the evaluation of humoral and cellular immune responses .Results Th1 responses and mucosal responses were not found in DNA group .Ad group induced both Th 1 responses and local mucosal re-sponses , and its IgG and IgA levels were significantly higher than those in DNA group .DNA/Ad group and Ad/DNA group induced stronger Th 1 responses and humoral responses than DNA and Ad group .Serum antibodies and mucosal antibodies in DNA/Ad group were higher than those in Ad/DNA group, but Th1 responses in DNA/Ad group were impaired than those in Ad/DNA group.Conclusion The recombinant adenovirus could induce Th1 responses and genital mucosal responses .Combined immunization of DNA/Ad or Ad/DNA showed enhanced specific immune responses in comparison with the single immunization with DNA or Ad .The differ-ences between the two combination methods were that DNA /Ad group stimulated higher levels of specific anti-body responses , while Ad/DNA group produced stronger Th 1 responses .The study provided a reference for the enhancement and development of vaccines against Chlamydia trachomatis in further studies .

Objective To analyze the ophthalmologic characteristics and examinations which caused by vertebral-basilar insuffi- ciency.Design Retrospective case series.Participants 469 eases of vertebral-basilar insufficiency.Methods Retrospective analysis of 469 cases of vertebral-basilar insufficiency patients with ophthalmologic signs' features,incidence,the main cause,color Doppler flow imaging and(or)digital subtraction angiography clearly stenosis location,the degree of stenosis from July 1st 2005 to July ist 2007 in PLA General Hospital.Main Outcome Measures Ophthalmologic signs' related reasons and supplementary examination.Results 96 cases were with the corresponding eye symptoms(20.4%).Vision decreased in 83 cases,diplopia was in 56 cases,ipsilateral hemiopia quadrant blind in 12 cases and depending on deformation in 10 cases,fundus arteriosclerosis in 72 cases,nystagmus in 39 cases,the eye movement disorder in 16 cases.Among 96 patients with associated ocular symptoms,75 patients with color Doppler flow imaging (CDFI)showed unilateral vertebral artery abnormal in 66 cases(88%),among those blood flow velocity decreased in 26 cases,17 cases with vascular plaque stenosis,thinning in 20 cases,3 cases with complete occlusion;different bilateral vertebral artery abnormal in 9 cases(12%),among those velocity dropped in 4 cases,two cases with vascular plaque stenosis and thinning in 3 cases.Forty-five cases with the digital subtraction angiography(DSA),left vertebral artery was abnormal in 24 cases,18 cases with stenosis,occlusion in 5 cas- es,abnormal bending in 1 case.Right vertebral artery was abnormal in 16 cases,13 cases with stenosis,occlusion in 3 cases.Bilateral vertebral artery stenosis was found in 2 cases;vertebral artery stenosis was in 3 patients.Conclusion Vertebral-basilar insufficiency may cause eye-related symptoms,therefore,vision acuity decrease,fundus 'arteriosclerosis,diplopia and nystagmus symptoms are more likely for patients with vertebral-patients with basilar artery insufficiency performance of the eye.(Ophthalmol CHN,2002,16:406-410)

Objective To prepare,purify and validate specific monoclonal antibodies(McAb) against fragment in C-terminal telopeptides of collagen type Ⅱ(EKGPDP)for clinical diagnostics and related research of osteoarthritis(OA).Methods 8 BALB/c mice were immunized with EKGPDP-KLH antigen complex.McAb against EKGPDP fragements were prepared by hybridoma technique.Immunoglobulin classes and subclasses were determined using an Immuno-Type~(TM)mouse McAb isotyping kit.Ascites were producted and McAb were purified by saturation ammonium sulfate(SAS)precipitation and protein G chromatography.Specificity and immunoactivity of antibodies were detected by indirect enzyme-linked imrnunosorbant assay(ELISA).Cartilage specimens were analyzed by immunohistochemical method.Results The hybridoma cells were obtained.10 IgG and 3 IgM single colonies were picked out by limiting dilution and ELISA kit.The titers of McAb in ascites were from 2.8?10~4 to 5.1?10~5.The McAb showed the characteristics of no cross reactions with KLH,BSA,cell culture supernatant,type Ⅰ,Ⅲ collagens and whole type Ⅱ collagen.The method of SAS could get better recovery,immunoactivity of the McAb than protein G chromatograply(t=25.26;P
