Objective To explore the biological characteristics and to compare the different regulation mechanisms of bone marrow mesenchymal stem cells(MSCs)derived from both lupus(MRL/Ipr)and normal (C57BL/6)mice on T iymphoeytes in vitro. Methods MSCs from MRL/lpr and C57BL/6 mice bone marrow were isolated and cultured. The surface phenotypes were detected by flow cytometry(FCM). The morphologi- cal changes of MSCs were evaluated in primary and passage cultures. The growth curves were assayed. CD3~+T lymphocytes from spleen of C57BL/6 mice were isolated by nylon wool columns and stimulated by ConA and co-cultured with or without the two strains of MSCs or supernatant for 72 h. The proliferation of T cells stained by CFSE and activation of T cells were detected. The apoptosis was assessed by now cytometry using Annexin V/PI. Totally 80 rats were randomly divided into the control group, the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) scutellarin( SC) groups and the colchicine ( Col) group. Apart from the blank group, all of the remaining groups were intraperitoneally injected with 0.5 mL pig serum twice every week for consecutively 13 weeks and orally administered with the corresponding drugs since the 9th week. The blank group and the model group were orally given equal volume of normal saline once every for consecutively four weeks. After the experiment, efforts were made to detect the contents of alanine aminotransferase (ALT), aspertate aminotransferase (AST), albumin (ALB), total protein (TP), total bilirubin (TBIL), hyaluronic acid (HA), laminin (LN) and collagen type IV (CIV), collect liver tissues of fixed positions, observe the pathological changes through hematoxylin-eosin (HE) staining, conduct the pathological grading for liver fibrosis, determine the expressions of hepatic collagen type I and III (C I, C III) and calculate their color rendering index. Compared with the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) SC groups could decrease the contents of ALT, AST, TBIL, HA, LN, CIV, increase the contents of ALB, TP in serum and reduce the contents of C I, C III in liver tissues. In conclusion, scutellarin has a certain therapeutic effect on immune liver fibrosis in rats induced by pig serum.

Objective: To explore the symptomatic level of personality disorders,family environment of college students, and the correlation between these two variables. Methods: PDQ-4+(Personality Diagnostic Questionnaire-4+) and AAQ3.1 were administrated to 320 college students by layered sample. Results: Significant correlation was found between several factors of personality disorders and family environment. The personality disorders group get significantly lower score in Intimacy factor and higher score in Conflicts factor of family environment than the normal group. Conclusion: There was significant correlation between symptomatic level of personality disorders and family environment.

Objective To observe the efficacy and safety of pegylated interferon apha-2a combined with ribavirin in the treatment of chronic hepatitis C.Methods One hundred and six patients with chronic hepatitis C were divided into 2 groups randomly.Patients in the observation group were treated with pegylated interferon alpha-2a,and patients in the control group were treated with interferon alpha-1b.All patients were given ribavirin according to the weight,and the treatment course was 48 weeks.HCV-RNA was tested before treatment,4 weeks, 12weeks and 24 weeks after the start of treatment,end of treatment,24 weeks after the end of treatment.The adverse reactions were also observed.Results In the observation group,the rapid virological response (RVR) was 77.4%,the complete early virological response (cEVR) was 83.0%,the end treatment virological response (ETVR) was 90.6%, the sustained virological response (SVR) of 24 weeks after the end of treatment was 84.9%.and these rates were significantly higher than the control group.All patients received the whole course of treatment.Condusion Treatment of chronic hepatitis C with pegylated interferon apha-2a combined with ribavirin is effective and safe.

Objective To explore the expression of angiopoietin-1 (Ang-1) and lung development in neonatal rat with hy-peroxia-induced bronchopulmonary dysplasia (BPD). Methods A total of forty-eight 1-to 3-day-old neonatal rats were random-ly divided into hyperoxia group and control group with 24 rats in each group, fed in high concentration oxygen (≥95%) or in air respectively. At 1st, 3rd and 7th day after high oxygen exposure, the histological changes in lung tissue were observed by HE stai-ning under a light microscope and the expressions of Ang-1 mRNA and its protein in lung tissue were detected by RT-PCR and Western blot. Results With extended exposure to high concentrations of oxygen, rats in hyperoxia group presented such patho-logic change of lung tissue dysplasia as alveolar simplification, reduction in alveolar number and arrested pulmonary microvas-cular development. At 7th day after high oxygen exposure, Ang-1 mRNA and protein expressions in hyperoxia group were (0.33± 0.18) and (0.20±0.07), significantly lower than those [(0.83±0.46) and (0.57±0.44)] in control group (P<0.05). Conclusions Ang-1 plays an important regulatory role in the pulmonary vascular development and participates in the pathogenesis of BPD.

Objective:To identify the effect of ?-synuclein overexpression on mitochondrial membrane structure with atomic force microscopy. Methods:?-syn expression was mediated by AAV (adeno-associated viral vector) and Recombinant AAV/?-syn and AAV/LacZ viral particles were stereotaxically injected in the left side of rat substantia nigra (SN) for rat model of ?-synuclein overexpression. Mitochondria were isolated from rats SN of Brain. Mitochondria were analysis with JC-1 staining,atomic force microscopy and Western blot. Results:By 16 weeks post-infection of AAV-?-syn,the level of ?-syn increased about 2 times in mitochondrial fraction with Western blot and mitochondrial membrane potential (??) decreased with JC-1 staining. Furthermore,mitochondria swelling and porous like structure formed on the mitochondrial membrane with atomic force microscopy. Conclusion:The data suggested that ?-syn could accumulate in mitochondria,might form mitochondrial membrane pores and lead to ?? decreases. ?-syn might lead to mitochondrial dysfunction in Parkinson's disease.

Many Chinese medicinal materials are difficult to be distinguished in characters resulting in easy confusion. In the pa-per, the basic concepts of three groups of easily confused Chinese medicinal materials ( schisandrae chinensis fructus and schisandrae sphenantherae fructus, phellodendri cortex and phellodendri amurensis cortex, akebiae caulis and clematidis armandii caulis) were illus-trated and the development of verification methods were reviewed in the respect of modern instrumental analysis technology. With the improvement of analysis technology, the identification of Chinese medicinal materials relies on experimental data instead of artificial ex-perience, which makes it more objective and accurate.

Objective To study the relation between phlegm-damp retention type of metabolism syndrome and microcosmic indices in order to find out the objective evidence for TCM syndrome differentiation.Methods Totally 49 cases of metabolism syndrome were observed,25 of them were phlegm-damp retention type in differentiation and 24 were non-phlegm-damp retention type.The height, body weight,waistline and blood pressure of two groups were measured,the serum leptin,insulin and blood lipid were determined, and the insulin resistance index(IRI)and body mass index(BMI)were calculated.Results The leptin,BMI,systolic pressure,FINS and IRI of the phlegm-damp retention group were all higher than those of the non-phlegm-damp retention group.Conclusion BMI, leptin,insulin,and IRI may be taken as the the index of syndrome differentiation in metabolism syndrome.

OBJECTIVE: To observe the effect of active component in Cichorii (N3) on lipid metabolism of rat with hypertriglyceridemia complicated by hyperuricemia and hyperglycemia. METHODS: Hypertriglyceridemia complicated by hyperuricemia and hyperglycemia was induced in rats by feeding a kind of feedstuff containing high fructose for 21 days. Then three different doses of N3 were administered continuously to the rats for 35 days. The effects of N3 on lipid metabolism were observed by determining the levels of serum triglyceride (TG), uric acid (UA), glucose (GLU) and free fatty acids (FFA) and the activities of fatty acid synthetase (FAS) and hepatic lipase (HL) from the liver and lipoprotein lipase (LPL) from the heart. RESULTS: Twenty-one days after feeding a high-fructose diet, the levels of serum UA, TG and GLU in the untreated group were significantly higher than those of the normal control group. Thirty-five days after administering N3, there was no statistical difference in serum TG and heart LPL activity between the untreated group and the normal control group, while liver FAS activity, serum FFA level and liver HL activity in the untreated group were significantly higher than those in the normal control group. Different doses of N3 treated rats all showed a decrease of TG, UA and GLU levels. N3 could decrease FAS activity and FFA level, and increase LPL and HL activities significantly. CONCLUSION: N3 in Cichorii may regulate the lipid metabolism of the rats with hypertriglyceridemia complicated by hyperuricemia and hyperglycemia by decreasing FAS activity and FFA level, and increasing LPL and HL activities.
