Hypoplastic myelodysplastic syndrome(Hypo-MDS),with the derangement of BM texture,is characterized by the hypoplasia of bone marrow(BM)accompanied with the deterioration of MDS. Hitherto there isn't a definite therapeutic definition about Hypo-MDS. Is therapy should be paid more attention to. This article introduces the study of therapy of Hypo-MDS.
2.Extracelluar matrix stimulates neurite outgrowth of dorsal root ganglion neurons differently depending on whether extracted from degenerated or normal intervertebral disk
Chinese Journal of Tissue Engineering Research 2014;(7):1039-1044
BACKGROUND:Compared with the normal intervertebral disk, the density of nerve fibers and number of nerve endings and neuropeptides appear to be more in the degenerated intervertebral disk. However, this phenomenon does not occur in the normal y aged disk.
OBJECTIVE:To evaluate the axonal growth and induction of a painful neuropeptide and substance P using rat dorsal root ganglion neurons and degenerated human disc cells in vitro.
METHODS:The human intervertebral discs were harvested from patients with discogenic low back pain and normal people. And extracelluar matrix extracted from human degenerative intervertebral discs was cultured with rat dorsal root ganglion neurons. The promotion of axonal growth and induction of substance P of dorsal root ganglion neurons in extracted medium were evaluated through morphology observation and enzyme-linked immunosorbent assay, respectively.
RESULTS AND CONCLUSION:Compared with the normal group, the content of nerve growth factor in the degenerative group was significantly higher and the average length of neuritis was significantly longer in the experimental group (P<0.05). After intervention with anti-nerve growth factorβ, the average length of neuritis became remarkably shorter. The percentage of substance P-immunoreactive cells was significantly higher in the degenerative group compared with the normal group (P<0.001). Nerve growth factors that highly express in the extracellular matrix from the degenerative intervertebral dick can promote neurite outgrowth of dorsal root ganglion neurons and induce release of neuropeptides related to pain transmission.

Objective To screen for Kaposi sarcoma (KS)-related genes. Methods Tissue samples were obtained from the lesion and normal skin of a patient with KS in Xinjiang Uygur Autonomous Region, and total RNA was extracted from these samples and reverse transcribed into cDNA. Real-time fluorescent quantitative reverse transcription PCR (RT-qPCR) was performed to determine the expression of K8.1, K2 and ORF50 in these samples. The cDNA was labeled with fluorescein and hybridized to a human 35K genome array containing 25 100 genes. Subsequently, the signal images were scanned by a laser scanner and acquired images were analyzed by software. Results RT-qPCR revealed the mRNA expression of K8.1, K2 and ORF50 in the KS tissues but not in the normal skin tissues, indicating that there was no crossed contamination in these specimens. Among the 25 100 genes, 1313 genes were identified to be differentially expressed between KS and normal skin tissues, including 756 up-regulated genes and 557 down-regulated genes. These differentially expressed genes, such as myeloid cell leukemia-1 gene (MCI-1), annexins (ANX) and serine proteinase inhibitor Kazal type 5 (SPINK5), were associated with apoptosis, angiogenesis, cell signaling, protein processing, cell cycle regulation, and so on. Conclusion The differentially expressed genes such as MCI-1 and SPINK5 may be associated with the development of KS.

OBJECTIVETo explore the effects of chronic hypoxia on left and right ventricular function and the expression of cardiac transient receptor potential canonical (TRPC) channels in rats.
METHODSForty eight SD male rats were randomly divided into control group (CON) and chronic hypoxic pulmonary hypertension model group (CH) (n = 24). In CH group, rats were exposed in chronic hypoxia environment (10% +/- 0.2% O2) to induce myocardial hypertrophy. After 3 weeks, mean systemic arterial pressure (mSAP), right ventricular systolic pressure (RVSP), left ventricular systolic pressure (LVSP), left or right ventricular pressure maximum rate of rise (LV/RV + dp/dt(max)), left or right ventricular pressure maximum rate of descent (LV/RV-dp/dt(max)), right ventricular hypertrophy index (RVMI) and left ventricular hypertrophy index (LVMI) were measured. Left and right ventricular myocardium tissue sections were stained by HE and observed under light microscope. Real-time polymerase chain reaction (real-time-PCR) and Western blot were performed to detect the expression of TRPC subfamily.
RESULTSRVSP, RVMI, RV + dp/dt(max) and RV-dp/dt(max) were markedly elevated in CH group (P < 0.01) in comparison to CON group. LVMI was markedly reduced in CH group in comparison to CON group (P < 0.01). LVSP, LV + dp/dt(max) and LV- dp/dt(max) had no significant changes in CH group in comparison to CON group. Right ventricular myocardial cells of CH group became thick, the nuclei stained deeply, the shape of nuclei became not regularity. Left ventricular myocardial fibers did not change significantly. There was significant difference in the levels of mRNA and protein of TRPC1 between CON and CH groups.
CONCLUSIONFor three weeks exposed to chronic hypoxia induced right ventricular hypertrophy specifically, raised the mRNA and protein expression of TRPC1 on right ventricular myocardial cells . TRPC1 might be involved in the development of cardiac hypertrophy.
