1.Application of the number of alleles identical by state in full sib test

Objective Establish and evaluate the method of number of alleles identical by state in full sib test.Methods Sib pair group and unrelated individual pair group were genotyped in 13 STR loci,statistical calculation was carried out.Results Through analysis by discriminant function the posterior probability of membership in each group and error of the posterior probability was obtained.The discriminant function in sib group is L sib =-27.087 03+3.202 32S(S:number of alleles identical by state);The discriminant function in unrelated individual group is:L unrelated =-7.495 63+1.685 09S.The average error of the posterior probability in sib and unrelated pair group is 0.026 5.Conclusion When the number of alleles identical by state is more than 17 or less than 8,posterior probability of a full sib or unrelated individuals is more than 0.998 0 or 0.999 4,respectively.The method based on the number of alleles identical by state in full sib test is credible in identifying sib pair relationship.

4.The prognostic value of modified early warning score of ICU elderly patients in emergency

Objective To study the prognostic value of modified early warning score (MEWS) and Acute Physiology and Chronic Health Evaluation (APACHE Ⅱ) score in emergency ICU elderly patients.Methods 113 elderly patients were collected in emergency ICU,after admission to calculate the MEWS score and APACHE Ⅱ score,the MEWS score and APACHE Ⅱ score difference were compared between death group and survival group.the differences were compared in MEWS score ≤ 3,4 ~6,7 ~ 9,≥ 10 scores groups respectively;analysis of the MEWS score and APACHE Ⅱ the relevance score.the correlation analysis of the MEWS score and APACHE Ⅱ score was given.Results The MEWS(7.91 ± 2.42) point and APACHE Ⅱ (21.9 ± 4.18) point in death group were greater than that in survival group [(4.51 ± 2.14) point,(19.53 ± 4.37) point] (t =7.49,2.70,all P < 0.01).With MEWS score increasing,the fatality rate rose(P <0.01).the MEWS score and APACHE Ⅱ score was correlated(r =0.617,P < 0.01).Conclusion The MEWS score was a simple and quick scoring system,as well as APACHE Ⅱ,it can predict the prognosis of elderly patients in emergency department,it has important application value.
5.Changes in lengths of the wrist ligaments during wrist radioulnar deviation: an in vivo study

Objective To investigate changes in lengths of the wrist ligaments during wrist radioulnar deviation. Methods Six volunteers were included in the study. CT scans was used to detect the changes under different positions of the wrist in the volunteers. The wrists were scanned from the position of radial deviation of 20° to the one of ulnar deviation of 40° at an interval of 20°. The 3-dimensional carpal and distal radioulnar joint structures were reconstructed with customized software and chauges in length of the palmar and dorsal wrist ligaments in radioulnar deviation were computed. Results From wrist radial deviation to neutral position, the lengths of the radioscaphocapitate ligament, the long radiolunate ligament and the dorsal intercarpal ligament increased significantly (P < 0.05), they increased (2.4±0.3) mm, (2.3±0.8) mm,(1.2±0.6) mm, (1.2±1.2)mm and (2.6±1.0) mm respectively. While from wrist ulnar deviation to neutral position, ulnocapitate ligament and the dorsal radiocarpal ligament increased significantly (P < 0.05), they increased (0.8 ±0.6) nm and (1.0±0.5) mm respectively. Conclusion At wrist radial deviation, the radioscaphocapitate, long radiolunate and dorsal intercarpal ligaments are slack. At wrist ulnar deviation, the ulnocapitate ligament and the dorsal radiocarpal ligament are slack. These positions may be favorable to healing of these ligaments.
7.Redution of false positivities of IgM antibodies against hepatitis E virus by a truncated immunodominant polypepfide of HEY open reading frame 2

Objective To exclude false positivities in detection of IgM antibodies against hepatitis E vires of genotype 4 (HEV-4) using a truncated immunodominant polypeptide of HEV open reading frames (ORF2). Methods The recombinant ORF2 immunodominant polypeptide corresponding to amino acids (AA) 459-607 and a truncated polypeptide corresponding to AA 472-607 were separately applied to coat ELISA plates. Anti-HEV IgM from 35 serum samples with HEV RNA positive, 69 serum samples from healthy individuals and 117 clinically suspicious HEV RNA positive serum samples was detected by an indirect ELISA and was confirmed by western blot in protein level and RT-PCR detecting in RNA level. Results Western blot analysis showed that the sera from HEV patients reacted with the dimmer of peptide 459-607, but they didn't react with the monomer and peptide 472-607. The ELISA showed that all 35 serum HEV RNA positive samples reacted with peptide 459-607 but not with peptide 472-607 and none of the 69 serum samples from healthy individuals reacted with either polypeptide. Among 117 chnically suspicious HEV RNA serum samples, 5 samples reacted simultaneously with both polypeptides. But the difference between 450 nm absorbance (A450) value was less than 0. 5. Western blot analysis demonstrated that all the 5 serum samples were anti-HEV IgM- negative. The 5 serum samples was detected negative by RT-PCR, indicating that the false pesitivities were caused by non-specific absorption. Conclusions ORF2 peptide 459-607 may be used to detect anti-HEV lgM efficiently. The false positivities caused by non-specific absorption can be largely excluded according to the difference between 45Ohm absorbance (A450) value when serum reacts with both polypeptides.
8.Effect of quality control cycle on reducing the false negative rate of minimal residual disease of flow cytometry in patients with acute myeloid leukemia

Objective To analyze the application of quality control cycle (QCC) in reducing the false negative rate of minimal residual disease (MRD) of flow cytometry in patients with acute myeloid leukemia (AML). Methods In AML patients with abnormal fusion gene detected in hematology laboratory of Changhai Hospital during the year of 2014, the prevalence of AML-MRD detected both by flow cytometry (FCM) and real-time fluorescence quantitative polymerase chain reaction (FQ-PCR) were analyzed retrospectively. The possible causes of false negative rate of flow cytometric MRD referring to PCR were further deeply analyzed, and the improvement measures were adapted from January 2015 to December 2015 and further judged all according to the QCC methods. Results Pareto diagram showed that the dilution and coagulation of the specimen, the improper analysis strategy and the incomplete combination of the MRD index [composition ratio:83.3 % (60/72)] were the main factors leading to the leakage of FCM MRD in 2014. The QCC group devised measures to reduce the dilution probability of bone marrow and develop a standard operating procedures (SOP) for sampling and testing, strengthen the maintenance of the flow instrument and more importantly, focused on optimizing the antibody panels and gated strategies referring to the current two main kinds of MRD detection combination modes on the basis of the latest advances published in 2015. Finally, the undetected rate of AML-MRD was reduced by FCM from 14.8 % (72/486) in 2014 to 2.6 % (16/620) in 2015. Conclusions The QCC can effectively reduce the leakage rate of flow cytometric AML MRD, improve the ability of laboratory quality control and the ability to solve problems. Solving problems with QCC is thus worthy of being popularized.
9.Clinical Significance of Autoantibodies and Thyroid Function in Patients with Recurrent Spontaneous Abortion

Objective To study the clinical significance of antinuclear antibody (ANA),anti-thyroid peroxidase antibody (TPO-Ab) and antithyroglobulin antibody (Tg-Ab),thyroid stimulating hormone (TSH),free triiodothyronine (FT3),free thyroxine (FT4) in patients with recurrent spontaneous abortion (RSA).Methods 46 women with RSA diagnosis in out patient department in Peking University Shenzhen Hospital from March 2015 to December 2016 were recruited as the study group,20 women with normal childbirth history were enrolled as the control group,ANA was detected by indirect immunofluorescence (IIF) assay and ELISA,TSH,FT3 and FT4 were detected by chemiluminescence immunoassay technology.Results The positive rates of ANA(IIF),TPO-Ab and Tg-Ab in patients with RSA were 24%,24% and 15% respectively which were significantly higher than those in healthy control (P<0.05).Serum levels of TSH,TPO-Ab and Tg-Ab in patients with RSA were 2.70 ± 1.38 mIU/L,38.99 ± 10.18 IU/ml and 3.07 ± 1.69 IU/ml respectively,which were significantly higher than those in healthy cases (P<0.05).Conclusion ANA and antithyroid antibodies were closely related with RSA.It is very important to screen ANA and antithyroid antibodies for etiological diagnosis and appropriate intervention in patients with RSA to reduce the incidence of abortion.
10.EFFECTS OF RGD-POLYPEPTIDE(224) ECHISTATIN AND 17?-ESTRODIOL ON OSTEOCLASTIC BONE RESORPTION ACTIVITIES

Objective This study is to understand the effects of RGD\|peptide(224), Echistatin and 17 ? estrodiol on bone resorption. Methods RGD\|polypeptide (224) (RGD), Echistatin(Ech) and 17 ? estrodiol (E\-2) were added into osteoclast like cells (OLC) and ivory bone slices co\|cultured system. Results It has been found that 10 -7 mol/L RGD, Ech and E\-2 decreased the number of resorption lacunae, resorption area and resorption cave on bone slices in various degrees.Conclusion RGD, Ech and E\-2 inhibits the bone resorption activities in various degrees.
