6.Progresses on Biosynthesis of Long Chain Polyunsaturated Fatty Acids in Plant

There is immense interest in the long-chain polyunsaturated fatty acids(LC-PUFAs) on account of their health-beneficial properties.There has been considerable interest in the production of LC-PUFAs in transgenic plants to provide a cheap,sustainable and clean source of these fatty acids.Several main LC-PUFAs,their roles and metabolic pathway in plant are introducedbriefly,progresses on biosynthesis of LC-PUFAs in transgenic plant are reviewed,and strategies for improving the yields of LC-PUFAs in plant are also discussed.
7.Advances in Application Studies of FAD2 on Biosynthesis of Industrial Fatty Acids

Many unusual fatty acids present in the seed oils of non-food plants are modified at the ?12 position either by the addition of epoxy or hydroxy groups or by the formation of triple (acetylenic) bonds or conjugated double bonds. The introduction of these functionalities into C18 fatty acids is catalyzed by a family of divergent forms of the fatty-acid ?12-desaturase (FAD2) enzyme. Genes encoding these enzymes have now been cloned from a variety of organisms and expressed in transgenic oilseeds. However,the accumulation of the intended fatty acids in transgenic plants has been relatively disappointing,and the mechanisms of these unusual fatty acids biosynthesis and transferring from phosphatidylcholine into storage triacylglycerols still need to be elucidated.
9.Dynamic changes of serum interleukin-18 level in patients with acute cerebral infarction

BACKGROUND: It has been proved that interleukin-18 (IL-18) involves in the development of acute cerebral infarction (ACI) and is positively correlated with the time of brain stroke onset, erythrocyte sedimentation rate (ESR), as well as neurological deficit and low-density value of brain CT.OBJECTIVE: To investigate the dynamic changes of serum IL-18 in the course of ACI.DESIGN: Verification analysis with patients as subjects and healthy volunteers as controls.SETTING: Department of neurology of a municipal hospital.PARTICIPANTS: Totally 46 inpatients (29 males and 17 females) with ACI were randomly selected from the Department of Neurology of People's Hospital, Maoming City, between December 2002 and January 2004.Meanwhile, 40 healthy controls (27 males and 13 females) were recruited from those who came to the hospital for routine physical examination. All the participants signed informed consent.METHODS: Fasting peripheral vein blood of 2 mL was collected from the patients on the 1st, 7rh, 14th and 21st days of ACI onset, and from healthy controls on the day of routine examination. The blood samples were centrifuged at 3 000 r/minute for 15 minutes at 4 ℃, and then the supernatant was collected for detecting IL-18 level by ELISA.MAIN OUTCOME MEASURES: The serum IL-18 level of patients on the 1st, 7th 14th and 21st days of ACI onset and that of healthy controls on the day of routine examination.RFSULTS: Totally 46 patients and 40 healthy controls were enrolled in this study and all data were statistically analyzed. The level of serum IL-18 was significantly higher in the patients with ACI than in normal controls on the 1st and 7th days of onset [(178±41) ng/L, (104±34) ng/L, (65±14) ng/L, P < 0.01],but was similar on the 14th and 21st days [(88±36) ng/L, (72±33) ng/L,(65±14) ng/L, P> 0.05]. The level of serum IL-18 in ACI patients was significantly higher on the 1st day than on the 7th, 14th and 21 st days of the onset (P < 0.05-0.01); moreover, it was also significantly higher on the 7th day than on the 21st day of the onset (P < 0.05).CONCLUSION: The level of IL-18 increases obviously on the 1st day of ACI onset, and gradually decreases with the extended course of disease and time of treatment.
10.The impact of Pan3CSK4 on the platelet activation and the expression of TLR2 on it

Objective To investgate role of TLR2 in the activation, the innate immune and inflammation of human platelets. Methods Human washed platelets were separated from healthy people(n=5) and were stimulated with different concentrations(1μg/ml, 5μg/ml, 10μg/ml) of TLR2 agonistPam3CSK4(a synthetic bacterial lipoproteins). Then the platelet aggregation rate, the expression of CD62p and TLR2 on the platelet surface were measured. Results The platelet aggregation rate were (28.32±5.67)%, (52.56±8.54)% and (76.24±11.23)%, respectively, at concentration of 1μg/ml, 5μg/mland 10μg/ml of Pam3CSK4, more than (12.83±2.43)% at 0μg/ml of it. In addition, the expression of CD62p were (18.45±2.66)%, (22.45±2.04)%, (29.53±4.08)%, respectively at above concentration of Pam3 CSK4, more than (11.20±1.67)% of CD62p at control group(P<0.01). The expression of TLR2 was not significantly increased at a lower concentration of Pam3CSK4(1μg/ml) with (16.85±6.10)% compared with(10.81±3.99)% at the control group. However, it were (21.15±9.90)% and (22.52±9.26)%, respectively, at a higher concentration(5μg/ml, 10μg/ml)more than(10.81±3.99)% at the control group(P<0.05). Conclusion Pam3CSK4 induce aggregation, activation and the up-regulation of TLR2 of platelet by stimulating TLR2 receptor of it. Thereafter, TLR2 play an important role in the innate immune of platelet.
