2.Effect of guanxin no. 2 on gene variant expression profile in rats after myocardial ischemia.

OBJECTIVETo study the effect of Guanxin No. 2 (GX2) on gene variant expression profile in rats after myocardial infarction (MI).
METHODSSix SD rats were equally randomized into the sham operated group, the model group and the GX2 group, and they received gastric perfusion with water and GX2 (10 g/kg) respectively. MI model was established by ligating the left-anterior descending branch of coronary artery after 10 days of perfusion, and rats' myocardial tissue in the junction zone was assessed 24 h later for gene chip detection with DNA microarray. Then a cluster analysis was conducted, and the different expressions of key genes were verified by real-time reverse transcription-polymerase chain reaction.
RESULTSThe up-regulating gene expressions in myocardial tissue in the junction zone increased after ischemia. After GX2 intervention, the up- or down-regulating gene expressions, especially the 2 genes, all-trans-13,14-dihydroretinol saturase (AF465614) and similar to expressed sequence AW413625 (AA799328) decreased significantly. In the common genes, more genes involving activity of signal transducer presented in the model group and the GX2 group and those in the latter showed a certain specificity.
CONCLUSIONGX2 could improve the characteristics of variant gene expression profile in MI rats to a certain extent.

3.Assessment of the Disease Severity by C Reactive Protein in Acute Pancreatitis

Objective To explore the relationship between serum C level of C-reactive protein and the classification of acute pancreatitis, and to provide reference for clinical diagnosis and treatment.Methods From January 2012 to January 2015, 100 cases of acute pancreatitis in our hospital were divided into light acute pancreatitis (MAP) group and severe acute pancreatitis (SAP) group.The levels of serum CRP were detected by 1, 3, 5, 7, and levels were compared with the patients of two groups.The sensitivity and specificity of ROC were compared with CRP curve.Results The serum levels of CRP in patients with MAP group and SAP group were increased and then decreased, and serum CRP in MAP group was lower than that in SAP group.The levels of serum CRP in pancreatic necrosis group were lower than those in pancreatic necrosis group.The sensitivity and specificity of CRP were improved with the increase of CRP levels.Conclusion Serum CRP levels can be used for early typing of acute pancreatitis, and it has a certain clinical reference value.
4.Transcriptome analysis of Aedes aegypti larvae before and after treatment with fipronil

Abstract: Objective In order to explore the application prospects of the phenyl pyrazole insecticide fipronil for mosquito control and identify potential target genes involved in the resistance of Aedes aegypti to fipronil, and lay the foundation for an in-depth study of the resistance mechanism of Aedes aegypti to fipronil. Methods Using Aedes aegypti sensitive strains as experimental materials, Aedes aegypti larvae were treated with fipronil, and the differences in gene expression of Aedes aegypti larvae before and after drug administration were compared at the transcriptome level using transcriptome sequencing combined with bioinformatics analysis, and the differential genes were analyzed. Results A total of 757 differentially expressed genes were identified between the fipronil-treated group and control group, including 217 and 540 up- and down-regulated genes, respectively. Among these, the expression of glutamate-gated chloride channel (GluCls) genes varied significantly before and after treatment. Gene ontology analysis revealed that differentially expressed genes were enriched in catalytic activity, binding, metabolic processes, and membrane-related functions, while KEGG pathway analysis indicated enrichment in biosynthesis, metabolism, and life regulation processes, while the glutathione metabolic pathway was enriched in 15 differentially expressed genes. Conclusions The transcriptome results revealed that GST gene expression was significantly upregulated in fipronil-treated Aedes aegypti larvae, indicating that GST gene is involved in the development of fipronil resistance in Aedes aegypti larvae. In addition, GluCls gene expression was also significantly different before and after treatment, suggesting that GluCls migh be a potential target receptor for fipronil resistance in Aedes aegypti. As GluCls is an ideal target receptor found only in invertebrates, this discovery provides a reference and basis for further exploration of the toxicological mechanism of fipronil on Aedes aegypti.
5.Lignans isolated from stems of Sambucus williamsii and their proliferation effects on UMR106 cells.

The present study aims to investigate the lignan constituents from Sambucus williamsii and their proliferation effects on osteoblast-like UMR106 cells. Seven compounds were isolated and purified by macroporous resin D101, silica gel, Sephadex LH-20, Toyopearl HW-40, ODS column chromatographies and Preparative HPLC(C-18). Their structures were elucidated by spectroscopic methods as threo-guaiacylglycerol-beta-0-4'-conifery ether (1), lirioresinol A (2), 1-hydroxypinoresinol (3), 5-methoxybalanophonin (4), balanophonin (5), 5-methoxy-trans-dihydrodehydrodiconiferyl alcohol (6), and p-hydroxybenzaldehyde (7). Compounds 3-7 were obtained from this genus for the first time. The proliferation effects of all isolated compounds on osteoblast-like UMR106 cells were determined. Compounds 1-7 (1 x 10(-12)-1 x 10(-7) mol x L(-1)) increased UMR106 cell proliferation to some extent.

6.Effect of hepatitis C virus serotype on the response of patients with chronic hepatitis C to interferon treatment.

OBJECTIVETo investigate the influence of hepatitis C virus (HCV) serotype on the interferon (IFN) treatment of patients with chronic hepatitis C.
METHODSNinety-eight patients with chronic hepatitis C were divided into two groups: patients in group 1 (n=53) were treated with Pegasys, 180 ug injected subcutaneously once a week for 24 weeks, and those in group 2 (n=45) were injected with Roferon-A 3 MU three times a week for 24 weeks and then patients in both groups were followed up for another 24 weeks. The virological response at the end of follow up was the primary endpoint for evaluating the effects of IFN treatment. The HCV RNA levels of the chronic hepatitis C patients were determined with COBAS AMPLICOR MONITOR Test, version 2.0, and the HCV serotypes were examined by the means of ELISA using Murex HCV Serotyping 1-6 Assay.
RESULTSOf the 98 cases, HCV in 44 cases was serotype 1, in 23 was serotype 2, in 10 was serotype 3, in 1 was serotype 4, 1 was serotype 5 and in 2 was serotype 6; HCV serotypes in the remaining 17 patients could not determined. In Pegasys treatment group, the biochemical and virological response was not significantly different at the end of treatment between the patients with serotype 1 and non serotype 1 or serotype undetermined patients, but the sustained virological response rate of HCV serotype undetermined group (66.7 percent) was significantly higher than that of serotype 1 patients (27.3 percent) (p=0.035). In Roferon-A treatment group, the virological response rate at 24 weeks and sustained viral response rate at the end of follow-up was not significantly different between serotype 1 and non serotype 1 patients or serotype undetermined patients.
CONCLUSIONAfter the six months treatment course, the HCV serotype had some effects on the treatment response to Pegasys treatment for chronic hepatitic C.

7.Liver histological changes after lamivudine treating in chronic hepatitis B patients with HBeAg positive.

OBJECTIVESTo investigate the histological changes in liver biopsy tissues taken from chronic hepatitis B patients with HBsAg and HBeAg positive and ALT abnormal after lamivudine therapy for one year.
METHODSLamivudine was given orally at the dose of 100 mg once a day for one year. 101 patients were enrolled into this open-label study. Paired liver biopsies from patients with hepatitis B before and after therapy with lamivudine were studied. Blinded biopsies were evaluated by a histopathologist and scored according to Knodell's histology activity index(HAI).
RESULTS53.5% (54/101), 51.5% (52/101) and 31.7% (32/101) patients had a reduction of their total hepatic HAI score, necroinflammation and fibrosis scores by >or=2 points or 1 points at the end of one year of lamivudine therapy, compared with their pretreatment values, respectively. There were significant reduction of HAI score, necroinflammation and fibrosis scores from 8.0+/-4.7 to 5.2+/-3.3 (t=7.358, P<0.01), from 5.9+/-3.8 to 3.6+/-2.5 (t=7.298, P<0.01), and from 2.1+/-1.2 to 1.6+/-1.2 (t=3.827, P<0.01), respectively. The histological improvement was independent on the HBeAg seroconvertion during the therapy.
CONCLUSIONSignificant improvement in liver histology, both necroinflammation and fibrosis, can be obtained in the majority of patients treated with lamivudine for one year.

Pharmaceutical preparations, particularly as a "secret recipe" of traditional Chinese medicine in medical institutions, are the product of China's medical and health industry, and they are also an important means of competing of different medical institutions. Although pharmaceutical preparations have advantages and characteristics than institutes for drug and pharmaceutical companies, the quality standards of pharmaceutical preparations in medical institutions has not reached the desired level over the years. As we all know, the quality of pharmaceutical preparations is important to ensure the efficacy, especially under the environment of people pay more sttention on drug safety and effectiveness and contry increase emphasis on the stste of pharmaceutical preparations. In view of this, we will improve the grade, stability, and clinical efficacy of pharmaceutical preparations by the advanced equipment, testing instruments and the process dynamic quality control technology. Finally, we hope we can provide new ideas for the quality control of pharmaceutical preparations.

9.Spinal cord injuries and repairs: research progress and prospects

Therapeutic intervention and rehabilitation after spinal cord injury (SCI) are currently confronted with tremendous challenges in basic and clinical medicine.The pathological process of SCI starts with the primary injury process directly caused by a mechanical force and follows the secondary injury process caused by microcirculation disorders and inflammatory responses.In this review,we interpret the neurobiological mechanisms underlying SCI.Damage to descending motor pathways and ascending sensory pathways interrupt their feedback and/or feedforward control of movement planning and modulation.To some extent,it also affetcs the normal function of local neural circuits with partial plasticity and independence.The animal models for SCI include transection models,contusion and compression models,ischemia-reperfusion injury and chemical injury models.Basic and translational studies in SCI have achieved remarkable progress in various aspects,including the gene network regulation of axonal regeneration,cell implantation,biomaterials applications,neuronal protection,neurostimulation and functional rehabilitation training,applications of artificial intelligence and brain computer interface.Obviously,future trends in basic and translational studies in SCI research will be enhancement of interdisciplinary research,cutting-edge technology application for clinical rehabilitation,database sharing and data standardization.
10.Radiological findings of the chondroblastomas on the atypical sites of the skeleton system

Objective To review the radiological findings of the chondroblastomas on the atypical sites of the skeleton system.Methods We collected the total image data of 13 patients who were pathologically confirmed the chondroblastomas on the atypical sites of the bone system from the department of orthopedics in shanghai No.6 hospital since 1991.Among all the patients,11 cases were male and others were female.The range of age was 10—50 years and the average age of the patients was 26.2 years old.A retrospective analysis of radiological signs from different diagnostic imaging modalities was made.Results X-ray examination was underdone on all case.On the plain X-ray films,all cases were lytic lesions.The radiolucent lesions were seen in 10 cases,mixed density in 3 cases.10 cases manifested expansible contour. Eleven cases were performed computed tomography(CT)examination.On CT,there were visible calcification in 8 cases,sclerotic margin in 10 cases,internal septation in 4 cases.Soft masses could be seen in 3 cases.Magnetic resonance examination(MRI)was done on 5 cases.On T_1 weighted images(T_1WI), the lesion was hypo and intermediate intense signal and heterogeneous hyperintense signal on T_2 weighed images(T_2WI).The fluid-fluid level and solid-fluid level were seen on 3 cases.On one post-contrast examination,the moderate enhancement was seen on the solid portion of the tumor and however,the obvious enhancement on the septation within the lesion.Conclusion The radiological findings of the chondroblastomas on the atypical sites of the bone system were not suggestive.However,it could display some particular signs of the chondroid tumors such as calcification,septation,etc.To effectively apply the different imaging modalities can be helpful to make a right diagnosis before the operation.
