Objective To investigate the influence of endothelial progenitor cells on the proliferation and differenti-ation of neural stem cells in vitro under the condition of co-culture. Methods We used the mature methods of iso-lation, culture and identification on cells to get the spinal cord NSCs and peripheral blood EPCs from the SD rats, and then put them together in the Transwell chamber in vitro. Set the untreated NSCs as a control group. On the 4th day, we calculated the number and diameter of neurospheres. After that, removing the insert of transwell with EPCs, and inducing the NSCs to differentiate. On the 7th day, we conducted MAP-2 immunofluorescence staining on the cells of induced-differentiation group with fluorescence microscopy, then observed the expression of the anti-gen and calculated the percentage of the differential neurons in the NSCs. All the data were analyzed using statisti-cal method. Results The average number of neurosphere in co-culture group was 10.3±2.2,it was significantly more than that in the control group, and the diameter was under the same situation, the average diameter was up to (49.0±5.3)μm. The percentage of the cells which differentiated into neurons in the co-culture group was signifi-cantly higher than that in the control group, the percentage was(32.1±1.8)%. Conclusion EPCs promote the proliferation of NSCs and induce the differentiation of NSCs into neurons in vitro.

Objective To study and design a new algorithm of QT interval measurement based on multi-scale morphological derivative transform(MMDT) according to the demands of mobile monitoring platform. Methods After the MMDT of ECG signals by inducing a triangle and a short line as a pair of structure elements, they were classified into four types by their morphological characteristics and the optimal strategy for each type was described to detect the onset of Q-wave.By introducing the "wing" function, two referenced points of T peak were picked out, which was helpful to locate the peak and offset of T wave, and improve the precision and recognition rate of MMDT method when detecting the bifid T wave(90.9%)and biphasic T wave(86.7%).Results Eighty data records from CSE database were used to evaluate the availability.By contrast with the wavelet transform method, the statistical results showed that the proposed algorithm had generally less error and smaller standard deviation especially for abnormal-phase T wave. Conclusion Compared with those algorithms based on wavelet transform and self-adaptive threshold techniques, our algorithm needs less empirical parameters and calculation. It is also suitable for mobile monitoring and HOLTER system, and has a wide prospect of application.

Objective: To explore the effect of Plastrum Testudinis on the three subtypes of NOS (nNOS,iNOS,eNOS) expression in rats with focal cerebral ischemia. Methods:Rat model of focal cerebral ischemia was set up by inserting nylon thread into the internal carotid artery . The expressions of nNOS, iNOS and eNOS was detected by immunohistochemical method to observe the effects of Plastrum Testudinis. Results: Expressions of nNOS and iNOS in cerebral cortex and caudate-putamen of Plastrum Testudinis treatment group were significantly lower than those of model control group (P

Flocculent gene FLO1 and its truncated form FLO1c with complete deletion of repeat unit C were expressed in a non-flocculent industrial strain Saccharomyces cerevisiae CE6 to generate recombinant flocculent strains 6-AF1 and 6-AF1c respectively. Both strains of 6-AF1 and 6-AF1c displayed strong flocculation and better cell growth than the control strain CE6-V carrying the empty vector under acetic acid stress. Moreover, the flocculent strains converted glucose to ethanol at much higher rates than the control strain CE6-V under acetic acid stress. In the presence of 0.6% (V/V) acetic acid, the average ethanol production rates of 6-AF1 and 6-AF1c were 1.56 and 1.62 times of that of strain CE6-V, while the ethanol production rates of 6-AF1 and 6-AF1c were 1.21 and 1.78 times of that of strain CE6-V under 1.0% acetic acid stress. Results in this study indicate that acetic acid tolerance and fermentation performance of industrial S. cerevisiae under acetic acid stress can be improved largely by flocculation endowed by expression of flocculent genes, especially FLO1c.

Chinese Journal of Geriatrics 2013;(3):271-274
Objective To evaluate estrogen receptor (ER) α and β expressions in elderly patients with non-small cell lung cancer (NSCLC) in China and their relationship with clinical and pathological characteristics. Methods Expressions of ER α and β were detected by immunohistochemical assay in 147 NSCLC patients over 65 years old,and its relationship with clinical and pathological characteristics was analyzed statistically. Results Both ERα and β expressed in nucleus. The positive expression rate of ER α and β was 4.1% (6/147) and 86.4% (127/147) respectively. There was correlation between ER α expression and the patient age, tumor stage, and pathology differential degree. There was higher ER α expression in patients 65 to 70 years old (8.8 %) compared with patients over 70 years old (0.0%) (x2 =7.267,P=0.007).The ER α expression was higher in patients in stage Ⅰ (9.4%) than that in later stage (0.0%) (x2=8.112,P=0.004),and also higher in patients with pathological high degree (14.3%) than that with pathological low degree (0.0%) (x2=7.820,P=0.005).ER β expression was associated with tumor stage, histology type and pathology differential degree.ER β expression was higher in patients in stage Ⅰ (76.6 %,x2 =9.322,P=0.002),much stronger in adenocarcinoma (71.0%,x2 =4.626,P=0.031),and in pathological high degree patients (82.1%,x2 =7.092,P =0.008).Conclusions ER α and β expressions correlate with clinical and pathological characteristics in elderly NSCLC patients. It indicates that expressions of ERα and β might play an important role in the occurrence and development of NSCLC,and might be a new therapy target in NSCLC.

OBJECTIVE To detect the condition of bacteria of the air in wards of our hospital in order to reduce the possibility of air spread. METHODS Ten tuberculosis wards were chosen at random for study.Detected the bacteria content of the air in wards used before and after respectively and analysed the results. RESULTS The average of backgroud bacteria was 164 CFU/m 3 before the wards were used. Two years later, the average of bacteria increased to 682 CFU/m 3.The ratio was 1∶3:2 on average. There were significant differences between them. CONCLUSIONS Humen are the main facters that make the air polluted in these wards. The bacteria content will decline by good environment cleaning sanitation, ventilation and strengthening steriling management.Infection will decline in the hospital.

Aim To investigate the changes of phosphodiesterase4(PDE4,type 4 cAMP-specific PDE) activity,TNF-? and neutrophil recruitment in experimental rat lung injury(ALI).Methods ALI in the rat was induced by lipopolysacdharide(LPS).PDE4 activity was measured with HPLC,and the level of TNF-? was detected with ELISA,neutrophil infiltration in bronchoalveolar lavage fluid(BALF) and lung tissues was detected by cell count and morphological analysis.Result Lung tissue PDE4 activity significantly increased as early as 1 h,peaked 6 h,and then markedly lowered at 24 h after intratracheal administration of LPS,while there was a same time-course change of total white cell and neutrophil in the BALF(r=0.83,P
