A modified dye test with microplate was to be established to detect Toxoplasma antibodies with cell-cultured Toxoplasma gondii. Numbers of stained and unstained tachyzoites were estimated in every 100 tachyzoites in each well after dyeing with methylene blue. The dilution with 50% tachyzoites stained was used as final dilution. Better results of the microplate dye test has been received when the concentration of tachyzoites in suspension reaches 109/ml with 1% sodium citrate as accessory factor.

Objective:To explore the way to separate and culture eutopic and ectopic endometrial glandular cells and their stromal cells, providing an in vitro cell model of endometriosis for study of its mechanism. Methods:Digestion, filtration and sedimentation were used to isolate and culture the eutopic and ectopic endometrial glandular cells and their stromal cells. The estrogen level was imatated to study the way of promoting cell growth. Morphological characters of eutopic and ectopic endometrial cells were examined using optical microscope. Results:The success rate of separation and culture of normal control endometrial glandular cells and its stromal cells was 91.7%(11/12);of eutopic endometrial glandular cells and its stromal cells of endometriosis was 93.8%(15/16);of etopic endometrial glandular cells and its stromal cells of endometriosis was 75.0%(12/16) . Conclusion:The cultured eutopic and ectopic endometrial cells is more like human body feature than the endometriosis model of animals. So the isolation and culture of eutopic and ectopic endometrial glandular cells and their stromal cells may serve as an in vitro experimental model.

Objective To investigate association between human leukocyte antigen DQB1 (HLADQB1 ) gene polymorphisms and bronchial asthma among Mongolian and Han nationalities. Methods Sequence-specific primer polymerase chain reaction (PCR-SSP) was used to detect frequencies of HLA DQB1 genotypes and alleles in 50 cases of Han and 68 Mongolian asthmatic patients, and 50 Han and 54 Mongolian healthy controls, respectively. Difference in gene frequencies between the two nationalities was estimated by odds ratio (OR) and chi-square test. Results Frequency of the HLA-DQB1 0602 allele was significantly higher in Han patients with bronchial asthma than that in healthy Han nationality (OR = 6.163,P <0.01 ). Frequency of the HLA-DQB1 0603/0608 allele decreased in Mongolian asthmatic patients, as compared to that in healthy Mongolians ( OR = 0.199, P < 0.05 ). Frequency of the HLA-DQB1 0301/4 allele was significantly higher in Mongolian asthmatic patients as compared to that in healthy Mongolians ( OR =2.074,P <0.05). Frequency of the HLA-DQB1 0301/4 allele was significantly higher in Mongolian than that in Han asthmatic patients ( OR = 2.482 ,P =0.05). Frequency of the HLA- DQB1 0602 allele was significantly higher in healthy Mongolians than that in healthy Han nationality ( OR = 3.341, P < 0.05 ), in contrast, frequency of the HLA-DQB1 0402 allele was significantly lower in healthy Mongolians than that in healthy Han nationality ( OR = 0.209, P < 0.05 ). Conclusions The HLA-DQB1 0603/0608 allele is possibly a protective gene and the HLA-DQB1 0301/4 allele a susceptible gene for bronchial asthma in Mongolians, and the HLA-DQB1 0602 allele is possibly a susceptible gene for bronchial asthma in Han nationlity.

Objective:
To analyze the epidemiological features of death comorbid with chronic kidney disease in Ningbo, and to provide reference for chronic kidney disease prevention and control.
Methods:
The information about death comorbid with chronic kidney disease in Ningbo from 2002 to 2017 was collected. The crude mortality, standardized mortality and the proportions of primary death causes were calculated. The annual percentage change(APC)was used to evaluate the trends of the mortality comorbid with chronic kidney disease.
Results:
From 2002 to 2017,the average annual crude mortality rate and standardized mortality rate of Ningbo residents comorbid with chronic kidney disease were 13.31/100 000 and 9.89/100 000,respectively. The crude mortality rate was 14.25/100 000 in males, which was higher than 12.37/100 000 in females(P<0.05). The crude mortality rate was 14.88/100 000 in urban residents, which was higher than 12.32/100 000 in rural residents(P<0.05). The crude mortality rates of Ningbo residents comorbid with chronic kidney disease from 2002 to 2017 showed an increasing trend and increased with age(P<0.05). The primary causes of death comorbid with chronic kidney disease were urogenital diseases, endocrine nutritional and metabolic diseases, circulatory diseases, tumors and infectious diseases or parasitic diseases, accounting for 44.45%,24.32%,13.51%,6.50% and 3.97%,respectively.
Conclusion
The crude mortality rates of Ningbo residents comorbid with chronic kidney disease from 2002 to 2017 showed an upward trend. Men,the elderly and the urban residents were at high risk. The death comorbid with chronic kidney disease mainly attributed to urogenital disease.
