2.Study on effect of scutellarin in resisting liver fibrosis in rats.

Totally 80 rats were randomly divided into the control group, the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) scutellarin( SC) groups and the colchicine ( Col) group. Apart from the blank group, all of the remaining groups were intraperitoneally injected with 0.5 mL pig serum twice every week for consecutively 13 weeks and orally administered with the corresponding drugs since the 9th week. The blank group and the model group were orally given equal volume of normal saline once every for consecutively four weeks. After the experiment, efforts were made to detect the contents of alanine aminotransferase (ALT), aspertate aminotransferase (AST), albumin (ALB), total protein (TP), total bilirubin (TBIL), hyaluronic acid (HA), laminin (LN) and collagen type IV (CIV), collect liver tissues of fixed positions, observe the pathological changes through hematoxylin-eosin (HE) staining, conduct the pathological grading for liver fibrosis, determine the expressions of hepatic collagen type I and III (C I, C III) and calculate their color rendering index. Compared with the model group, low, middle and high dose (25, 50, 100 mg x kg(-1)) SC groups could decrease the contents of ALT, AST, TBIL, HA, LN, CIV, increase the contents of ALB, TP in serum and reduce the contents of C I, C III in liver tissues. In conclusion, scutellarin has a certain therapeutic effect on immune liver fibrosis in rats induced by pig serum.

3.Intervention of Tert-butyl Hydroperoxide-Induced Injury in Human Umbilical Cord Derived Mesenchymal Stem Cells with Ginkgo Biloba Extract

Objective To observe the intervention effect of Ginkgo biloba extract EGb761 on tert-butyl hydroperoxide ( t-BHP)-induced injury in human umbilical cord-derived mesenchymal stem cells ( hUC-MSCs) . Methods Proliferation of hUC-MSCs after primary culture was detected by methyl thiazolyl tetrazolium (MTT) assay, and then the optimal concentrations of t-BHP and EGb761 for oxidative stress injured MSC model were screened. Cell apoptosis was assessed by flow cytometry after Annexin V-fluorescein isothiocyanatel propidium iodide ( Annexin V-FITC/PI) staining. Content of malondialdehyde ( MDA) and superoxide dismutase (SOD) activity in hUC-MSCs were evaluated, and the expression levels of p53 and p21Cip1/Waf1 were analyzed by real-time fluorescence PCR. Results Pretreatment with 10~200 mg/L of EGb761 for 3 hours reduced the sensitivity of hUC-MSCs to t-BHP ( 100 μmol/L) induced proliferation inhibition, while EGb761 over 100 mg/L had no significant effect on enhancing the protection of hUC-MSCs . EGb761 at 100 mg/L prohibited hUC-MSCs apoptosis and MDA accumulation in hUC-MSCs induced by 100μmol/L of t-BHP acting for 6 hours, maintained the enzymatic activity of SOD, and decreased the expression of p53 and p21Cip1/Waf1 in hUC-MSCs with t-BHP-induced injury. Conclusion EGb761 is capable of protecting hUC-MSCs against oxidative stress injury, and its mechanism is probably related with the modulation of p53/p21 signal pathway.
4.Studies on germination of dormant bud and seed of Elaeagnus mollis under tisssue culturing

Object To study the factors causing the dormancy of seed and to broaden the scope in the choice of explant for the tissue culture of the rare and near extinct plant Elaeagnus mollis Diels.Methods Dormant buds from one-year old branch collected each month from Autumn to next Spring, and seed of the very year were selected and cultured on basic media with the addition of 6-BA, NAA and GA 3 Results The germination rate of dormant bud follows the regular order of the months when it was taken. Those taken in Autumn declined every month until next Spring, and the nrevived monthly with those taken in March showing the most prosperous germination.The dormant bud can germinate into shoots. When seed was cultured, there are several factors leading to its dormancy: hardness of the mesocarp, keratinization of periderm, refractory to water permeation and the presence of certain inhibitory subtances. Seed kernal is rich in nutrition, but susceptible to bacterial infection leading to its decay. But the germination rate was well over 70% which can be cultured to give bacteria free shoots. Conclusion Both dormant buds obtained in March and bacteria free shoots germinated from naked kernal can be used as new sources for the tissue culture of E mollis .
5.Clinical study on antibiotic-induced endotoxin release in Gram-negative bacterial infection

Objective To observe the various influence of Imipenem/Cilastatin(IPM) and Cefoperazone(CPZ) on Gram-negative bacterial endotoxin release in severely burned patients.Methods Patients were divided into IPM group and CPZ group randomly. The bacterial cultures and microbial drug-sensitivities of burn wound swabs and subeschar tissues were performed on the 1st and 3rd day, At the same time, the levels of LPS in plasma were detected in seven burned patients administrated with IPM and six with CPZ at the time 0?2?4?12?24?48?72h of antibiotics administration.Results Patients were infected by Gram-negative bacterial. The counting of bacteria in subeschar tissues decreased 10~100 times after the administration of antibiotics 3 days; Plasma LPS levels elevated to the peak value at 2h after infusion of IPM or CPZ , and then showed significant decrease. The magnitude of increased LPS in CPZ group was higher than that in IPM group; The LPS levels of 72h were lower significantly than that of 0h.Conclusion IPM and CPZ could effectively control Gram-negative bacterial infection and induce the releases of LPS from bacteria, of which the former induced less LPS release than the latter; The levels of LPS came down with the decreasing of bacterial counting.
7.A Control Study of N-acetylcysteine Therapy for Chronic Obstructive Pulmonary Disease

Objective NAC therapy was used to observe the effect of TNF-?,hs-CRP,PaO2,PaCO2and lung function in patients with COPD,so as to evaluate the therapeutic effect. Methods 65 patients with COPD were recruited and the patients were randomly divided into 2 treatment groups:control group (n=31) and experimental group (n=34). Patients in the two groups received the conventional treatment of COPD in unchanged dosage for the duration of the study except stopping using antibotic drug after infection was resolved. NAC tablets were taken orally at a dose of 600mg once a day in experimental group. After 8 weeks treatment,we appraised lung function and the plasma level of TNF-?,hs-CRP,PaO2 and PaCO2. Results Compared with normal controls,the plasma levels of TNF-? and hs-CRP increased significantly. 8.Effects of Different Doses of Diphtheria Toxin on the Cochlear Structure and AuditoryFunction in Wildtype Auditory-Mature Mice

Objective To investigate the effects of different doses of diphtheria toxin on cochlear structure and auditory function of adult wildtype mice.Methods The auditory-mature wild type C57BL/6J mice 4 weeks old were randomly devided into 50 ng/g group, 100 ng/g group and control group.C57BL/6J mice in the 50 ng/g or 100 ng/g group were injected 50 ng/g or 100 ng/g diphtheria toxin intraperitoneally for one time, respectively, and the control mice were injected equal volume of normal saline for one time.Then we investigated the ABR threshold change and morphological change of inner and outer hair cell and spiral ganglion neuron 7 days after the injection.Results At 7 day post diphtheria toxin injection compared with those of in control group, in the 50 ng/g group, there was no threshold elevation across frequencies(8 kHz ABR threshold was 20.0±3.78 and 20.83±2.04 dB SPL for 50 ng/g and control respectively), and no loss of inner and outer hair cells (for both groups, the HC loss rates were 0.3%~1%) or SGN (the SGN density was 39.45±3.65, 41.03±3.73/105 μm2, in 50 ng/g and control, respectively).However, the 100 ng/g group, compared with those of in control group, the ABR threshold (8 kHz ABR threshold was 63.0±4.47 dB SPL, respectively)was significantly elevated across each frequency(t=19.62,P<0.001), and there was significant loss of outer hair cell (the loss rate of IHC and OHC was 0.5%±0.1%, 10.7%±0.3%, respectively), which was 10% loss in the apical, middle and basal turn(t=42.219,P<0.001).And the loss of spiral ganglion neuron (the SGN density was 25.55±3.66/105 μm2) was 38%, which was significantly different from the control(t=10.985,P<0.001).Conclusion High dose injection of diphtheria toxin can cause loss of outer hair cell and spiral ganglion neuron in wild type auditory-mature C57BL/6J mice.
9.Evaluation on problem-based social practice improving preventive medicine students' tobacco control capacity

Problem-based social practice is organically combined with health education lessons,to improve tobacco control capacity of preventive medicine students,through study in class and social practice after class.The students' awareness rate of tobacco regulations,support rate of smoke-free school,rejecting smoking rate,and lifetime non-smokers rate have significantly increased ( 40%~90% ) after intervention.The teaching method is adult learning model,and it has effectively enhanced preventive medical students' tobacco control capacity.
10.Comparative study on health risk behaviors between male and female freshmen among medical universities, scientific and technologic universities

ObjectiveTo explore the distribution and characteristics of health risk behaviors between the male and female college freshmen in the medicine and science,and to provide evidence for appropriate health intervention measures.MethodsA questionnaire investigation was performed on college students'health risk behaviors among 1019 freshmen from medical and science college in urban area of Harbin.Results (1)There were statistically significant among the different gender in difference of risky behaviors except playing video games,net surfing,considered suicide,drug abuse and a few activities outside the 60 minutes every day (P < 0.01 ).(2)Medical male risk behavior of the frequency(56.4%,19.2%,12.7% and 47.1% )higher than that of science and engineering male(35.6%,5.2%,1.6% and 17.3% )in trying to smoking,current smoking,sexual behavior and breakfast skipping.Recent medical males drinking rate was 85.3% higher than male of science and engineering 72.2%.The incidence of female science and engineering students,respectively( 89.3%,50.8% and 38.5% ) than female medical students (72.4%,36.9% and 8.0% ) in trying to drinking and recent drinking and breakfast skipping.ConclusionThe risk behaviors of college freshmen were prevalent.The frequency of risk behaviors are different among the College Freshmen in different genders and with different major,respectively.Therefore,it is necessary to take measures for intervention.
