strains that could produce raw starch-digesting glucoamylase were isolated from soil and mildewed rice.The highest raw starch-digesting glucoamylase activity strain named OR-1 was identified as Rhizopus.sp.The raw starch-digesting glucoamylase activity of the strain is 90U/mL.Through UV and NTG mutagenesis,the raw starch-digesting glucoamylase activity raised to 200U/mL and 325U/mL respectively.The RDA were 70% and 65% respectively.
2.BML-111 attenuats acute lung injury induced by intestine ischemia-reperfusion via inhibiting p38 MAPK/NF-κB signaling pathway

Objective This study aims to investigate the effect of Lipoxin A4 receptor on acute lung injury (ALI) induced by intestine ischemia-reperfusion (IIR). Methods Thirty-two 8-week old SD rats were randomly divided into four groups: sham, intestine ischemia-reperfusion (IIR), IIR + BML111 (BML-111), Boc-2 + IIR +BML111 (Boc-2). BML-111 (1 mg/kg) was given intraperitoneally at the onset of reperfusion in the BML-111 and the Boc-2 group. Boc-2 (50 μg/kg) was given intraperitoneally after anesthesia in the Boc-2 group. Rats were subjected to superior mesenteric artery occlusion consisting of 45-min ischemia and 6-h reperfusion, and the sham laparotomy was served as controls. The lung pathology was assayed by the H&E staining. Lung water content was detected using dry/wet ratio. Concentrations of TNF-α, IL-1β, and IL-6 in lung tissue were determined by ELISA. The protein expression of p38 MAPK and NF-κB of lung was assayed by western blot. Results IIR induced serious ALI, with poor lung pathology and increased lung water content, elevation of TNF-α, IL-1β, and IL-6 levels in lung, accompanied with activation of p38 MAPK/NF-κB pathway. However, BML-111 could inhibit the activation of p38 MAPK/NF-κB pathway, leading to the reductions of TNF-α, IL-1β, and IL-6 in lung and attenuation of IIR-induced ALI. Conclusion BML-111 treatment could attenuate inflammation in lung after IIR injury via inactivating the p38 MAPK/NF-κB signaling pathway.
3.Epidemiology of enterovirus 71 infection in children with hand, foot, and mouth disease in Shanghai, 2010-2011

ObjectiveTo understand the clinical epidemiology of enterovirus 71 (EV71) in children with hand,foot,and mouth disease (HFMD) in Shanghai during 2010 to 2011.Methods The demographic,etiological and clinical data of children with HFMD were analyzed retrospectively.EV71 was tested in stool samples by one-step quantitative reverse transcription-polymerase chain reaction (RT-PCR).The date were analyzed by Chi-square test.ResultsEV71 was detected in 820 (54.45%) of 1506 inpatients in 2010 and in 924 (59.84%) of 1544 inpatients in 2011,respectively.The detection rates in severe cases and uncomplicated cases were 86.31% and 46.67% (x2 =247.146,P<0.01) in 2010 and 88.78% and 48.75% (x2 =201.664,P<0.01) in 2011,respectively.The detection rate of EV71 was the highest (60%- 67 %) during May and June when HFMD peaked.Among 1744 EV71-infected HFMD inpatients,the male-to-female ratio was 1.78 ∶ 1 ; the proportion of cases was the lowest in infant <6 months of age (0.46%,8/1744),and the highest in children 1 years of age (34.92%,609/1744); children aged 1-3 years accounted for 76.78% (1339/1744);and migrant children accounted for 72.76 % (1269/1744).The demographic characteristics in severe cases were similar to those in general EV71-infected children.Nine severe cases of pulmonary edema/hemorrhage were all infected with EV71.Conclusions EV71 was a major pathogen causing the outbreaks of HFMD and severe complications in Shanghai in 2010 and 2011.Most severe cases and all critically severe cases were associated with EV71 infection.
4.Study on retreatment of CHC patients with initial treatment failure

Objective To explore the retreatment of CHC patients with initial treatment failure and how to achieve SVR.Methods 54 patients who had experienced treatment failure were enrolled and retreated with standard treatment of pegylated interferon and ribavirin or intensive treatment,respectively.Their SVR rates were statistically compared,to decide two therapies' application.Results 54 patients had been retreated,and total SVR rate was up to 75.92%,with 88.46% in relapsed patients and 64.29% in non-responders.After retreatment with pegylated interferon and ribavirin,SVR rate was 95.45% in patients with prior interferon monotherapy,and 64.71% in patients with prior interferon and ribavirin,and 60% in patients with prior pegylated interferonα-2a monotherapy.SVR rate of relapsed patients was significantly higher than that of non-responders.Conclusions In CHC patients with treatment failure,SVR rate of retreatment with standard treatment or intensive treatment still can be up to 60%-90%.Retreatment with standard therapy can be applied to patients who had received interferon monotherapy or interferon plus ribavirin.Three types of patients who need intensive retreatment were as following:patients nonresponsive to interferon plus ribavirin or pegylated interferon α-2a monotherapy,and patients with treatment failure who had received prior standard treatment.

6.Comparison of effects of kaempferide and anhydroicaritin on biomineralization of cultured osteoblasts.

This study is to compare the effects of kaempferide and anhydroicaritin on biomineralization of rat osteoblasts (ROB) in vitro. Calvarias were dissected aseptically from newborn SD rats, the osteoblasts were obtained by enzyme digestion and were cultured in MEM containing 10% FBS. The medium was changed every three days, and serial subculture was performed when cells covered with 90% of the dish. Kaempferide and anhydroicaritin were separately added with final concentrations of 1 x 10(-4), 1 x 10(-5), 1 x 10(-6) and 1 x 10(-7) mol x L(-1) under the conditions of osteogenic differentiation. The proliferation was measured by MTT, and the optimal concentration was detected by the ALP activity at the 9th day after osteogenic induction culture. The osteogenic indexes of kaempferide, anhydroicaritin and control group with the optimal concentration were compared. The result showed that the anhydroicaritin at concentration of 1 x 10(-5) mol x L(-1) had significantly promoted the activity of ALP, calcium content and osteocalcin content, increased the number of CFU-F(ALP) and mineralized nodules, enhanced the mRNA level of BMP-2, OSX and Runx-2, which are key genes of osteogenic differentiation, and raised the protein content of collagen-I. However, the kaempferide group had not significantly represented the ability that promoted osteogenic differentiation of ROB. The difference of osteogenic differentiation on ROB between kaempferide and anhydroicaritin was caused by the prenyl group on C-8 of icariin.

7.Dose distribution prediction of breast-conserving postoperative intensity-modulated radiotherapy for breast cancer based on deep learning

Objective:To develop the method based on deep learning to predict the dose distribution of breast-conserving postoperative intensity-modulated radiotherapy(IMRT) for breast cancer, and to evaluate accuracy of the prediction model.Methods:The data of 110 left-sided breast-conserving postoperative IMRT for breast cancer patients were reviewed, among them, 80 cases were randomly selected for training set, 10 cases for validation set and the remaining 20 cases were used as test set.Firstly, the four-channel characteristics of the patients′ computed tomography(CT) images, regions of interest, distances between voxel and planning target volume(PTV), and corresponding dose distributions were taken as input data.The established U-Net was used for training and obtaining prediction model which was utilized to perform dose prediction on the test set, in order to verify the influence of the features of distance between voxel and PTV in dose prediction, and to compare the dose prediction result with the actual manual planned dose.Results:By incorporating the features of distance between voxel and PTV, the model achieved higher accuracy in predicting the dose distribution.The dose scores and dose volume histogram(DVH) scores of the testing set, consisting of 20 patients, were 2.10±0.18 and 2.28±0.08, respectively, and the predicted dose distribution was closer to the manually planned distribution( t=2.52, 2.40, P<0.05). The deviation between the predicted doses of the PTV and the organ at risk (OAR) and the manually planned doses were within 4%, the average dose to the contralateral breast was increased by 13 cGy, all of them within the clinically acceptable range. Except for the statistically significant differences in D2, D98( Di represents the dose received by i%of the PTV volume), Dmean(mean dose) of PTV 60 and V5( Vi was the volume percentage of OAR receiving i Gy dose.), Dmeanof the ipsilateral lung ( t=3.74, 2.91, 2.99, 3.47, 2.29, P < 0.05), there were no statistically significant differences in other parameters. Conclusions:The deep learning-based method can accurately predict the dose distribution of breast-conserving postoperative IMRT for breast cancer, and it has been proven through experiments that by incorporating the features of distance between voxel and PTV can effectively improve the prediction accuracy, which helps physicists to improve the quality and consistency of treatment planning.
8.Grape seed extract inhibits the growth of prostate cancer PC-3 cells.

OBJECTIVETo investigate the inhibitory effect of grape seed extract (GSE) on the growth of prostate cancer PC-3 cells.
METHODSPC-3 cells were treated with GSE at the concentration of 100, 200 and 300 microg/ml for 24, 48 and 72 hours, respectively. The the inhibitory effect of GSE on the growth of the PC-3 cells and the kidney cells of SD rats was determined by MTT reduction assay, with primarily cultured kidney cells of 1-3 days old SD rats as the normal control.
RESULTSGSE significantly inhibited the growth of PC-3 cells in a concentration- and time-dependent manner, but had only a mild inhibitory effect on the kidney cells.
CONCLUSIONGSE inhibits the growth of prostate cancer PC-3 cells and can be used as a new drug for the treatment of prostate cancer.

9.Short-term safety and efficiency of cryoablation for renal sympathetic denervation in a swine model.

BACKGROUNDRenal sympathetic nerves are involved in the reflective activation of the sympathetic nervous system in circulatory control. Catheter-based renal denervation (RDN) ameliorated treatment-resistant hypertension safely, but 10%-20% of treated patients are nonresponders to radiofrequency denervation. The purpose of this study was to investigate the safety and efficiency of cryoablation for sympathetic denervation in a swine model and to explore a new way of RDN.
METHODSSeven swines randomly assigned to two groups: Renal cryoablation (CR) group and control group. The control group underwent renal angiogram only. The CR group underwent renal angiogram plus bilateral renal cryoablation. Renal angiograms via femoral were performed before denervation, after denervation and prior to the sacrifice to access the diameter of renal arterial and the pressure of aorta abdominalis. Euthanasia of the swine was performed on 28-day to access norepinephrine (NE) changes of the renal cortex and the changes of renal nerves.
RESULTSCryoablation did not induce severe complications at any time point. There was no significant change in diameter of renal artery. CR reduced systolic blood pressure (BP) from 145.50 ± 9.95 mmHg at baseline to 119.00 ± 14.09 mmHg. There was a slight but insignificant decrease in diastolic BP. The main nerve changes at 28-day consisted of necrosis with perineurial fibrosis at the site of CR exposure in conjunction with the nerve vacuolation. Compared with the control group, renal tissue NE of CR group decreased by 89.85%.
CONCLUSIONSPercutaneous catheter-based cryoablation of the renal artery is safe. CR could effectively reduce NE storing in the renal cortex, and the efficiency could be maintained 28-day at least.

10.Comparison of the parameters obtained by sperm quality analyzer V and computer-aided sperm analysis system.

OBJECTIVETo assess the differences between the main parameters obtained by sperm quality analyzer V (SQA-V) and computer-aided sperm analysis system (CASA), and to investigate their application to sperm quality analysis for fertile and infertile men.
METHODSTwelve fresh semen samples from fertile volunteers and 73 from infertility patients were detected with SQA-V and CASA for sperm concentration and motility, the percentage and concentration of motile sperm, sperm motility index (SMI), amplitude of lateral head displacement (ALH), beat cross frequency (BCF), curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), linearity (LIN = VSL/VCL) and straightness (STR = VSL/VAP). The correlation between the parameters obtained by the two devices were analyzed.
RESULTSSignificant differences were observed in the above parameters between the fertile and infertile groups. An obvious consistency was noted between the results from SQA-V and those from CASA in sperm concentration (r = 0.58, P < 0.01), motile sperm concentration (r = 0.75, P < 0.01) and average sperm velocity (r = 0.59, P < 0.01). Significant correlations were found between the SMI from SQA-V and STR, LIN, BCF, VCL, VSL and VAP from CASA (P < 0.05).
CONCLUSIONThere is a consistency between the results from SQA-V and those from CASA. Both the devices can detect the seminal differences between different cohorts of patients.
