Objective To study the effect of angelica polysaccharides (APS) on recoverable ability from cryopreservation damage of umbilical cord blood (UCB) hematopoietic cells and APS with hematopoietic growth factors (HGFs) on in vitro expansion of UCB mononuclear cells (MNC) after thawing. AIM: To investigate the changes of cardiac function, oxidative stress and apoptosis in myocardium of rat model of heart failure induced by adriamycin (ADR). METHODS: At the end of the study, we observed content of malondialdehyde (MDA), activity of superoxide dismutase (SOD) and apoptosis. Expression of P53 protein and p53 mRNA were measured with immunohistochemical method and RT-PCR, respectively. RESULTS: Our data showed that content of MDA increased, activity of SOD decreased and apoptosis in myocardium happened while cardiac function decreased after ADR treatment. p53 gene expression and P53 protein obviously increased in heart failure group. CONCLUSION: There were oxidative stress and apoptosis occurred significantly in a model of heart failure induced by ADR. p53 gene might play an important role in the apoptosis. Correlation analyses suggested that apoptosis in myocardium is related to oxidative stress in ADR-induced heart failure.

Objective:To investigate the mRNA expression of β_2-adrenergic receptor(β_2-AR) and the receptor post signaling materials cyclic AMP(cAMP) and cAMP-dependent protein kinase (PKA) activity in myocardial infarction(MI)rats.Methods:MI model was made by ligation of rats'left anterior descending coronary artery.A total of 48 adult Wistar rats were randomly divided into four groups:Control group,the rats were sham operated,and 2 weeks post MI group,4 weeks post MI group and 8 weeks post MI group.The myocytes suspensions were untreated or treated in each group by salbutamol,isoproterenol,ICI118,551,atenolol,propranolol and pertussis toxin(PTX)respectively.The mRNA expression of β_2-AR and β_1-AR from left ventricular myocardium were detected by RT-PCR,the concentration of cAMP was determined by competitive enzyme immunoassay and the activity of PKA was determined by non-radioactive protein kinase assay.Results:Compared with Control group,the mRNA expression of β_1-AR was lower in MI groups(P<0.05),and the ratio of β_2-AR to β-AR increased from 19% to 38%(P<0.01)after MI.Compared with the untreated cells,cAMP concentration was increased by salbutamol treatment in Control group and 2 weeks,4 weeks,8 weeks post MI group by 98.1%,133.6%,147.7% and 150.7% respectively(P<0.05).Compared with isoproterenol treatment,ICI118,551 decreased cAMP concentration in 8 weeks post MI group(P<0.05).Atenolol decreased cAMP in Control group,2 weeks and 4 weeks post MI groups(P<0.05);Propranolol decreased cAMP concentration in Control and three MI groups(P<0.05).β-receptor inhibitor inhibited PKA activity in 8 weeks post MI group(P<0.05),Atenolol inhibited PKA activity in Control group and 2 weeks post MI group(P<0.05),Propranolol inhibited PKA activity in Control group and three MI groups(P<0.05,respectively).Conclusion:The ratio of β_2-AR to β-AR increased after MI.β_2-AR agonist increased cAMP concentration and PKA activity in myocytes in post MI rats.

AIM To study the effect of ketamine on proliferation,cell cycle in the cultured rat neural stem cells. METHODS The growth inhibition of ketamine on neural stem cell was evaluated by an MTT assay. The effect of ketamine on cell cycle was measured by flow cytometry. RESULTS Ketamine inhibited the growth of cultured rat neural stem cells. Flow cytometry analysis showed that G 0/G 1 phase rate was increased but S phase rate was decreased. CONCLUSION Ketamine can inhibit proliferation of cultured rat neural stem cells,and this inhibitition is associated with cell cycle block.

OBJECTIVETo explore the clinical effects of minor bone anchors and palmaris longus tendon graft in treating chronic mallet fingers deformity.

METHODSFrom January 2008 to June 2013, 26 patients with chronic mallet fingers deformity were treated with minor bone anchors and palmaris longus tendon graft. There were 18 males and 8 females, aged from 18 to 52 years old with an average of (32.0±1.3) years. Among them, 8 cases caused by machine injury, 6 cases by fall injury, 6 cases by sprain from fight, 4 cases by tendon spontaneous rupture, 2 cases by knife trauma. There was no tendon attachment of extensor tendon check in 16 cases, and with 0.3 to 0.5 cm tendon attachment in 10 cases. All patients had the flexion deformity and the disability of dorsiflexion activity. During operation, the distal interphalangeal joint was fixed in 10° to 20° dorsiflexion by a Kirshner wire, the minor bone anchor was used to reconstruct the extensor tendon insertion, the palmaris longus tendon slice was transplanted the decayed area of extensor tendon insertion. Four weeks postoperatively, the Kirshner wire was removed and the plaster external fixation was used, and the patient began function exercises. Postoperative complications were observed and fingers functions were assessed according to Dargan standard.

RESULTSThe patients were followed up from 6 to 14 months with an average of (5.0±0.3) months. Wound superficial infection occurred in 2 cases, the skin pressure ulcer in 2 cases, joint activities disability in 1 case; these symptoms got improvement after symptomatic treatment. Traumatic arthritis occurred in 2 cases, 1 case was improved after treatment, and 1 case had chronic pain for a long time. No internal fixation loosening or breakage and tendon rupture were found. According to Dargan standard to evaluate the finger function, 17 cases got excellent results, 8 good, and 1 poor.

CONCLUSIONIt is an effective way to treat the chronic mallet finger deformity using minor bone anchors and palmaris longus tendon graft, and the method has advantages of reliable fixation, easy operation, satisfactory effect and less complication.
