1.Clinical efficacy of small dose of antithymocyte globulin and zenapax in kidney transplant recipients

Objective To evaluate the effects of small dose of antithymocyte globulin(ATG)and zenapax in the induction therapy for kidney transplant recipients.Methods A series of 150 cadaver-donor kidney transplant recipients were randomly divided to 3 groups,ie,small dose of ATG group(total dose,2.1 -3.0 mg/kg;n=72),zenapax group(50mg,on the first and 14th d after operation;n=15)and controls without induction therapy(n=63).Follow-up was 6 months.The rates of acute rejection,delayed graft func- tion(DGF)and pulmonary infection were statistically compared among the 3 groups.Results During a 6-month period,in ATG,zenapax and control groups,acute rejection episodes occurred in 4 cases(5.5%), 1(6.7%)and 10(15.9%),respectively;DGF occurred in 3(4.2%),0 and 8(12.7%),respectively;pul- monary infection occurred in 4(5.1%),1(6.7%)and 3(4.8%),respectively;leucocytopenia occurred in 3(4.2%),1(6.7%)and 5(7.9%),respectively;thrombocytopenia occurred in 2(2.8%),1(6.7%)and 5(7.9%),respectively.Conclusions In the early stage of kidney transplantation,small dose of ATG and zenapax can be the optimal choice for induction therapy.
2.Conversion from calcineurin inhibitor to sirolimus for renal function deterioration in kidney allograft recipients

3.Clinical Observation of Moxifloxacin Combined with Jiehe Pill in the Treatment of Multidrug-resistant Tuber-culosis

OBJECTIVE:To observe the efficacy and safety of moxifloxacin combined with Jiehe pill in the treatment of muilt drug-resistant tuberculosis(MDR-TB). METHODS:116 patients with MDR-TB were randomly divided into control group and ob-servation group. Control group was given Moxifloxacin hyohochloride tablet 0.4 g once a day,observation group was additionally given Jiehe pill 9 g,twice a day,and 2 groups were given rifapentine,albendazole isoniazid,amikacin,pasiniazide,pyrazin-amide,p-amino salicylic acid and other conventional treatment. The treatment course for both groups was 18 months. The clinical efficacy,and T cell subsets (CD3+,CD4+,CD4+/CD8+),interferon(INF)γ,interleukin(IL)4 levels before and after 6,12 and 18 months,and incidence of adverse reactions in 2 groups were recorded. RESULTS:The total effective rate in observation group was significantly higher than control group,the difference was statistically significant(P<0.05). After treatment,CD3+,CD4+,CD4/CD8 and INF-γ level in 2 groups were significantly higher than before and gradually increased by time,and observation group was high-er than control group;IL-4 level was significantly lower than before and graually decreased by time,and observation group was lower than control group,the differences were statistically significant(P<0.05). There was no significant difference in the inci-dence of adverse reactions(P>0.05). CONCLUSIONS:Based on the coventional treatment,moxifloxacin combined with Jiehe pill has better efficacy than moxifloxacin in the treatment of MDR-TB,with similar safety.
4.Caffeic acid phenethyl ester against cellular injuries in the rotenone-induced Parkinson’s disease model

BACKGROUND:Caffeic acid phenethyl ester (CAPE) can inhibit lipid peroxidation after rat brain injury. However, the trend of 5-lipoxygenaseis (5-LOX) and cysteinyl leukotrienes (CysLTs) in model of Parkinson’s disease, and whether CAPE protects against rotenone-induced cel ular injuries by inhibiting the levels of 5-LOX and CysLTs stil need further research.
OBJECTIVE:To investigate the protective effect of CAPE on the rotenone-induced Parkinson-like injury, and to determine whether 5-LOX involved.
METHODS:(1) PC12 cel s in good-growth were col ected and divided into five groups cultured with different concentrations of rotenone (0, 0.01, 0.1, 1, 10μmol/L). 24 and 48 hours later, changes of cel ular morphology and activity were observed to single out the optimum concentration of rotenone;at 24 hours, the levels of 5-LOX and CysLTs were detected by western blotting and ELISA, respectively. (2) PC12 cel s were pretreated with different concentrations of CAPE (0, 0.01, 0.1, 1, 10 μmol/L) for 30 minutes, and 1 μmol/L rotenone was then added. The other cel s received no intervention as blank control group. Subsequently, the cel activity was detected, and the CysLTs production was detected by ELISA at 24 hours.
RESULTS AND CONCLUSION:(1) Rotenone (0.1-10μmol/L) could induce PC12 cel injury with overt morphological and cel activity changes at 24 hours, especial y the 1 μmol/L rotenone. (2) Rotenone also significantly increased the 5-LOX expression and CysLTs production in a concentration-dependant manner. (3) CAPE (1-10μmo/L) significantly attenuated rotenone-induced CysLTs production and cel viability reduction in a concentration-dependant manner. (4) These results suggest that CAPE protects against PC12 cel injuries in the model rat with Parkinson’s disease induced by rotenone involving 5-Lox.

Dendritic cells (DCs) are uniquely well-equipped antigen (Ag)-presenting cells. This function of DCs, coupled with their remarkable plasticity, renders them attractive therapeutic targets for immune modulation. Recent data have demonstrated a promising role for pharmacologic treatment as a means of generating potent regulatory DCs. Herein, the evidence that the potential of regulatory DC the-rapy is considerable and that there are compelling reasons to evaluate it in the setting of organ transplantation in the near future are discussed in this paper.

7.Experimental study on cervical lymph nodes removal enhance allograft survival in alkali-burned cornea

AIM:To explore the inhibitive effects of cervical lymphadenectomy on kerstoplasy after alkaline burns.METHODS:The Wistar rats' corneas were transplanted into Sprague-Dawley(SD)rats' eyes which were randomly divided into 4 groups:group A(control group);group B,the cervicallymphadenectomy group;group C,corneal transplantation after the alkali burn injury;group D,cervical lymphadenectomy following group C.Out of 6 rats in each group,the cornea of one rat Was used for mawophage immunohistochemistry at day 14 after the transplantstion,and the remaining 5 rats were used for studying corneal immune rejection with a slit lamp.The time when allograft rejection occurred was recorded and mean survival times(MST)were compared among the groups.RESULTS:Compared with the MST of group A(10.40±1.14 days),the MST of group B(46.30±9.46 days)Was significantly longer(P<0.05).MST of grafts between group C(7.00± 1.58 days)and group D(15.00±3.39 days)was also significant (P<0.05).At 14th day after the transplantation,there was no CD68 immunoreactivity in the graft of group B,and CD68 proteins were expressed to some extent in the grafts of group A and D.However,in the graft of group C,the expression of proteins Was dramatically up-regulated.CONCLUSION:Cervical lymphadenectomy therapy has a significant effect in preventing corneal allograft rejection in normal and alkali burned oorneai beds.
8.Ozonation with ultrasonic enhancement of p-nitrophenol wastewater

Synergetic effects for p-nitrophenol degradation were observed in the ozonation with ultrasonic enhancement. The enhancements of removal rate for p-nitrophenol and TOC were around 116% and 294% respectively in comparison with the individual ultrasound and ozonation systems. The synergetic phenomenon is attributed to two physicochemical mechanisms: (1)Ultrasound decomposes ozone causing augmentation of the activity of free radicals; (2) Ultrasonic wave increased the concentration of O3 in solution because of ultrasonic dispersion.

10.Reduced Incidence and Severity of Collagen-induced Arthritis in Mice Lacking LFA-1

Lymphocyte function associated antigen-1 (LFA-1) is a member of integrin family, that plays an important role in the adhesion of lymphocytes with other cells and matrix. To investigate the role of LFA-1 in collagen-induced arthritis (CIA), the incidence of CIA, histological and radiological assessments in the LFA-1 deficient (LFA-1~ -/- ) mice and control mice were examined. LFA-1~ -/- mice and control mice were immunized with 100?g collagen type II(CII) emulsified with an equal volume of Freund’s complete adjuvant (CFA), followed by the booster injection of the same amount of CII in CFA on day 21. Then, clinical, histological and radiological assessments were done. It showed that 57% control mice developed arthritis and apparently changed in the histological and radiological assessment, whereas the all of LFA-1~ -/- mice had the normal histological and radiographic response and none developed arthritis. These results suggeste that LFA-1 is indispensable for the onset of CIA.
