ObjectiveTo study the risk factors of childhood functional constipation.MethodsA cross-sectional analysis of risk factors associated with 182 cases of functional constipation in children was conducted from March 2013 to February 2014. ResultsThe male patients were more than females, mainly in preschool children. The risk factors were did not like fruits and vegetables (52.2%), drank less water (44.0%), did not develop the habit of daily bowel movement (39.5%), defecation fear (36.3%), inadequate food intake (25.8%). There were different risk factors in different age stages.ConclusionsFunctional constipation is related to gender, age, diet habits and mental psychology.

Objective To explore the clinical manifestation, imaging examination, treatment and prognosis of Alagille syndrome in a child combined with hepatocellular carcinoma. Method The clinical manifestation, assistant examination and diagnosis of Alagille syndrome combined with hepatocellular carcinoma were analyzed in the child, and the pertinent literature were reviewed. Results The 6-year-old girl was admitted to hospital for repeated jaundice, and had a history of cardiac surgery. After admission, the patient was found to have a typical face look such as frontal bossing, sunken eyes, pointed chin and hypertrophy of nasal tip. Blood biochemistry showed intrahepatic cholestasis and increased alpha-fetoprotein. Abdominal ultrasonography revealed diffuse multiple solid lesions in the liver. And magnetic resonance imaging of the liver indicated that the liver was enlarged and multiple solid space occupying masses. Jagged 1 gene detection showed heterozygosis mutation of c.1205delC. Conclusion Alagllie syndrome complicated with hepatocellular carcinoma in childhood is extremely rare, and early diagnosis and long-term follow-up are of positive significance for its treatment and prognosis.

Objective To investigate the relationship between diastolic function of the left ventricle and common carotid artery (CCA) elasticity in patients with hypertension. Methods Forty-one primary hypertension patients and 31 healthy volunteers underwent echocardiography to obtain the indices of LV diastolic function and also echo tracking techniqne (ET) to obtain the CCA elasticity indices. The LV diastolic function indices included Ve (early diastolic peak velocity), Va (late diastolic peak velocity ), Ve/Va, Em (early diastolic mitral annular motive velicity), Am (late diastolic mitral annular motive velicity) and Em/Am. The indices of CCA elasticity included Ep (pressure-strain elasticity modulus), β(stiffness parameter), AC (arterial compliance), AI (augmentation index) and PWVβ (pulse wave velocity). Results Va was higher and Em, Ve/Va and Em/Am were lower in patients than that in the healthy , P＜0.05. Ep and PWVβ were higher in patients,P＜0.05. Am and Va were correlated positively with PWV β and Ep , respectively in patients, P＜0.05. Ve/Va was correlated positively with AC, and negatively with PWV β, P＜0.05 in patients. Em/Am was correlated negatively with β and PWV β in patients. Conclusions The LV diastolic function and common carotid artery elasticity were decreased in patients with hypertension. The decreased common carotid artery elasticity was along with the LV diastolic function decreased.

Objective To investigate the relationship of acute graft versus host disease (aGVHD) and the regulatory T (Treg) cells,NK cells as well as their function-related cytokines such as IL-10,TGF-β,and perforin in mice with allogeneic bone marrow transplantation.Methods H-2 completely mismatched C57BL/6→ BALB/c aGVHD mice model was constructed.Flow cytometry analysis was used to detect the proportion of CD4+CD25 + Treg cells and NK-1.1+ NK cells in the spleen of aGVHD mice after transplantation.ELISA method was used to detect the serum levels of IL-10,TGF-β and perforin in the aGVHD mice intervened with CSA prophylactic or not.The normal C57BL/6 mice were used as controls.Results Compared with those of normal mice,the proportion of Treg cells in aGVHD mice after transplantation was decreased (mean 3.6％ vs.1.55％) and the proportion of NK cells increased (mean 3.3％ vs.11.5％).In the aGVHD mice treated with cyclosporine A,serum IL-10 expression level was significantly increased (treated group (125.79 ± 0.27)pg/mL,untreated group [(103.09 ± 3.27)pg/mL,P＜0.01)],TGF-β expression level was increased [(252.05 ±7.84)pg/mL vs.(241.61±15.41)pg/mL,P＞0.05],perforin expression level was significantly increased [(186.97 ± 4.68)pg/mL vs.(144.35 ± 14.42)pg/mL,P＜0.01].Conclusion ① The occurrence of aGVHD is correlated with the decreased number of Tree cells after transplantation in mice.Treg cell function-related cytokines IL-10 and TGF-β are involved in the treatment of aGVHD by cyclosporine A-mediated immunosuppression.②NK cells are involved in the occurrence of aGVHD after allogeneic bone marrow transplantation,and the increased level of perforin is related to the inhibition of aGVHD.

AIM To investigate that Danhong Injection may decrease inflammation and oxidative stress response in rat peritoneal mesothelial cells (RPMCs) cultured with high glucose.METHODS The third passage of RPMCs were used for the experiment.After being incubated with DMEM for 24 hours,RPMCs were divided into normal control group,high glucose group (RPMCs were incubate with 100 mmol/L glucose for 12 h,24 h,48 h,72 h),Danhong Injection group (complete medium with 80 mL/L Danhong Injection for 48 h) and high glucose + Danhong Injection group (100 mmol/L high glucose with 40 mL/L,80 mL/L,160 mL/L Danhong Injection for 48 h respectively).The cell proliferation was detected by MTT method.The levels of interleukin-18 (IL-18),interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in culture fluid were determined by enzyme linked immunosorbent assay.The contents of malondialdehyde (MDA),superoxide dismutase (SOD),glutathione (GSH),in the culture fluid and intracellular reactive oxygen species (ROS) were measured by kits.The expressions of Bcl-2,Bax,fibronectin (FN),endothelin-1 (EDN1) and haem oxygenase-1 (HMOX1) mRNA were detected by quantitative real-time PCR.The protein expressions of EDN1 and HMOX1 were measured by Western blot.RESULTS High glucose can significantly inhibit RPMCs' viability.High glucose can also up-regulate the expressions of IL-18,IL-6,TNF-α,ROS,MDA,Bax mRNA,FN mRNA,EDN1 mRNA and down-regulate the levels of SOD,GSH,Bcl-2 mRNA,HMOX1 mRNA in RPMCs.Treatment with Danhong Injection can effectively reverse aforementioned effect induced by high glucose.CONCLUSION Danhong Injection can reverse the inhibition of cell viability,the inflammation and oxidative stress response induced by high glucose in RPMCs,thus protect peritoneal membrane.

Chondrosarcomas has low morbidity in China,but we should pay more attention to the results it contributed to.Patients usually have poor prognosis because of defect of surgical resection and resistance to con-ventional chemotherapy and radiotherapy.Chemotherapy is a significant method to the chondrosarcoma treatment. If we overcome the difficulty of chemoresistance,chemotherapy will kill the sarcoma cells efficiently and inhibit the capacity of metastasis and invasion.The patients will have better prognosis.Finding the mechanisms of che-moresistance in chondrosarcomas is the key, now we review sevaral recent mechanisms of chemoresistance in chondrosarcoma.

Cancer stem cells are cells with stem cell characteristics within a tumor. With increased proliferative capabilities, they are able to self-renew and develop into various cell types, and thus play important roles in the formation, development, metastasis, and recurrence of tumors. MicroRNAs are a class of endogenous, small non-protein coding RNA molecules. Through regulating the expression of genes depending on the complementation between the microRNAs and their targets, microRNAs play important roles in various human cancers. This article summarizes recent research advances in the roles of microRNAs in cancer stem cells.

Objective To develop a BP26 recombinant BCG (rBCG-BP26) vaccine,and to observe the effects of rBCG-BP26 on CD4+,CD8+ T cells in immunized mice.Methods The recombinant shuttle vector pMV261-Ag85B-BP26 was constructed by using traditional molecular biological technology.The recombinant strains were obtained by kanamycin resistance screening and PCR identification after electroporation.Western blotting was used to detect the expression of recombinant BP26 vaccine in immunized mice.Safety experiment was carried out in three different groups:the target experiment(rBCG-BP26) group,the positive control(BCG) group and the negative control(PBS) group,15 BALB/c mice in each group.Intradermal inoculations of 100 μl rBCG-BP26 [containing 106 colony forming units(CFU)],BCG,and PBS were carried out,respectively.Signs of mice in each group were observed.After immunization for 10,20,30,and 40 days,body weight was weighed,and tail blood was collected to observe the change of peripheral blood CD4+ and CD8+ T cells by flow cytometry.Results The rBCG-BP26 was successfully constructed.The expression of BP26 protein was detected in the liquid medium and the bacteria cells.The results of safety test analysis showed that there were no significant differences in signs and body weights(F=2.468,0.331,1.520,0.739,all P＞ 0.05),between PBS group[ (19.24 ± 0.54),(21.37 ± 0.66),(22.83 ± 0.62),(25.06 ± 0.37)g],BCG group[ (19.90 ± 0.02),(21.53 ± 1.57),(21.95 ± 0.55),(24.70 ± 0.39)g]and rBCG-BP26 group[ (19.16 ± 0.55 ),(20.89 ± 0.20),(22.15 ± 0.76),(24.60 ± 0.64)g].The results of flow cytometry showed that the percentages of CD4+ T cell level were lower in BCG group(26.70％,33.07％) and rBCG-BP26 group( 13.40％,26.70％) than that of the PBS group(33.85％,29.33％) and the values of CD4+/CD8+ T cells increased in rBCG-BP26 group (0.69％,1.27％,1.57％,1.70％ ) 10,20 and 30 days after immunization.Conclusions Recombinant BCG-BP26 vaccine strain can express brucella BP26 protein efficiently.Furthermore,its virulence is mild,and it can activate CD4+,CD8+ T cells in the body.It can be used as one of candidate vaccine strain against brucellosis.
