1.Effect of short-term intensive treatment with insulin pump on the level of circulating endothelial progenitor cells in patients of newly diagnosed type 2 diabetes mellitus and its antioxidation mechanism

Objective To observe the protective effects of short-term continuous subcutaneous insulin infusion (CSⅡ) therapy on the peripheral blood circulating endothelial progenitor cells (EPCs) in patients with newly diagnosed type 2 diabetes mellitus (T2DM) and its antioxidation mechanism. Methods Forty T2DM patients (T2DM group) and 30 controls (control group) were involved in this study. Density gradient centrifugation was used to collect peripheral blood mononuelear cells. After 7 days' inducement and differentiation, FITC-UEA and DIL-acLDL double staining cells were identified to be differentiated EPCs by laser scanning conform microscope, and they were counted under inverted microscope. Serum levels of reactive oxygen species (ROS), malondialde-hyde (MDA), glutathione (GSH), superoxide dismutase (SOD) were examined. Results In T2DM group, the EPCs in peripheral blood were significantly reduced (46 ± 3 vs 23 ± 4, P< 0.01), BOS and MDA increased[(1125 ± 687) U/ml vs(2088 ± 765) U/ml, (4.58 ±3.03) μ mol/L vs (8.32 ± 4.15) μ mol/L, P < 0.01] , and GSH and SOD decreased [(263 ± 35) mg/L vs (206 ± 41) mg/L, (103 ± 7) U/ml vs (80 ± 15) U/ml, P < 0.05] . EPCs were negatively correlated with BOS and MDA (r = -0.7352,P < 0.01 ;r = -0.8454, P < 0.01) and was positively correlated with GSH and SOD (r =0.5435,P <0.05;r =0.6437,P <0.05). After short-term CSⅡ therapy, EPCs, GSH, and SOD in patients with T2DM significantly increased (P < 0.05),but BOS and MDA significantly decreased (P <0.05). Conclusion Short-term CSⅡ therapy can significantly reduce oxidative stress in patients with T2DM, promote the restoration of EPCs, and then prevent vascular disease.

Objective To examine expression of PTEN gene in ovarian cancer cisplatin-sensitive cell line OV2008 cells and cisplatin-resistant cell line C13K cells,and evaluate the effect of wild-type PTEN gene on reversing cisplatin-resistance of C13K cells and underlying mechanisms.Methods The expression of PTEN mRNA and protein in OV2008 and C13K cells were detected by semi-quantitative RT-PCR and western blot.Recombinant eukaryotic expression plasmid containing human wild-type PTEN gene was transfected into C13K cells by lipofectamine 2000.The expression of PTEN mRNA was monitored by RT- PCR and the expression of PTEN,protein kinase B(AKT),phospho-AKT(p-AKT)protein were analyzed by western blot in PTEN transfected and untransfected C13K cells.Proliferation and chemosensitivity of cells to cisplatin were measured by methyl thiazolyl tetrazolium(MTT),and cell apoptosis was detected by flow cytometry after treatment with cisplatin.Results(1)The expression of PTEN mRNA and protein(1.02 ?0.05,1.02?0.07)in OV2008 cells were significantly higher than those in C13K cells,which were 0.45 ?0.03 and 0.55?0.03 respectively(P
3.Effect of ketamine on proliferation and the cell cycle of cultured rat neural stem cell

AIM To study the effect of ketamine on proliferation,cell cycle in the cultured rat neural stem cells. METHODS The growth inhibition of ketamine on neural stem cell was evaluated by an MTT assay. The effect of ketamine on cell cycle was measured by flow cytometry. RESULTS Ketamine inhibited the growth of cultured rat neural stem cells. Flow cytometry analysis showed that G 0/G 1 phase rate was increased but S phase rate was decreased. CONCLUSION Ketamine can inhibit proliferation of cultured rat neural stem cells,and this inhibitition is associated with cell cycle block.

5.Effect of resveratrol combined with DDP on FRH-0201 cholangiocarcinoma cell line

Objcetive: To investigate the effect of resveratrol (RES)combined with cisplatin (DDP)on the cholangiocarcinoma cell line FRH-0201 in vitro. Methods: FRH-0201 cells were exposed to different concentrations of RES or (and)DDP. Cell morphological changes were observed by light and fluorescence microscopy. Inhibition of cell proliferation was measured by MTT and colony-forming assay. And the flow cytometry were performed to detect the cell apoptosis rate and cell cycle. Results: Resveratrol inhibited the proliferation of FRH-0201 cells in a time-and dose-dependent manner in the concentration of 5~320 ?mol/L and the IC50 values after the exposure of 24, 48, and 72 h were 55.35, 32.84, and 28.01 ?mol/L respectively. RES and DDP had synergetic effect in the inhibition of FRH-0201 cells. Conclusion: RES can inhibit the growth of FRH-0201 cells and has the synergetic effect with DDP.
6.Clinical assessment of one-level posterior lumbar interbody fusion performed with microendoscopic approach

Objective To evaluate the clinical outcome of one-level posterior lumbar interbody fusion (PLIF) performed with microendoscopic approach.Methods A consecutive series of 56 patients who under went one-level PLIF procedure(24 cases performed with microendoseopic approach and 32 cases with tradition- al open approach)were studied from January 2005 to May 2006.The following data were compared between 2 groups with 12 to 27 months follow-up:estimated blood loss,postoperative drainage,transfusion needs,surgi- cal time,length of hospital stay,postoperative back pain by visual analogue scale,complications,and the clinical and radiographic results.Results The microendoscopic approach was found to have a significantly less blood loss,less postoperative drainage,less needs of transfusion,less postoperative back pain,shorter re- covery time and shorter length of hospital stay.However,the microendoscopic approach needed significantly longer surgical time.There was no significant difference between 2 groups in the aspects of the complications and the clinical and radiographic results.Conclusion The one-level PLIF performed with microendoscopic approach minimize estimated blood loss,length of hospital stay,postoperative back pain.It also shows the ex- cellent surgical efficacy of the microendoscopic approach for suitable patients.
7.1H-MRS study on the metabolites of first dorsal interossei.

OBJECTIVE:
To estimate the application of prognosis evaluation of ulnar nerve injury by 1H-magnetic resonance spectroscopy (1H-MRS).
METHODS:
The metabolites of first dorsal interossei (FDI) of two hands from 12 healthy volunteers and 1 volunteer with complete ulnar nerve injury were detected by 1H-MRS and the data were statistically analyzed.
RESULTS:
For the FDI of healthy adults, the female peaks area of extra-myocellular lipids (EMCL) was higher than the male (P < 0.05); There was no significant difference in Cho, Cr and intra-myocellular lipids (IMCL) between male and female (P > 0.05); There was no significant difference in all the peaks area between the left and right hand (P > 0.05). The EMCL peak of the injury side was higher than that of the healthy side, and the area of FDI was reduced in the volunteer with ulnar nerve injury.
CONCLUSION
Noninvasive and quantitative detection of 1H-MRS may be valuable for prognosis evaluation of peripheral nerve injury.

8.Correlation between expression of aquaporin-4 and the apparent diffusion coefficient on diffusion-weighted imaging after focal cerebral ischemia/reperfusion in rats

Objective To investigate the correlation between the apparent diffusion coefficient (ADC) and the expression of aquaporin-4 (AQP4) in brain tissue after ischemia/reperfusion in rats. Methods A model of right middle cerebral artery occlusion (MCAO) induced by suture method. Seventy Wistar rats were randomly divided into 7 groups: sham-operation (A), MCAO 30 min (B), MCAO 30 min and reperfusion 30 min (D), MCAO 30 minand reperfusion 60 min (E), MCAO 60 min (C); MCAO 60 min and reperfusion 30 min (F), and MCAO 60 min and reperfusion 60 min (G) groups (n=10 in each group). The rats in all groups underwent diffusion-weighted imaging (DWI). The relative apparent diffusion coefficient (rADC) was calculated. Triphenyltetrazolium chloride (TTC) staining was used to detect the ratio of ischemic area. Immunohistochemistry, in site hybridization, and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect the expressions of AQP4. Results No abnormal intensity signals were observed on DWI in sham-operation group. The ranges of the high signal intensity lesions on diffusion-weighted imaging (DWI) were from small to large in groups B, C, D, E, F, and G. Then it reduced gradually, so did the ischemic area. The AQP4 expression was up-regulated significantly in groups B and E. The AQP4 expression was decreased significantly after the reperfusion in all groups. 1here was significant difference between ischemic groups and reperfusion groups (all P<0. 05). The expressions of rADC and AQP4 showed negative correlation (r=0. 72, P<0.01). Conclusions The AQP4 expression in brain tissue is closely associated with the changes of ADC after cerebral ischemia/reperfusion. DWI may indirectly reflect the levels of AQP4 expression.
9.Effect of flupentioxl melitracen and pinaverium bromide treatment on the changes of anorectal motility and rectal sensation in the patients of diarrhea-predominant irritable bowel syndrome accompanying with depression and/or anxiety status

Objective To study the effect of flupentioxl melitracen and pinaverium bromide treatment on the changes of anorectal motility and rectal sensation in the patients of diarrhea-predominant irritable bowel syndrome (IBS-D) accompanying with depression and/or anxiety status. Methods Forty-four patients with IBS-D accompanying with depression and/or anxiety status were divided into group A (flupentioxl melitracen and pinaverium bromide) and group B (pinaverium bromide) by random digits table,and treated for 4 weeks. Twenty-five healthy subjects were included as control group. The anorectal motility and rectal sensation before and after taking medicines were compared. Results When abdominal pressure was increased, the net increased pressure of anal sphincter was (3.0 ± 1.2 ) kPa in group A and (2.9 ± 1.2)kPa in group B. They were lower than that in control group [(3.6 ± 1.6) kPa](P< 0.05). The rectal lowest volume of sensory threshold, the maximal volume of tolerance and maximal compliance were (55 ± 20) ml,( 145 ± 78 ) ml, ( 21.9 ± 12.9 ) ml/kPa in group A, ( 56 ± 38 ) ml, ( 150 ± 50 ) ml, (20.8 ± 11.2) ml/kPa in group B. They were lower than those in control group [(80 ± 38 ) ml, ( 190 ± 50 ) ml, (30.8 ± 15.2 ) ml/kPa](P < 0.01 ). The rectal lowest volume of sensory threshold, the maximal volume of tolerance and maximal compliance were higher than those before taking medicines. Only the rectal lowest volume of sensory threshold in group B was higher than that before taking medicines. The rectal lowest volume of sensory threshold, the maximal volume of tolerance and maximal compliance in group A after taking medicines were higher than those in group B (P < 0.05 or < 0.01 ). Conclusions Higher sensitivity, lower tolerance,lower compliance of rectum and weakened anal automatic control function in IBS-D may be associated with diarrhea and frequent defecation. Treatment combining flupentioxl melitracen with pinaverium bromide may preferably improve the aperception functions of rectum in the patients of IBS-D accompanying with depression and/or anxiety status.
10.Effect evaluation of two kinds of wet dressing on peripheral phlebitis above stage Ⅱ

Objective To observe the effects of Suprasorb G and Suprasorb H on peripheral phlebitis above stage Ⅱ. Methods 64 patients with peripheral phlebitis above Stage Ⅱ were randomly divided into the observation group (36 patients) and the control group (28 patients). The observation group used Suprasorb G, and the control group used Suprasorb H. All cases were tested with the change of stage of peripheral phlebitis, the pain score, the red swelling of the skin and the palpable vascular cord. Results After treatment with Suprasorb H or Suprasorb G, there were significant differences in the change of stage of peripheral phlebitis, red swelling of the skin, pain score and the change of palpable vascular cord by self-contrast before and after treatment. Significant differences existed in change of stage of peripheral phlebitis, and red swelling of the skin and the change of palpable vascular cord at 48 and 72 hours, but no change was seen in pain score between 2 groups. Conclusions Suprasorb G and Suprasorb H beth show good effect in treatment of peripheral phlebitis, but Suprasorb G is better than Suprasorb H in improving the stage of peripheral phlebitis, also in reducing the area of red swelling of the skin and the palpable vascular cord with passage of time. But there is no difference in releasing of pain caused by peripheral phlebitis between them.
