Objective To observe the effects of T-lymphocyte function subsets,IL-4 and IFN-γ cell factor in different dose and stage after irradiation.Methoda The C57BL/6j mice were divided into shammed irradiation group and model groups. The radiation hurt model was induced by 60Co gamma rays(0.7,1.4,2.8 and 5.6 Gy).The changes of T-lymphocyte subsets CD3,CD4,CD8,IL-4 and IFN-γ in spleen cells were analyzed by flow cytometry in acute injury stage and recovery stage after irradiation. Results The lymphocyte subsets CD3 +,CD4+ and CD8 + decreased after irradiation, which were related to the irradiation dose.At 1 day after irradiation, the decreasing level of IFN-γ was higher than that of IL-4.When irradiation dose was over 2.8 Gy,IL-4 / IFN-γ showed a markedly increased compared with control group. At 25 days after irradiation,CD3 +,CD4+,CD8 +,CD4 +/CD8 +,IL-4 and IFN-γ recovered obviously, but they did not recover to the normal level of shammed irradiation group. Conclusions The depression of mouse immune function induced by γ-irradiation might be caused by changes of CD3,CD4,CD8 and CD4/CD8 ratio, especially the imbalance of IL4 and IFN-γ.

Chinese Acupuncture & Moxibustion 2010;30(8):679-682
A PubMed search is carried out using the keyword "acupuncture" from in the top four journals: New England Journal of Medicine (NEJM), The Journal of American Medical Association (JAMA), The Lancet, and British Medical Journal (BMJ). The papers with full texts on randomized controlled trials (RCTs) of acupuncture are included up till July 2009, and the corresponding trial design and trial quality in these papers are extracted and analyzed. The results reveal that papers published in these four journals are mostly on randomized controlled trials with good design, high quality and large sample size. Therefore, if Chinese researchers publish papers in these journals, appropriate trial design adhering to the characteristics of acupuncture should be used; the results should be reported according to CONSORT statement and STRICTA recommendations. Moreover, obtaining assistance from foreign research team is also important.

Objective Diabetic mice models were established.Hearts were separated from the body, reperfused after ischemia, and disposed of with the extraneous calcitonin gene-ralated peptide (CGRP),to prove if the CGRP had protective effect on the early-staged diabetic mice with cardiac ischemia.Method ICR mice were injected with Streptozocins intraperitoneally to establish diabetic model. The model mice and the normal mice were randomly divided into 4 groups:two diabetic groups, two control groups.Hearts of the experimental mice were taken out from the chest cavity alive, and hanged on the Langendorff steadily for 30min.Western blot and radioimmunoassay techniques were used to test expression of Vanilloid receptor(VR1) and CGRP in myocardic tissue;Madlab system was used to test the cardiac function, which including left ventricular end-diastolic pressure (LVEDP),left ventricular developed pressure (LVDP),heart rate (HR),and coronary artery flow (CF),in the process of ischemic reperfusion,And ELISA assay kit was used to measure the concentration of the lactate dehydrogenase (LDH) in perfusion fluid collected from the heart.Result Expressions of VR1 and CGRP in diabetic hearts were significantly lower than those in normal ones 2 weeks later(P<0.01).By comparing with normal hearts, diabetic hearts had higher LVEDP and CF (P<0.05),and lower LVDP and HR(P<0.05).However,release of LDH were lower than normal ones (P<0.05).Predisposition of normal and diabetic hearts with CGRP can improve the cardiac function after ischemie injury. And the beneficial effect was more profound in early-staged diabetic hearts than in normal ones. Conclusion The diabetes disease(DM) can impair the expression of CGRP in myocardiac tissue. The extraneous CGRP may exert more potent protective effect on cardiac function in the early-staged diabetic heart.

Objective To study the effects of semiconductor laser irradiation and high voltage static electric fields on small vessel damage in diabetic rats. Methods Fifty healthy male Wistar rats were randomly divided into two groups:8 rats in a normal group and 42 in a diabetic model group. The diabetic models were created by intrape- ritoneal injection of streptozocin. The diabetic model rats were randomly divided into four subgroups:a diabetes group,a semiconductor laser treatment group,a high voltage static electric field treatment group and a comprehensive treatment group receiving combined semiconductor laser and high voltage static electric field exposure. The rats in each treatment group were subjected to the corresponding intervention. After 20 days of treatment, the venous blood, kidney tissue and myocardium tissue of the rats were collected, and the concentrations of blood glucose,insulin,en- dothelin and tissue were detected. Results Compared with the normal group,a significant increase in blood glu- cose and endothelin was observed in the diabetic model group, along with significantly decreased blood insulin and significant small vessel endothelium proliferation in the kidney tissue.Compared with the diabetes group, endothelin levels were significantly lower in all 3 treatment groups, and blood insulin was also higher in the comprehensive treat- ment group. Conclusion There were severe abnormalities in blood glucose, insulin and endothelin as well as mild impairment of small vessel endothelium proliferation in the diabetic rats.Semiconductor laser and high voltage static electric field exposure have a role in treating and preventing these conditions in diabetic rats.

Background Most animal models of experimental autoimmune uveitis (EAU) are single attacked procedure, with a different feature from the natural course of human recurrent autoimmune uveitis. So establishing a recurrent EAU model is necessary for the clinical study on EAU.Objective This study was to establish the recurrent EAU model in rat and investigate the ocular inflammation and pathological manifestation and interleukin-17 (IL-17)expression in the eye. Methods T cells isolated from the spleen and draining lymph nodes of Lewis rats immunized with interphotoreceptor retinoid-binding protein (IRBP) 1177-1191 peptide fragments (R16) 10 days earlier were re-stimulated with R16 in vitro and injected into naive syngeneic rats to establish the recurrent EAU models, and the normal Lewis rats were used as controls. The eyes of model rats were then examined daily for clinical signs of uveitis by slit-lamp biomicroscopy and scored Caspi's criteria. The rats were sacrificed 1 month,2,3months after injection respectively, and the retinal sections were prepared for the pathological examination by hemotoxylin & eosin staining.Immunohistochemistry was performed to detect the expression of IL-17 in the retina. Results Adoptive transfer of R16-specific T cells to Lewis rats induced recurrent uveitis. The inflammatory scores on the fourth day,the sixth day,and the inflammatory response disappeared on the tenth day after injection. However, the inflammatory reaction occurred repeatedly 4 or 5 times in the 2-month duration after that, and the right and left eyes of a single recipient showed a different pattern of relapse, and the clinical manifestations of EAU was similar to the natural course to those of human autoimmune uveitis. In the retinal specimens of 1-,2-and 3-month group, the number of inflammatory cells was gradually decreased as the time lapse.Compared with the normal group, the thicknesses of the entire retina, outer nuclear layer and inner nuclear layer decreased with a significant difference among the 4 groups (F=20.46,288.40,4.43,all P=0.00).The number of RGCs in the normal group,1-,2-and 3-month group was 231.27 ± 15.36,225.36 ± 17.79,132.18 ±9.39 and 67.45 ± 11.90,respectively,showing a significant difference among them (F=68.94,P=0.00).Immunohistochemistry showed that the scores of the IL-17 expression in the rat retina were 0.64 ± 0.17,1.92 ± 0.19,1.17 ± 0.23 and 0.83 ± 0.23,showing statistically significant difference (F=64.10,P=0.00).Conclusions The stimulation of R16-specific T cells can induce recurrent EAU in Lewis rat.Th17 is involved in the disease course.

OBJECTIVETo investigate the elastic limit and relevant enclasp force of the non-precious metal casting clasp.
METHODSCasting clasp samples of five cobalt-chromium alloys and one 18 - 8 nickel-chromium alloy were made from prefabricated clasp wax by invesing, casting, sandblasting, and ultrasonic cleaning. The process of casting clasp samples deflected by loading and returned by unloading was tested and electric signals were collected by an omnipotent material machine. The analog electric signal was converted to digital signal by an analog to digital converter and stored in a computer. The elastic limit and the relevant enclasp force were analyzed using a relative software.
RESULTSThe elastic limit and the relevant enclasp force of the casting clasp made from the 18 - 8 nickel-chromium alloy were smallest and those of the clasps made from the cobalt-chromium alloys in various brands were different. The range of the elastic limit of the cobalt-chromium alloy casting clasp with the length of 5.0 mm in undercut was 0.28 mm-0.33 mm and the relevant enclasp force was 14.42 g-19.28 g.
CONCLUSIONSIn clinic, we should select the suitable undercut deepness wherein the cobalt-chromium alloy casting clasps, according to different brands of the casting alloy, undercut length, undercut slope, and the clasp thickness.
