Objective To investigate the effect of angiotensin Ⅱ(Ang Ⅱ)on the expression of integrin-linked kinase(ILK)in rat glomerular mesangial cells.Methods SD rat glomerular mesangial cells were separated and cultured.They were treated with 10-7 mol/L Ang Ⅱ in various time-point(0,12,24,48,72 h),and then cultured rat glomerular mesangial cells were treated 48 h with Ang Ⅱ in various concentration(0,10-11,10-9,10-7,10-5 mol/L).Then total protein expression in rat glomerular mesangial cells stimulated by Ang Ⅱ at every time-point and every concentration-point was extracted.The expression of ILK protein was measured by Western blot.Results 1.Expression of ILK protein in rat glomerular cells stimulated by Ang Ⅱ in various time-point,or various concentration was significantly increased.In group of 10-7 mol/L Ang Ⅱstimulated cells,the protein semiquantity of ILK at 0,12,24,48,72 h respectively was 0.18?0.02,0.37?0.07,0.90?0.10,1.73?0.12,and 1.72?0.13(F=166.78 P
4.Transcription factor SP1 decoy ODNs targeting α1, 3-GT renders porcine endothelial cells resistant to complement-mediated cytotoxicity

Objective To investigate whether porcine endothelial cells transfected with SP1 de-coy ODNs could resist complement-mediated cytotoxicity during the model of SV-40-PED with human serum in vitro. Methods Immortalized porcine aortic endothelial cells of the line PED were divided in-to three groups. The transfected group was SV-40-PED with SP1 decoy ODNs. The mismatched group was SV-40-PED with scrambled SP1 decoy ODNs. The negative group was cells with oligo-fectamine only. The expression of α1,3-GT mRNA and αGal was detected after 26 h by using fluores-cence microscope, Western blot, RT-PCR and lactate dehydrogenase (LDH) activity assay. Results Fluorescence microscopy observed the decreased fluorescence of αGal after SP1 decoy ODNs transfec-tion. Dotted fluorescent decrease could be observed on some membrane while the mismatched group and negative group with bright green fluorescence. Western blot showed that the average absorbance of the PED cells transfected with decoy ODNs was decreased to 52.6% of the negative group (P<0.05). The expression of α1,3-GT mRNA in the PED cells transfected with decoy ODNs was 0.42±0.20 (isoform 1) and 0.27±0.12 (isoform 2) respectively, significantly lower than in the negative group (isoform 1, 0.72±0.17; isoform 2, 0.50±0.19; both P<0.05). The expression of α1, 3-G Tin the mismatch group was not different from that in the negative group (P>0.05). 20% normal hu-man serum (NHS) and 40 % NHS had cytotoxic effect on both mismatch and negative groups, but de-coy ODNs could confer SV-40-PED protection from the cytolysis effect (P<0.05), which made a re-markable reduction of complement-mediated cytotoxicity towards SV-40--PED. Conclusions SP1 decoy ODNs could confer porcine endothelial cells protection from complement-mediated cytotoxicity effect in vitro.
5.Preliminary application of 64-slice multislice computed tomography in postmortem after a road traffic accident

Objective To investigate the value of 64-slice multislice computed tomography (MSCT)in full-body postmortem examination after a road traffic accident.Methods The process of road traffic accident involving a pedestrian was obtained from scene monitoring photographic recordings and scene investigation photos.First,the external autopsy was performed by professional forensic pathologist who then reported the autopsy results.Then,64-slice MSCT was employed to perform a full-body scanning,when the CT sectional images were processed with muhiplanar and 3D reconstructions.The CT imaging results were then compared with the autopsy findings to find difference.Results The external autopsy found many soft tissue injuries over the body,subarachnoid hemorrhage,haematothorax,multiple rib fractures and nasal bone fractures and viewed that craniocerebral injury and thoracic injury caused the death after road traffic accident.While 64-slice MSCT revealed severe injuries in many organs including head,chest,abdomen,pelvis and spine.Conclusions Compared with the external autopsy,64-slice MSCT can obtain more important information of tissue and organ injuries.Combined use of 64-slice MSCT and external autopsy can provide sufficient and reliable proofs for identification of road traffic accident injuries.
6.Discussing of influence mechanism of Chinese herbal monomer on physical stability of cream.

This study left flavonoids and alkaloids Chinese herbal monomer with common parent nucleus as cream base carriages drug respectively, cream base were prepared with stable span 60-tween 80 emulsification system. The near-infrared stability analysis technology was performed to quantitatively characterize the physical stability of cream. Base on the theory of gel network structure, theory of emulsification, theory of solubility parameter and theory of double layer, the influence mechanism of Chinese herbal monomer on physical stability of cream was discussed. The results showed that tetrahydropalmatine, matrine and naringenin had similar solubility parameter value with cream base material, creams prepared with those Chinese herbal monomer have higher Zeta potential value and stronger physical stability, and that those creams had similar microstructure information with cream base. However, a larger solubility parameter difference exists between baicalin, baicalein, berberine, palmatine and cream base material. Creams prepared with those Chinese herbal monomers had lower Zeta potential value and poorer physical stability, and that those creams had great different microstructure information with cream base.

7.Treating Persistent Henoch-Schonlein Purpura Nephritis by Injecting Methylprednisolone into Intra-Renal Capsule in Children

Objective To evaluate the results of treating the persistent Henoch-Schonlein purpura nephritis(HSPN) by injecting methylprednisolone into intra-renal capsule in children.Methods Twenty-two patients(aged from 6 to 13 years) with persistent HSPN were randomly divided into 3 groups.Group I was treated with prednisone;group Ⅱ was treated with high dosage methylprednisolone by venous injection,while group Ⅲ was treated by injecting methylprednisolone into intra-renal capsule.The 24 h urinary protein excretion,the levels of serum albumin and creatinine,or the blood cholesterole in children with persistent HSPN were detected at the beginning,4 weeks and 8 weeks of the study.The blood pressure,body weight were detected in the study duration.Results The values of 24 h urinary protein excretion were(2.35?1.09),(0.97?0.37),and(0.99?0.52) g(P
8. Clinical outcome of fresh or frozen-thawed blastocyst transfer: A historic cohort study

Objective To explore the relevant parameters and clinical outcome of frozen-thawed blastocyst transfer compared with fresh blastocyst transfer. Methods A historic cohort study was conducted to analyze the blastocyst implantation rate and clinical pregnancy rate of 61 patients receiving fresh blastocyst transfer and 372 receiving frozen-thawed blastocyst transfer from Mar. 2015 to Dec. 2016 in Changhai Hospital of Second Military Medical University. And then the clinical outcomes of day five (n=308) or day six (n=64) embryo were analyzed in the frozen-thawed blastocyst transfer group. Results There was no significant difference in age or number of the transferred blastocysts between the two groups. Blastocyst implantation rate in frozen-thawed blastocyst transfer group was significantly higher than that in fresh blastocyst transfer group (40.2% vs 29.6%, P=0.036), while no significant difference was found in clinical pregnancy rate between the two groups (57.8% vs 47.5%, P=0.134). Blastocyst implantation rate and clinical pregnancy rate of day five embryo were significantly higher than those of day six (42.3% vs 30.1%, P=0.016, and 60.7% vs 43.8%, P=0.012, respectively). Conclusion The pregnancy outcome of frozen-thawed blastocyst transfer is better compared with fresh blastocyst transfer, especially when transferring on day five.
9.Observation on therapeutic effect of acupuncture combined with chinese herbs on polycystic ovary syndrome of kidney deficiency and phlegm stasis type.

OBJECTIVETo observe the clinical therapeutic effect of acupuncture combined with Chinese herbs on polycystic ovary syndrome of kidney deficiency and phlegm stasis type and probe into the mechanism.
METHODSSixty-three cases of polycystic ovary syndrome of kidney deficiency and phlegm stasis type were randomly divided in to a combined acupuncture and Chinese herb group (n=32) treated with acupuncture at Qihai (CV 6), Guanyuan (CV 4), et al. and oral administration of Chinese herbs, and a simple Chinese herb group (n=31) treated with oral administration of the same Chinese herbs as in the combined acupuncture and Chinese herb group. The therapeutic effects and changes of follicle stimulating hormone (FSH), luteotropic hormone (LH), testosterone (T) and LH/FSH were compared between the two groups.
RESULTSThe total effective rate was 93.8% in the combined acupuncture and Chinese herb group and 80.6% in the simple Chinese herb group, the former being significantly better than the latter (P < 0.05). The decrease of T in the combined acupuncture and Chinese herb group was significantly su perior to that in the simple Chinese herb group (P < 0.01).
CONCLUSIONAcupuncture combined with Chinese herb therapy is superior to the simple Chinese herb in the clinical therapeutic effect on polycystic ovary syndrome of kidney deficiency and phlegm stasis type and decrease of T level, indicating this method is a better one for polycystic ovary syndrome of kidney deficiency and phlegm stasis type.

10.Relationship of Proteinuria,Immunopathogenesis and Humoral Immunity in Children with Henoch-Schonlein Purpura Nephritis

Objective To explore the relationship of proteinuria,immunopathogenesis and humoral immunity in children with Henoch-schonlein purpura nephritis(HSPN).Methods Serum IgG,IgA,IgM,C3 and C4 levels were detected with complete-automatic biochemistry analyzer in 47 children with HSPN and 18 heathy children,and the differences in the findings were compared among groups.The levels of IgG,IgA,IgM,C3 and C4 were compared among groups according to the count of proteinuria.The renal tissues were adopteded with immunity in the children with HSPN were taken for biospy examination,and the differences in immunopathalogy were compared among groups with immunof-luorescence assay.Results The IgG,IgA levels in children with HSPN were higher than those of heathy children,on the contrary,the IgM,C3 and C4 levels were lower than those of heathy children(Pa0.05),but the IgM level of the nephritic syndrome group were higher than that of other groups(Pa0.05).Conclusions There is severe disturbance of immunologic function in children with HSPN.There is no connections among serum IgG,IgA,IgM,C3 and C4 levels,proteinuria and renal immunopathlogy.
