1.Interactive tobacco control drama:its design and effect evaluation

Objective To explore the methods of tobacco control through interactive sitcoms and evaluate its impact on audience in their empathy, knowledge acquisition and behavior, providing basis for health communication activities in relation to tobacco control. Methods Relevant knowledge and skills on tobacco control were integrated into the drama, which ran for 8 performances in Shanghai.Four of them were randomly selected for evaluation.All the audience were invited to complete a questionnaire. Results Over 90% of the audience gave affirmation to the design, performance and interaction of the drama.More than 70% smokers established their supportive attitude for smoke free environment.Audience who preferred interactive activities (
2.Determination of Zeranols in Plant Tissue by Rapid Resolution Liquid Chromatography-Mass Spectrometry/Mass Spectrometry

A method was developed for the determination of zeranols (α,β-zearalanol,α,β-zearalenol,zear-alanone,zearalenone) with RRLC-MS/MS in the plant tissue. The samples were extracted with acetonitrile and reextracted with aqueous alkaline solution,cleaned up with MAX column and then determined by RRLC-MS/MS using multiple reaction monitoring ( MRM) scan mode. The results showed that the working curves were linear in the range of 0 -20 μg/kg. The limits of detection ( LOD) of zeranols were 0.5 μg/kg and the limits of quantification (LOQ) was 1. 0μg/kg. Extraction recoveries for zeranols ranged from 75. 8% to 105.4% and RSD was between 2.4% and 12.1%. The method is suitable for the determination of zeranols in the plant tissue.

4.Effects of activation of ALDH2 by ethanol on the expression of JNK in kidney of diabetic rats.

OBJECTIVETo observe the effect of activation of aldehyde dehydrogenase 2 (ALDH2) by ethanol on the expression of c-Jun N-terminal kinase (JNK) in the kidney of diabetic rats.
METHODSEightheen healthy male SD rats were randomly divided into 3 groups (n = 6): normal control group, diabetes group and ethanol + diabetes group. After 8 weeks, 24 h urine samples from rats were collected to detect urinary protein content. The kidney was isolated and the ratio of kidney weight/body weight (index of kidney weight) was detected. The levels of fasting blood glucose, glycosylated hemoglobin serum urea nitrogen and serum creatinine were measured. Morphological changes of renal tissue were observed by optical microscope. The protein expressions of ALDH2 and JNK in renal tissue were detected by Western blot.
RESULTSCompared with the normal control rats, the levels of fasting blood glucose, glycosylated hemoglobin, serum urea nitrogen, serum creatinine and the index of kidney weight were increased markedly in diabetic rats. The expression of ALDH2 protein was decreased, while p-JNK, JNK protein expressions and the ratio of p-JNK/JNK were increased. The morphological observation was shown that the amount of glomerular mesangial matrix were increased, basement membrane were thickened and capillary lumen were narrowed. However,in ethanol + diabetes group, renal function was improved and the damage of renal structure was attenuated. The expression of ALDH2 protein was increased, while p-JNK, JNK and the ratio of p-JNK/JNK were decreased.
CONCLUSIONEnhanced ALDH2 expression can protect kidney in diabetic rats, which may be relevant with inhibitting the activity of JNK pathway.

5.Recent advancement in miniSTR research.

As the most popular and important inherited marker used in forensic identification, short tandem repeat (STR) always have partial DNA profiling or even no results when handling degraded or minute DNA sample. Through redesigning primers close to STR core repeats, MiniSTR can access shorter STR loci and increase success rate of DNA profiling in degraded or minute DNA sample. The review provides an update on the advancement of miniSTR research to give information in the practice of forensic science.

6.Establishment of miniCTTA multiplex set with fluorescence-labeled primers.

OBJECTIVETo establish a miniCTTA multiplex set including short tandem repeat (STR) markers as CSF1PO, TH01, and TPOX and amelogenin gene, whose amplified fragments are shorter than 130 bp.
METHODSThe length of the gene fragments amplified with fluorescence-labeled primers were analyzed by ABI3100 Genetic Analyzer for genotyping of 100 unrelated individuals, 10 genealogies and 30 highly degraded specimens.
RESULTSThe genotypes derived form miniCTTA were consistent with the results by AmpFLSTR Identifiler kit.
CONCLUSIONMiniCTTA multiplex set is useful for personal identification and paternity test, especially applicable for degraded DNA sample.

8.The application of proton magnetic resonance spectroscopy and computerized tomography in evaluating nonalcoholic fatty liver disease.

OBJECTIVETo investigate the application of proton magnetic resonance spectroscopy (1HMRS) and computerized tomography (CT) in evaluating nonalcoholic fatty liver disease (NAFLD).
METHODSTwenty-two NAFLD patients were selected, according to the Chinese Medical Association standard, and compared with 20 healthy persons (as the control group). Their body mass index (BMI), waist to hip ratio (WHR), and blood pressure (BP) were examined. The serum ALT, the concentration of fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), and uric acid (UA) were tested simultaneously. The severity of hepatosteatosis was evaluated by 1HMRS and CT scans of their livers. The intrahepatic content of lipid (IHCL) and CT liver and spleen ratios were measured.
RESULTSThe BMI, WHR, serum ALT, FBG, TG, and UA were all elevated significantly in the NAFLD group and were (28.4+/-2.4) kg/m2, 0.91+/-0.04, (71.5+/-24.8) U/L, (5.67+/-0.61) mmol/L, (2.48+/-1.46) mmol/L, (420.7+/-57.5)micromol/L, respectively, P less than 0.01 or 0.05. Meanwhile, in the NAFLD group, the IHCL calculated by 1HMRS were increased and CT value ratios were decreased significantly compared with those of the control group (27.49%+/-12.27% vs 1.34%+/-0.79%, P less than 0.01). However, there was no correlation between the clinical features and the IHCL and between the clinical features and CT value ratios, but a negative correlation existed in the CT value ratio and IHCL.
CONCLUSIONSThe intrahepatic content of lipids can be measured precisely by 1HMRS, and 1HMRS is better than CT in quantitative evaluations of NAFLD.

9.Correlation of multislice spiral CT findings with vascular endothelial growth factor expressions and microvessel density in renal cell carcinoma.

OBJECTIVETo study the correlation of multislice spiral CT features and vascular endothelial growth factor (VEGF) expressions and microvessel density (MVD) in renal cell carcinoma (RCC).
METHODSFourth-seven patients with pathologically confirmed RCC were examined by multislice spiral CT, and VEGF expressions and MVD of the RCC and the adjacent normal tissues were determined by immunohistochemistry with specific monoclonal antibodies.
RESULTSVEGF expression and MVD in the RCC and adjacent normal tissues increased with the pathological grades of RCC (P<0.05), VEGF expression was found to significantly correlate with MVD (r=0.67, P<0.01). The positive expression of VEGF and MVD were associated with the findings by multislice spiral CT scan of tumor size, intratumor necrosis, cystic degeneration, intensity signal, lymph node metastases, invasion of the renal vein or inferior vena cava, and invasion of the adjacent organs or distant metastases (P<0.01).
CONCLUSIONMultislice spiral CT findings can be indicative of the histopathology of RCC, and some CT findings are closely correlated with MVD and VEGF expressions in RCC, which may help evaluate the biological behavior and malignancy of the tumor and predict tumor invasion and metastasis.

10.Preliminary investigation of acaroid mites breeding in commercial food in Hsinchu City,Taiwan Province,China

Objective To investigate the breeding of acaroid mites in the stored food in Taiwan Province,China.Methods The breeding mites from samples selected from the markets in Hsinchu City,Taiwan Province were separated under a micro-scope.The isolations were used for slide preparation,then observed and identified under a microscope.Results Among the 39 species samples,13 species of acaroid mites were isolated from the samples.They belonged to 11 genera of 6 families.They were Acarus siro,Tyrophagus putrescentiae,Tyrophagus longior,Aleuroglyphus ovatus,Caloglyphus berlesei,Suidasia nesbitti,Lar-doglyphus konoi,Lepidoglyphus destructor,Dermatophagoides pteronyssinus,Chortoglyphus arcuatus,Carpoglyphus lactis,Der-matophagoides farina and Gohieria fuscus.Conclusions There are acaroid mites breeding in the commercial food in Hsinchu City,Taiwan Province,China.Therefore,relevant measures should be taken to control the breeding of acaroid mites.
