1.Construction and ldentification of Cell Division Cycle 2 Promoter Reporter Gene Vector

Objective To construct the luciferase reporter gene vector of cell division cycle 2 (Cdc2) gene promoter and determine its transcriptional activity. Methods Primers were designed based on human Cdc2 promoter sequence from UCSC software. Then Cdc2 promoter from human genome DNA was replicated. After pGL3-Basic vector and Cdc2 promoter were digested with restriction enzymes SacⅠand XhoⅠseparately, Cdc2 promoter was inserted into pGL3-Basic vector. The recombinant plasmid named pGL3-Cdc2-promoter was transiently co-transfected into U2OS cells with control vector pRL-SV40, and then the activity of dual luciferase was detected. Results pGL3-Cdc2-promoter was constructed successfully. The restriction analysis and sequencing proved the entirely correct sequencing results. The luciferase activity was higher in pGL3-Cdc2-promoter/pRL-SV40 group than that of pGL3-Basic/pRL-SV40 group (1.591 5±0.199 8 vs 0.049 9±0.010 4). Conclusion pGL3-Cdc2-promoter can be transcribed and activated in U2OS cells. This study provided an important basis for screening and evaluation of anticancer drugs.
2.Clinical effect analysis of external nutrition support via jejunosto-my catheter after surgery of esophageal carcinoma

Objective:To investigate the clinical application of enternal nutrition support via jejunostomy catheter after esopha-geal carcinoma excision. Methods:The patients of the parenteral nutrition group (PNn group) received completely parenteral nutrition support. The patients of the external nutrition group (EN group) received total intravenous nutrition support on the first day after the sur-gery. And the nutritional targets and immunological function were timely examined at different points. Results:The time of postopera-tive exhaust, defecation and hospital stay were all shorter in EN group than in the PN group (P<0.05). The index of nutritional and im-mune function was better in en group than in pn group. Conclusion:The enteral nutrition can improve the nutritional status, and can maintain and promote the postoperative gastrointestinal function of the esophageal cancer patients.
4.Rapid Screening Method for Ten Sedative-hypnotica Drug in Functional Food by Liquid Chromatography with Mass Spectrometry

Objective To develop a method for the simultaneous separation and determination of 10 kinds of sedative hypnotica drugs in the functional food with high performance liquid chromatography mass spectrometry system.Methods The mobile phase consisted of acetonitrile and 15 mmol/L ammonium acetate solution(0.1% formic acid ),chromatographic column was Zobax SB C 18.Identification was based on the compound's absolute retention time,protonated molecular ion,and representative fragment ion by multiple reaction monitoring.The condition of determination was investigated and optimized.Results With this method,the linear range for the 10 drugs was 10-1 080 ?g/kg,the average recoveries ranged from 80.5%-97.1% and the detection limits were from 0.35-12.0?g/kg respectively.Conclusion The method established in the present paper is applicable to monitoring sedative hypnotica drug in the functional food.
5.Generation of thymic epithelial cells in mouse by blastocyst injection of induced pluripotent stem cells

Objective To examine an in vivo method for the differentiation of induced pluripotent stem cells (iPSCs) into thymic epithelial cells (TECs) in mice. Methods Green fluorescent protein-expressing iPS cells, derived from C57BL/6 mice, were injected into blastocysts from ICR mice. Chimeric blastocysts were then transferred into uteri of E2.5 pseudopregnant mice. Chimeric mouse could be identified by coat color 10 days after birth. The chimeric thymus was transplanted under the renal capsule of BALB/c nude mice. The spleen was cut out from the thymus-transplanted nude mice and the cells were dispersed and analyzed by a flow cytometer 4 weeks after transplantation. Results Chimeras were born 17 days after embryo transfer and 13 live-born chimeras were obtained. The contribution of iPSC-derived cells in the chimeras ranged from 5% to at most 90%. Typical thymic epithelium structure consisted of green fluorescent protein-expressing cells in chimera. The iPSCs-derived thymic epithelial cells could support the generation of new T cells. Conclusion The results indicate that mouse iPS cells can differentiate in vivo towards normally functioning TECs.
6.CT Diagnosis of Thymoma

Objective To study CT features of thymoma,so that to improve the accuracy of CT diagnosis and differential diagnosis.Methods 31 cases of thymomas proved by surgery and pathology were examined with conventional CT scans.CT findings of thymoma were analyzed.Results The lesions in 27 cases(87.1%)were located in the anterior upper and middle mediastinum.There were benign lesion in 11,including mass-cardiovascular interface(MCI) with convex type(8 cases),flatness type(1 cases) and concave type(2 cases).20 cases were malignant lesion,including MCI with cast type(18 cases) and concave type(2 cases).Irregular invasion to adjacent organs was found in 11 cases,others included pericardiac effusion(n=6),pericardial and mediastinal invasion(n=2),pleural effusion(n=4),pneumonia(n=2),lung,bone,mediastinal lymphadens metastasis(n=2) and liver,pancreas metastasis(n=1).Conclusion CT scans is of significant value in diagnosis and differential diagnosis of thymoma.
7.Induced Pluripotent Stem Cells-derived chimeric Thymus transplantation enhances T cells Reconstitution and prevents GVHD in Murine after allogeneic Bone Marrow Transplantation

Objective To explore the effects of iPS cells-derived chimeric thymus transplantation on T cells reconstitution and graft versus host disease of murine after allo-BMT. Methods iPS cells-derived chimeric thymus was grafted under the renal capsules of mice after allogeneic IBM-BMT. The mice were divided into three groups:IBM-BMT group, IBM-BMT+TT group and IBM-BMT+DLI group. Four weeks after BMT, T lymphocyte subsets in the peripheral blood were analyzed by flow cytometry, the degree and pathological examination of GVHD were observed, respectively. Results Percentage of CD8+T cells in IBM-BMT group, IBM-BMT+TT group and IBM-BMT+DLI group was(5.52 ± 0.83)%,(11.10 ± 1.49)%and(8.49 ± 0.82)%respectively, there was signifi-cant difference between pairwise comparisons(P<0.05), and percentage of CD4 + T cells of the peripheral blood in IBM-BMT+TT group(9.60 ± 0.69)%was significantly higher than IBM-BMT group(6.42 ± 1.40)%and IBM-BMT+DLI group(8.07 ± 0.65)%(P<0.05) . IBM-BMT group and IBM-BMT+TT group showed less clinical and histopathological scoring of GVHD than IBM-BMT + DLI group. Conclusion iPS cells-derived chimeric thymus transplantation could effectively accelerate T cells reconstitution and prevent GVHD after allo-BMT.
8.Treatment of nonunion of humeral shaft fracture with plate fixation and bone graft

Objective To evaluate the operative treatment of nonunion of humeral shaft fracture with in- ternal plate fixation and autogenous cancellous hone graft.Methods Forty-one cases of nonunion of humeral shaft fracture operatively treated from February 2002 to December 2004 were analyzed retrospectively.There were 32 males and nine females.Their average age was 37.5 years(range,17 to 67 years).Sixteen nonunions were defined as hypertrophic and 25 as atrophic.We followed all the patients and obtained their complete medical information. Results Our average follow-up was 22.6 months(range,8 to 42 months).Forty fractures(97.6%)were united within an average of 5.8 months(range,3 to 12 months).Complications included iatrogenic radial nerve injury in three patients,wound infection in one patient and fracture nonunion in one patient.At the final follow-up,shou]der and elbow functions were found to be satisfactory.Conclusion Open reduction and plate internal fixation sup- plemented with autogenous cancellous bone graft is an effective treatment for nonunion of humeral shaft fracture.
9.Sternocleidomastoid myocutaneous flap for reconstruction after resection of carcinoma in floor of mouth.

OBJECTIVETo evaluate the clinic outcome and indications of sternocleidomastoid (SCM) myocutaneous flap for reconstruction after resection of carcinoma in floor of mouth.
METHODSThirty-two patients underwent SCM myocutaneous flap for reconstruction of the floor of mouth were reviewed. According to the clinical data and the follow-up, the appearance and function were analyzed.
RESULTSOnly one myocutaneous flap was partial necrosis in termination of the flap, and growth with granular. In eight cases, partial epidermal loss over the skin paddle occurred with secondary healing with mucosal epithelium. All patients had oral diet with good separation of the oral floor and the tongue, and had no difficulty in speech intelligibility. The patients' appearance, function and the overall quality of life were improved.
CONCLUSIONThe SCM myocutaneous flap appears to be easy to use and suitable for reconstruction of the defect after resection of oral floor carcinoma, the indications of this flap should be selected carefully.

10.Current situation of the project of water improvement to reduce arsenic in endemic arsenism area in Shanxi province

Objective To investigate the actual situation of implementation of the project to reduce water arsenic in endemic arsenic poisoning areas in Shanxi province,and to provide a scientific basis for prevention and control of the disease.Methods In June 2011,a questionnaire survey of 151 arsenic endemic villages was carried out in Shanxi province.The actual situation of implementation of the project to reduce water arsenic was investigated.Water samples were collected and arsenic level was determined by atomic fluorescence spectrometry.Results In the 151 arsenic poisoning villages 126 villages had changed the water,and the rate was 83.44%.In these villages,22 villages did not use the water improvement utilities properly.Of the 33 water improvement projects to reduce arsenic,23 operated normally.The projects covered 110 villages,beneficiary population of 97 920 people,accounting for 46.9%(97 920/208 736) of the total population.Water arsenic exceeded the national standards(≤0.05 mg/L) in 10 of the water improvement projects which covered 31 villages,including 15 nonendemic villages and 16 diseased villages.There were a population of 24 764 people in the diseased villages.Conclusions In Shanxi endemic arsenic poisoning areas,water improvement efforts need to be strengthened; the quality of water improvement project needs to be improved,and the project's later management and maintenance also need to be further strengthened.
