A sensitive LC-MS/MS method to determine cocaine and its major metabolite benzoylecgonine in guinea pig's hair has been established. About 20 mg of decontaminated hair sample was hydrolyzed with 0.1 mol·L-1 HCl at 50 ℃ overnight, in the presence of cocaine-d3 and benzoylecgonine-d8 used as internal standards, and then extracted with dichlormethane. The analysis was performed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Positive electrospray ionization (ESI+) and multiple reactions monitoring (MRM) mode were used. The limit of detection (LOD) for cocaine and benzoylecgonine was 1 pg·mg-1. The calibration curves of extracted standards were linear over the range from 5 pg·mg-1 to 250 pg·mg-1 (r2≥0.999 7). The method was validated and applied to the analysis of guinea pig's hair after a single dose administration of cocaine hydrochloride. Cocaine and benzoylecgonine were not only detected, but also quantified in guinea pigs hair.

Chinese Pharmaceutical Journal 2015;50(15):1353-1358
Pharmaceutical excipients are important ingredients of drugs, and the quality of it directly related to the safety of drug. Compared to the 2010 edition, one of the most significant improvement of the 2015 edition of Chinese pharmacopoeia is the monographs of pharmaceutical excipients will be available in an individual book (Chinese Pharmacopoeia Book IV). And also, some new features will be found in the new edition: the doubled varieties of chronicled pharmaceutical excipients, the application of new techniques, the strict standards of the excipients for injection, the establishment of functionality-related characteristics. This article will interpret the developing trend of the standards of pharmaceutical excipients and the effect of the trend on the entire pharmaceutical industry in the future via the improvement of Chinese Pharmacopoeia.

To discuss the protective effect of aralosides (AS) on I/R-induced rat myocardial injury. The adult rat ventricular myocyte ischemia model was established through perfusion with sodium lactate perfusate and reperfusion with Ca(2+) -containing Tyrode's solution simulation. The cell contraction and ion concentration synchronization determination system was applied to detect the effect of AS on single I/R cell contraction and Ca2+ transients. According to the findings, AS could increase resting sarcomere length, contraction amplitude, ± dL/dt(max), calcium transient amplitude and speed of post-reperfusion myocardial cells (P < 0.05, P < 0.01), and decrease in time for achieving 90.0% of maximum relaxation, time for achieving peak value, resting calcium ratio, contraction period [Ca2+] i, time for achieving 50.0% of maximum relaxation and attenuation rate of intracellular calcium transient (P < 0.05, P < 0.01). Therefore, it is suggested that AS improved the post-reperfusion cell contraction and injury of calcium homeostasis.

Objective To study the effects of semiconductor laser irradiation and high voltage static electric fields on small vessel damage in diabetic rats. Methods Fifty healthy male Wistar rats were randomly divided into two groups:8 rats in a normal group and 42 in a diabetic model group. The diabetic models were created by intrape- ritoneal injection of streptozocin. The diabetic model rats were randomly divided into four subgroups:a diabetes group,a semiconductor laser treatment group,a high voltage static electric field treatment group and a comprehensive treatment group receiving combined semiconductor laser and high voltage static electric field exposure. The rats in each treatment group were subjected to the corresponding intervention. After 20 days of treatment, the venous blood, kidney tissue and myocardium tissue of the rats were collected, and the concentrations of blood glucose,insulin,en- dothelin and tissue were detected. Results Compared with the normal group,a significant increase in blood glu- cose and endothelin was observed in the diabetic model group, along with significantly decreased blood insulin and significant small vessel endothelium proliferation in the kidney tissue.Compared with the diabetes group, endothelin levels were significantly lower in all 3 treatment groups, and blood insulin was also higher in the comprehensive treat- ment group. Conclusion There were severe abnormalities in blood glucose, insulin and endothelin as well as mild impairment of small vessel endothelium proliferation in the diabetic rats.Semiconductor laser and high voltage static electric field exposure have a role in treating and preventing these conditions in diabetic rats.

Objective To explore the protective effect of Salvianolic acid B on isolated lung in rats.Method Twenty-four SD rats were randomly divided into 2 groups:control group and experimental group (n =12 each). The isolated lung in control group was perfused with raffinose-low potassium dextran (R-LPD),and that in experimental group was perfused with R-LPD and 800 mg/L of Salvianolic acid B.The model of isolated lung was established in all these rats.Wet/dry weight ratio (W/D),myeloperoxidase (MPO),malondialdehyde (MDA) and superoxide dismutase (SOD) were measured, and the pulmonary structures were observed by HE staining at different preservative periods of 6,9,12 and 24 h after infusion.Result Intragroup comparison was made in both groups:all parameters had no statistically significant difference before 12 h.However,W/D,MPO,MDA,and SOD at 24 h were higher than those at 12 h (P<0.01).The result of interclass comparison showed that after 6,9 and 12 h preservation, all parameters of these two groups showed no significant difference.However,W/D,MDA and MPO were lower, and SOD was higher after 24 h preservation in control group (P<0.05).Moreover,before 12 h preservation, the two groups did not show inflammatory injury, only manifested slight inflammatory reaction histologically.Destructive pulmonary structure, alveolar interstitial edema and inflammatory damage were seen more clearly in control group than in experimental group. Conclusion The perfusion with Salvianolic acid B had no apparent superiority before 12 h.However, after 12 h,it could ensure the quality and prolong the preservation time of the donor lungs more effectively.Salvianolic acid B might play an important role in donor lungs protection.

Objective To know the influence of short-term hard health education combined with net-working fellow-up on diabetes related costs. Methods Divided 83 diabetes patients into the experimental group (41 cases) and the control group (42 cases) randomly. Short-term hard health education combined with networking fellow-up and traditional health education was used in the two groups respectively, and then evaluat-ed the diabetes related costs between the two groups on the time points of the third, the sixth and the dozenth month. Results The costs of health products in the control group was significant higher than that of in the experimental group from the first month to the third month after the intervention, while the inspection expenses in the experimental group was significant higher than that of in the control group from the fourth month to the sixth month after the intervention. Conclusions Short-term bard health education can not change the average level of diabetes related costs, but it can change the constituent ratio of costs.
