1.Determination of Adenosine in Chongcaoqizhi Granule by HPLC
Chinese Journal of Information on Traditional Chinese Medicine 2006;0(04):-
Objective To develop a RP-HPLC method to determine adenosine in Chongcaoqizhi granule. Method The separation was performed in a Kromasil ODS-1 column (250 mm?4.6 mm, 5 ?m) with a mobile phase of phosphate buffer solution (pH 6.5) - methanol (85∶15). The flow was 0.7 mL/min and the detective wavelength was set at 260 nm. Results The standard adenosine showed a good linear correlation at range of 1.016~12.192 ?g/mL. The linear regression equation and correlation coefficient (r) were Y =169339X -82318, r =0.9990, respectively. The average recovery of the loading was 98.02% and the RSD was 1.72%. The average content of adenosine was 12.45 ?g/g. Conclusion This method is accurate, reproducible and highly selective, and can be used for quality control of Chongcaoqizhi granule.

Objective To observe the effect of Sijunzi Decoction (SD) on the mitochondrial DNA deletion of the aging model mice, and to explore its anti-aging biomolecular mechanism. Methods Kunming mice were divided into 6 groups according to the body weight: normal control group, .model control group, high-, middle-and low-dose SD groups (at the dose of 20, 10, 5 g·kg-1 respectively), and vitamin E group (250 mg·kg-1), 20 mice in each group. The mice models of aging were induced by D-galactose. Water maze test was used to evaluate the learning and memory ability of the aging model mice. The effect of SD on the mitochondrial DNA deletion of the aging model mice was ob-served. Results In the hippocampal mtDNA of model control group and low-dose SD group, there were about 3.87 kb fragment missing. But there was no missing pieces in high-and middle-dose SD groups. Conclusion The anti-aging biomolecular mechanism of SD is probably related to the preventive effect on the mutation and deletion of the mitochondrial DNA in the aging model mice.

To improve the yield of industrial fermentation, a method based on near infrared spectroscopy was presented to predict the growth of yeast. The spectral data of fermentation sample were measured by Fourier-transform near-infrared (FT-NIR) spectrometer in the process of yeast culture. Each spectrum was acquired over the range of 10000-4000 cm1.Meanwhile, the optical density (OD) of fermentation sample was determined with photoelectric turbidity method. After that, a method based on competitive adaptive reweighted sampling (CARS) was used to select characteristic wavelength variables of NIR data, and then extreme learning machine (ELM) algorithm was employed to develop the categorization model about the four growth processes of yeast.Experimental result showed that, only 30 characteristic wavelength variables of NIR data were selected by CRAS algorithms, and the prediction accuracies of training set and test set of the CARS-ELM model were 98.68% and 97.37%, respectively. The research showed that the near infrared spectrum analysis technology was feasible to predict the growth process of yeast.

Objective To observe the expressions of AQP9 and AQP9 mRNA in cerebral cortex and hippocampus of rats after moderate traumatic brain injury(TBI).Methods The moderate TBI model was established according to Feeney's method. At different time points after TBI,the degree of cerebral ischemia by TTC staining, the expressions of AQP9 and its mRNA in brain by immunohistochemistry and RT-PCR respectively were performed. Results No obvious ischemia in brain was found 6 h after TBI,but gradually the ischemic focus enlarged, and reached the maximum 72 h after TBI.The expressions of AQP9 and its mRNA were similar, increasing significantly 3 h after TBI,decreasing 6 h after TBI,then increasing again at 24 h in bilateral cerebral cortex and the hippocampus and reaching the maximum 48 h or 72 h after TBI.Conclusion The increased expression of AQP9 after TBI is involved in the stress of early TBI and in later brain energy metabolism.

Abstract: Mosquitoes are involved in the transmission of serious diseases such as malaria, dengue, chikungunya, yellow fever, Zika virus disease, and filariasis, and their prevention and control have always been a research hotspot. Currently, mosquito control methods mainly include physical control, chemical control and biological control. Physical control methods are environmentally friendly, but they are slow to take effect and have unsatisfactory control effects; although chemical control can quickly eliminate mosquitoes, it has been eliminated due to its high pollution, high residual, and easy drug resistance; biological control uses natural enemies or pathogens to kill mosquitoes and reduce their ability to transmit disease. Therefore, environmentally friendly biological control has become the main measure for controlling and preventing mosquitoes. In recent years, significant progress has been made in bacterial mosquito control agents, among which Bacillus thuringiensis has been the most extensively studied. Bacillus thuringiensis is a Gram-positive soil microorganism, which is the pathogenic bacterium of a variety of agricultural pests such as Lepidoptera, Coleoptera and Diptera. During the sporulation process, its strains produce a variety of insecticidal crystal proteins (ICPs) or δ-endotoxins with insecticidal activity against mosquito larvae. This review firstly introduces the crystal proteins of Bacillus thuringiensis, describes in detail the types and structures of crystal proteins in detail, and also reveals the mechanism of action of crystal proteins related to receptors.
