The function that participate in modification and degradation of some cancer factors is vital in tumor cells,and followed by tumor biological behavior changing. Researches show that UbE2D3 correlates with human telomer-ase reverse transcriptase( hTERT),radiation sensitivity,aggressive,etc in breast cancer.

Objective To study the regulation role on molecular level of hCG on P450scc,P450c17 and 3?HSD in adult mouse Leydig cells. Methods Leydig cells of adult mice were cultured in vitro for 1 and 8 hours with or without hCG,then P450scc,P450c17 and I,VI total 3?HSD mRNA levels were measured respectively by the semi quantitative RT PCR as well as Scal restriction endonuclease incised methods.Meanwhile,the testosterone contents were measured in two groups. Results 1.In stimulated group Leydig cells cultured for 1 and 8 hours with hCG,the testosterone contents were higher significantly than those in control groups(1 hour P 0.05),but the ratio of type VI/I 3?HSD mRNA gradually increased. Conclusion hCG can stimulate the transcription of P450scc,P450c17 and type VI 3?HSD in adult mouse Leydig cells.

Objective Jasminodin in Mei Rong capsule was determined by the method of thin layer chromatography. Methods The sample jasminodin was extracted with methanol and then the extract was chromatographed on silica gel GF254 plate with chloroform-methanol-water (10:3:1) as mobile phase and the chromatograms were scanned by using Shimadzu CS-930 dual wavelength TLCS in zegzag scanning mode at λs 370nm and λR 250nm.Results Calibration graph was rectilinear between 2.7μg～16.2μg per spot for the jasminodin. Conclusion Satisfatory results were obtained with the average recovery ratio of 99.76% ,RSD 2.27%.

Objective To study the protective effect of normothermic ex vivo perfusion to prevent donor-lung injury.Method After being flushed with perfadex solution through the pulmonary artery and vein,the donor lungs were divided to control group (preserved with 4℃ cold static preservation,n =8) and experimental group (preserved with ex vivo oxygenated blood continuous perfusion,n =8).Pulmonary function indexes,including pulmonary vascular resistance (PVR),lung ventilation resistance(LVR),the left inferior pulmonary vein(LIPV) blood PO2,and lung water content,were evaluated.Lung injury were studied by light microscopic observation and detecting the expression of proimflammatory factor IL-1β was detected.Result (1) Lung function evaluation:in both group,PVR,LVR and lung water content gradually increased,LIPV PO2 gradually declined;But in the same time point PVR and LVR in group were were significantly less than in control group (P＜0.05),LIPV PO2 in experimental group were were significantly more than in control group(P＜0.05).(2)Lung injury evaluation:Lung tissue edema classification in both group were significantly higher than in normal lung(P＜0.05),but classification in experimental group were significantly lower than in control group(P＜0.05).The expression of IL-1β in experimental group (0.422 ± 0.132) were significantly lower than in control group(0.578 ± 0.163,P＜0.05).Conclusion Ex vivo oxygenated blood continuous perfusion without ventilator is a feasible technique for lung preservation.It ameliorates hypothermic time-dependent ischemic injury.But further systems evaluation in complete physiologic condition is necessary.

Objective To observe blood glucose values and serum insulin levels at different times after acupoint injection of insulin with PVP as a sustained release agent and analyze the correlation between them in SD Rats.Methods One hundred and forty-four male SD rats were randomized into groups A (acupoint injection of insulin alone), B (acupoint injection of sustained-release insulin) and C (control). At one hour after every group received an oral gavage of 40% glucose solution (2.2 g/kg), groups A and B were given point Zusanli injection and group C was not treated. In the three groups of rats, blood glucose values and serum insulin levels were measured at six time points: 0.5, 3, 6, 8, 10 and 24 hours after acupoint injection (in group C at the corresponding time points). In every group, the reaction of Point Zusanli region to the stimulation was observed by optical microscopy in at three time points: 3, 10 and 24 hours after acupoint injection.Results The glucose-lowering rate reached its peak at 3 hours after acupoint injection and then declined in groups A and B. The hypoglycemic effect tended to be stable at 6 hours after acupoint injection in group A and at 10 hours after acupoint injection in group B. The hypoglycemic effect was better in group B than in group A at 6 and 8 hours after acupoint injection (P<0.01). In group A, serum insulin reached its peak at 3 hours and tended to be stable at 6 hours after acupoint injection (P<0.05). Serum insulin levels were significantly lower in group B than in group A at four time pints: 0.5, 3, 6 and 8 hours after acupoint injection (P<0.05). An analysis of the correlation between rat serum insulin levels and the glucose-lowering rate showed a significant positive correlation in group A (P<0.01) and a low positive correlation in group B (P<0.05). In groups A and B of rats, clear stripes and complete structure of muscle fibers were seen with no obvious degeneration, necrosis and inflammatory reaction around point Zusanli at three time points: 3, 8 and 24 hours after acupoint injection; there were no obvious pathological changes compared with group C.Conclusions PVP as a sustained release agent can slow down the absorption rate of insulin in rat point Zusanli region. Meanwhile, it prolongs the continuous time of the hypoglycemic effect of insulin. There is a low correlation between serum insulin levels and the hypoglycemic effect, suggesting that acupoint injection with a sustained release agent can prolong the effect of acupoint injection of medicine.

Objective To provide a scientific reference for the choice of antimicrobial drugs by analyzing the distributions and the antimicrobial resistances of Carbapenem‐resistant Acinetobacter baumannii (CR‐AB) and methicillin‐resistant Staphylococcus au‐reus (MRSA) in the Northern Jiangsu People′s Hospital .Methods All the CR‐AB and the MRSA isolated from hospitalized pa‐tients from January 2011 to March 2014 were collected and analyzed .Use the disk diffusion method for antimicrobial resistance tes‐ting .Results A total of 170 CR‐AB strains and 168 MRSA strains were isolated .CR‐AB mainly distributed in the ICU ,which ac‐counting for 61 .17% (104/170) .MRSA mainly isolated in neurosurgery ,about 27 .98% (47/168) .67 .06% of CR‐AB strains and 54 .17% of MRSA strains were from Sputum specimens .The antimicrobial resistance rate of CR‐AB to most antimicrobial drugs were reached 50 .00% ,the resistance rates to cefoperazone/sulbactam (27 .65% ) ,minocycline (21 .18% ) ,amikacin (19 .41% ) and tobramycin (12 .35% ) were all less than 30 .00% ,the strain that resistant to polymyxin was not found .The sensitivity rates of MR‐SA to linezolid ,vancomycin ,and tigecycline were 100 .00% ,and the resistance rates to nitrofurantoin and kuinuputing/ dalfopristin were low (less than 2 .00% ) .Conclusion The antimicrobial resistances of CR‐AB and MRSA are serious .In order to control effec‐tively the spreading of CR‐AB ,MRSA and other multi‐drug resistant bacteria ,it′s necessary for us to strengthen the management of key departments ,and select antimicrobial drugs based on the results of drug susceptibility testing .

Methods The technique of in situ RP\|PCR was used to detect the expression of type Ⅰ and Ⅲ 17? HSD mRNA. Results In the cells of X\|zone which located in the interface between adrenal crotex and medulla, there were expressions of type Ⅰ and Ⅲ 17? HSD.Conclusion\ The X\|zone cells of adrenal gland in immature mouse could synthesize androgen, enhanced the male sexual differentiation together with the androgen synthesized from the testis.\;[

Objective:To optimize the processing technology of prepared Morinda officinalis based on nystose content. Methods:Orthogonal experiments and the nystose determination method in Chinese Pharmacopoeia (2010 edition) were used to optimize the pro-cessing technology. A Welch AQ-C18(250 mm ×4.6 mm,5 μm)chromatographic column was applied with methanol-water(3∶97)as the mobile phase, the column temperature was at 25℃, the flow rate was 0. 8 ml·min-1 , the injection volume was 10μl, and the drift tube temperature was at 40℃ with low temperature atomization. Results:The linearity of nystose was good within the range of 0. 214 8-0. 644 4 mg·ml-1 and the recovery was 99. 5%(RSD=3. 5%,n=6). Conclusion: On the basis of nystose content determination, the optimal processing technology of prepared Morinda officinalis is as follows:liquorice of 4%, stir fry time of 15min and the boiling pot temperature of 200℃.
